    Optoelectronic characteristics of InGaAs (P)/InP strained quantum well and superlattice

    【中文摘要】 用 Si H4 气体的减压 CVD法 ,在氧化硅以及石英基板上自然形成了高密度的 (~ 10 11cm-2 )纳米尺寸的半球状硅晶粒 (硅量子点 ) ,并且对其光学吸收和发光 (Photo- luminescence,PL)特性进行了评价。用表面热氧化了的硅量子点样品 ,在室温条件且在高于 1.2 e V以上的能量范围内观察到了 PL谱。随着量子点尺寸的减少 ,PL谱的光学吸收限移向高能方向。 PL谱的峰值能呈现大幅度的 (约 0 .9e V)斯塔克移动 ,并且 PL谱的强度几乎与温度无关 ,说明发光来自与局域能级相关联的发光和复合过程。 【英文摘要】 Using the low pressure CVD method with SiH 4 gas, we have accumulated the silicon quantum dots in self assembled and nano sized on a silica glass substrate. The optical absorption and photoluminescene spectra have been observed. With decreasing the size of silicon quantum dot, the limit of optical absorption shifts to higher energy.     New Method of Nanosilicon Preparation by Pulsed-laser Deposition

    采用高强度激光灼蚀沉积(PLD)的方式,在流动N2做为保护气体的情况下,成功制备出Si纳米结构,并利用扫描电镜(SEM)、光致发光(PL)进行表征。结果表明,该方法制备的纳米Si尺寸在几nm到几10nm之间,同时它具有较强的发光,发光强度比同等测量条件下的多孔Si(PS)样品高10倍以上,而且方法比较简单。The photo-luminescence of Si-based material is one of the most interesting and difficult subjects in research field.Nano-silicon will be a useful way to realize silicon-based photoluminescence.A new pulsed-laser-deposition(PLD) method to produce nano silicon is reported then according to SEM and photo-luminescence(PL) it′s characters are expressed.The results show that n-Si produced by the new pulsed-laser deposition(PLD) style has a great PL intensity.For the simpleness of new PLD style,it will be of some reference value for silicon-based PL rereach.国家自然科学重点基金资助项目(60336010

    THE EFFECTS OF COMBINATION OF CHINESE MEDICINE EFFECTIVE CONSTITUENTS ON CONFIGURATION AND COMPONENTS OF NUCLEAR MATRIX OF THE OSTEOSARCOMA MG-63 CELLS

    目的研究中药有效成分人参皂甙Rg1、肉桂酸CINN及丹参酮TanⅡA的组合(简称RCT),诱导人成骨肉瘤MG-63细胞分化过程中,细胞核基质构型与蛋白质组成的变化。方法应用流式细胞仪检测细胞周期变化,用四甲基偶氮唑蓝(MTT)法绘制细胞生长曲线,以选择性抽提整装光镜与电镜技术及蛋白质组学技术分析观察处理前后MG-63细胞核基质构型与蛋白质组成的变化。结果生长曲线显示,RCT处理后MG-63细胞的增殖受到明显抑制,增殖抑制率在第7天达到72.37%,细胞周期出现明显的G0/G1期阻滞。MG-63细胞核基质为典型的肿瘤细胞恶性表型,在RCT处理后发生了向正常细胞转变的恢复性变化。RCT处理后的细胞中多种核基质功能蛋白的表达发生了变化。结论RCT能显著抑制体外培养MG-63细胞的增殖活动,将细胞周期阻滞于G0/G1期,诱导核基质构型发生了显著的恢复性变化,并改变核基质蛋白的组成,对MG-63细胞具有显著的分化诱导作用。Objective To explore the alteration of nuclear matrix proteins and the configurational changes of nuclear matrix(NM) system during differentiation of the osteosarcoma MG-63 cells in vitro after being induced by the combination of ginsenoside Rg1,cinnamic acid and tanshinoneⅡA(RCT).Methods Cell cycle was investigated by flow cytometry and the growth curve was drawn with MTT method.The cells treated with or without RCT were subjected to selective extraction method and prepared for whole mount electron microscopy observation.The nuclear matrix-intermediate samples were examined under light and transmission electron microscopes respectively.The nuclear matrix proteins were subjected to analysis of proteomic technique.Results The proliferstion of the MG-63 cells treated with RCT were repressed markedly;the rate of repression was 72.37% on the seventh day,and the cell cycle was blocked at G_0/G_1 phase.The typical malignant phenotype of nuclear matrix of MG-63 tumor cell were reversed to normal cells.The expression of many functional proteins of nuclear matrix were changed in the MG-63 cells after the treatment with RCT.Conclusion RCT can repress markedly the proliferation of MG-63 cells,block cell cycle,induce a restorational change to the nuclear matrix architecture similar to that of normal cells,and change the constitutes of nuclear matrix proteins.Thereby RCT have a notable effect on MG-63 cells differentiation.国家自然科学基金资助项目(30470877);; 福建省自然科学基金资助项目(2008J0302);; 中国博士后科学基金资助项目(20070420754

