    Preparation of Immobilized Nucleoside Phosphorylase and Research on Its Property

    目的优化核苷磷酸化酶的固定化工艺,并对固定化酶的性质进行研究。方法通过正交试验优化核苷磷酸化酶(包括嘌呤和嘧啶核苷磷酸化酶)固定化条件,并进一步对固定化核苷磷酸化酶的最适P H、温度及其稳定性进行研究。结果固定化酶条件为P H=8.0,温度为25℃,蛋白载量为20 Mg/g载体,固定化时间为6 H;固定化核苷磷酸化酶的最适P H=7.5,最适温度为40℃,固酶连续使用20批次酶活力无明显损失。结论固定化核苷磷酸化酶稳定可靠,适合应用于工业化生产。Objective To optimize the immobilization process of the nucleoside phosphorylase and research on its property.Methods The immobilization process of the nucleoside phosphorylase(including purine and pyrimidine nucleoside phosphorylase)was optimized by orthogonal test,and took further study of the finest p H,temperature and stability of immobilized nucleoside phosphorylase.Results The finest condition of immobilized enzyme was p H = 8.0,25 ℃,20 mg/g protein loads and immobilize for 6 h; the finest condition for the immobilized nucleoside phosphorylase to catalyze was p H = 7.5 and 40 ℃.There was no significant loss of activity of the enzyme after use for catalyzing 20 batches consecutively.Conclusion The immobilized nucleoside phosphorylase is stable and reliable,which is suitable to application in industrial productio

    以十二烷基硫酸钠(SDS)为模板剂,正硅酸乙酯(TEOS)和N-三甲氧基硅丙基-N,N,N-三甲基氯化铵(TSPMNC)为硅源,以NaOH为催化剂,合成出带有季铵荷正电基团的有序介孔材料(QAS)。采用XRD,TGA、BET、SEM,XPS、TEM、FT-IR等手段对产品结构进行了表征和分析。结果表明,产品具有较为均一的六方介孔结构,孔径约为3.5nm,产品中季铵荷电基团的含量约为1.41mmol/g

    利用农药对酯酶活力的抑制反应,测定了不同浓度的敌敌畏对游离的不同动物肝脏酯酶的酶活性。实验采用从动物肝脏中提取的粗酶液在水解后与显色剂固兰B盐作用,用分光光度计测定其在595nm处的吸光度。结果表明,猪肝中α-乙酸萘酯酶(α-NA Esterase)的活力较高,且对较低浓度的敌敌畏也有较好的灵敏度。鸡肝和鸭肝α-乙酸萘酯酶对敌敌畏浓度变化不是很明显。Based on the inhibiting mechanism of esterase,the general esterase activities and the specific activities in livers of various animals materials such as chicken liver,duck liver and pork liver were studied and compared for the detection of organophosphate pesticides.     γ-分泌酶复合体对β-淀粉蛋白前体蛋白(Beta-amyloid precursor protein,βAPP)的水解是生成β-淀粉蛋白(Aβ)的关键步骤,在阿尔茨海默氏症(Alzhei mer′s disease,AD)的病理发生中起着重要作用.PEN-2蛋白(Presenilin enhancerprotein 2)是γ-分泌酶的一个重要组分,在γ-分泌酶的组装和成熟中扮演重要的角色.此外,PEN-2还具有促进神经细胞存活和防止细胞凋亡的作用.为了进一步研究PEN-2的生理功能,以PEN-2为诱饵蛋白用酵母双杂交系统筛选与之有相互作用的蛋白,发现PCBP4和GRK5肽段可结合PEN-2,并在酵母体系用β-半乳糖苷酶报告基因验证了它们的相互作用.这些结果表明PCBP4和GRK5可能影响PEN-2的正常生理功能,并且有可能在阿尔茨海默氏症的形成中起着一定的作用.Presenilin enhancer protein 2(PEN-2) is an essential component of the γ-secretase complex which plays an important role in the pathogenesis of Alzheimer′s disease(AD).In addition to PEN-2,the γ-secretase complex consists of other components including presenilin(PS),nicastrin(NCT) and Anterior pharynx defective1(APH-1).PEN-2 has been demonstrated to be crucial for the endoproteolysis of the full-length PS to generate its N-terminal fragments and C-terminal fragments,which keep associated and interact with PEN-2 and other γ-secretase components.Moreover,PEN-2 is involved in promoting neuronal cell survival and protecting neurons from apoptosis in vivo.To further understand the function of PEN-2 in the pathology of AD,we utilized PEN-2 as a bait to identify PEN-2 interacting proteins in a yeast two-hybrid system.Two candidate proteins were identified:Homo sapiens poly(rC) binding protein 4(PCBP4) and Homo sapiens G protein-coupled receptor kinase 5(GRK5).The β-galactosidase activity assay demonstrated that the two proteins can interact with PEN-2 in yeast strain AH109.These results suggest that PCBP4 and GRK5 may play potential roles in facilitating the physiological functions of PEN-2.国家自然科学基金(30572077);; 厦门市海洋与渔业局基金(k53008)资
