TLR4和IFN-γ激活的间充质干细胞通过调节巨噬细胞极化改善血吸虫病肝纤维化
目的探讨共激活的间充质干细胞(MSCs)是否通过调节巨噬细胞极化发挥治疗血吸虫病的作用。方法取20只成年Balb/c雄性小鼠,随机分为四组,分别为未感染组、感染组、MSC组和MSCTLR4+IFN-γ组。通过腹部敷贴尾蚴法建立日本血吸虫感染模型。在感染后第5周,经灌胃给予小鼠吡喹酮进行抗虫治疗;感染后第6周,分别给予不同处理的MSCs治疗或未治疗。每周监测各组小鼠体质量变化率。采用HE染色和Masson染色观察肝组织病理变化,通过RT-qPCR检测肝组织中α-SMA和胶原蛋白mRNA水平,以评估肝纤维化程度;检测肝脏炎症因子和基质金属蛋白酶(MMP)的 mRNA水平,分析纤维化相关机制;通过RT-qPCR检测肝组织中i-Nos和Arg-1表达,并利用免疫荧光染色检测M1或M2巨噬细胞的比率,分析MSCs治疗与巨噬细胞极化的相关性;采用体外共培养体系,研究MSCs对巨噬细胞极化的直接调控作用。结果与感染组相比,MSCTLR4+IFN-γ组小鼠体质量增加(P<0.01),肝脏虫卵肉芽肿面积显著减少(P<0.001),α-SMA、Col-Ⅰ和Col-Ⅲ在RNA水平表达下降(P<0.01)。此外,MSCTLR4+IFN-γ组小鼠肝组织中炎症因子TNF-α和IL-1β表达下降(P<0.05),基质金属蛋白酶MMP2、MMP9和MMP13表达增高(P<0.01)。MSCTLR4+IFN-γ组较感染组显示M2标志物Arg-1 mRNA高表达(P<0.001),而M1标志物i-Nos表达下降(P<0.05);免疫荧光结果显示MSCTLR4+IFN-γ组肝脏i-Nos+细胞比率较感染组显著降低(P<0.05),而F4/80+CD206+细胞比率升高(P<0.000 1)。体外共培养实验进一步验证了这一现象:和MSCTLR4+IFN-γ共培养后,骨髓来源巨噬细胞中Arg-1在RNA水平表达升高,且促炎因子i-Nos和TNF-α的表达下降,ELISA结果与之相符。结论本研究表明,TLR4与IFN-γ协同激活的MSCs可减轻日本血吸虫感染小鼠的肝纤维化,该作用可能与激活MSCs调节巨噬细胞向M2型极化,发挥抗炎作用并促进肝脏细胞外基质降解相关
Optimization of solid culture conditions for TNF-α inhibitor qiaonanmycin via response surface methodology
目的利用响应面法对潜在的Tnf-α抑制剂桥南霉素固态发酵培养基进行优化。方法用HPlC做检测方法,以桥南霉素的洗脱峰面积为指标,先通过PlACkETT-burMAn(P-b)实验筛选出影响固态发酵产量的主要因素,进而进行单因素优化实验和响应面中心组合设计实验,确定主要影响因素的最佳浓度,得到最优的固体发酵培养基组成。结果优化后的发酵培养基组成:土豆400g/l、葡萄糖35g/l、蛋白胨10.5g/l、(nH4)2SO4 0.6g/l。按此配方固态发酵10l,对得到的粗提物进行HPlC检测,桥南霉素的实际产量为4.3Mg/l,与模型的理论值基本相符,较优化前的产量1.5Mg/l提高了186.7%。结论响应面方法是一种有效的培养基优化方法,经此优化方法桥南霉素的产量得到了较大的提高。Objective The solid fermentation culture conditions of compound qiaonanmycin were optimized via response surface methodology.Methods Based on quantitative detection method of qiaonanmycin by HPLC, the key fermentation factors were determined via P-B test, then their optimized concentrations were confirmed through response face method.Results The optimum solid fermentation culture medium compositions of qiaonanmycin were: potato 400g/L, dextrose 35g/L, peptone 10.5g/L, and(NH4)2SO4 0.6g/L.Under this condition, the yield of qiaonanmycin was 4.3mg/L, resulting in a 186.7% increase comparing with original production yield, and the optimized yield was consistent well with theoretical value predicted by response face method.Conclusion The optimized solid fermentation culture conditions were reliable and repeatable, it was warranted for large-scale fermentation process.中央高校基本业务费(No.2010121092); 厦门市科技计划项目(No.3502Z20123010

~(162)Lu的高自旋态及A=160区双奇核晕态能级的符号因子反转
利用147 ̄Sm(19 ̄F,4nγ)162 ̄Lu反应研究了162 ̄Lu的高自旋态。由7个带BGO康普顿抑制的高纯锗探测器和一个小平面探测器进行了在束γ测量,首次建立了双奇核162 ̄Lu转晕带的能级纲图。发现在低转动频率下,162 ̄Lu的转晕带能级发生符号因子反转。对质量数A=160核区双音核的转晕带重新进行了分析和考察,讨论了转晕带能级符号因子反转的系统性。High-spin states in 162Lu have been roduced and studied via 147Sm (19F, 4nγ) 162Lu reaction. lh-beam γ-rays were measured by using planar detector and 7 high purityGe detectors with BGO anti-Compton Shield. 一种新的角分布测量方法

双奇核~(170)Ta的转动带结构
用能量为105MeV 的~(16)O束流,通过~(159)Tb(~(16)O,5nγ)~(170)Ta 反应研究了~(170)Ta的高自旋态,观察到~(170)Ta 的3个转动带,其中一个耦合带和一个半退耦带中的非优先ΔI=2转动系列是由本工作发现的.文中还讨论了这些转动带可能的准粒子组态.High spin states in ~(170)Ta have been studied via ~(159)Tb (~(16)O,5nγ)~(170)Ta reaction at 105 MeV.Three rotational bands are observed,among which one coupled band and an unfavored ΔI=2 E2 squence are newly found in this work.The possible quasiparticle configurations of these bands are discussed.国家教委和国家自然科学基金资