    A Study on the Photoluminescence Spectroscopy of Silicon Nanocrystal Formed by Pulse-laser Deposition

    在激光灼蚀(PLD)系统中,采用流动的N2作为环境气体成功制备了尺寸从2纳米到几纳米之间的纳米硅,并在1.60~1.75eV之间观测到了较强的光致发光谱:结合Raman散射和光致发光谱的分析,推断强光致发光来源于纳米硅的量子效应。It has been successful to fabricate silicon nanocrystal from 2 nm to several nanometers by pulse-laser deposition using flowing N_2 as atmosphere gas.Strong photoluminescence between 1.6 eV and 1.75 eV has been observed.The origin of the photoluminescence was investigated by Raman scattering spectroscopy and photoluminescence spectroscopy.It was deduced that the strong pholuminescence stems from quantum effect.国家自然科学重点基金(批准号:60336010)资助项

    Changes in expression and localization of heat shock protein 70 during curcumin-induced apoptisis of human esophageal cancer cell line EC9706

    目的探讨姜黄素对人食管癌EC9706细胞凋亡的诱导作用,热休克蛋白70(HSP70)在肿瘤细胞凋亡过程中在核基质上的变化及其与凋亡调控相关蛋白的关系。方法用细胞计数和流式细胞仪检测姜黄素对人食管癌EC9706细胞的增殖抑制作用,以光学显微镜和透射电镜观察姜黄素诱导人食管癌EC9706细胞凋亡前后的细胞结构变化,琼脂糖凝胶电泳观察人食管癌EC9706细胞凋亡前后的dnA结构变化。双向凝胶电泳和质谱鉴定分析HSP70在核基质中的存在与变化;并以WESTErn blOTTIng进行确证;激光扫描共焦显微镜观察HSP70在EC9706细胞凋亡过程中的定位及其与bAX、bCl-2等基因产物的共定位关系。结果姜黄素能显著抑制人食管癌EC9706细胞增殖并诱导人食管癌EC9706细胞凋亡,双向凝胶电泳、质谱鉴定和结果发现并证实,HSP70在姜黄素处理前后的EC9706细胞核基质蛋白中的存在及其表达下调变化。激光扫描共焦显微镜观察结果显示,HSP70在EC9706细胞凋亡过程中与bAX、bCl-2等基因产物具有共定位关系,且其共定位区域发生了变化。结论姜黄素对人食管癌EC9706细胞具有显著的凋亡诱导作用;HSP70作为一种新发现的核基质蛋白,在姜黄素诱导人食管癌EC9706凋亡过程中的表达与分布发生了显著变化。HSP70与凋亡相关基因的关系对EC9706细胞凋亡具有重要影响。Objective To investigate the effect of curcumin on human esophageal cancer EC9706 cells and explore the role of heat shock protein(HSP70) in cell apoptosis by examining changes in the nuclear matrix and its relationship with apoptosis-related proteins.Methods Cell counting and flow cytometry were performed to probe the inhibitory effect of curcumin on cellular proliferation.Transmission electron microscopy and optical microscopy were used to observe the structural changes in EC9706 cells before and after apoptosis.Agarose gel electrophoresis was conducted to investigate the DNA structure of EC9706 cells before and after apoptosis.Two-dimensional polyacrylamide gel electrophoresis(2-D PAGE) and mass spectrometry(MS) analysis were performed to investigate the presence and changes of HSP70 in the nuclear matrix of EC9706 cells before and after curcumin treatment,which was further corroborated by Western blotting assay.Laser confocal scanning microscopy was used to observe the colocalization of HSP70 with Bax and Bcl-2 during apoptosis.Results The results indicated that curcumin could markedly inhibited EC9706 cell proliferation and finally induced apoptosis.Data from 2-D PAGE,MS,and Western blotting showed that HSP70 was involved in the nuclear matrix proteins and expression of HSP70 was downregulated after curcumin treatment.Laser confocal microscopy showed that HSP70 colocalized with Bax and Bcl-2,and the colocalized regions were altered by the curcumin treatment.Conclusion Our work proves that curcumin could definitely induce EC9706 cells into apoptosis.As a new found nuclear matrix protein,the expression and distribution of HSP70 are altered during the apoptosis of EC9706 cells.The colocalization of HSP70 with apoptosis-related genes evidently affects the apoptosis of EC9706 cells.福建省自然科学基金资助项目(2011J01256); 中央高校基本科研业务费专项资金(2011121061
