    提高水稻产量,改良稻米品质是育种学家广泛研究的课题.随着现代生物技术的发展,水稻已成为植物基因工程的重要研究对象.许多实验室已成功地建立了一系列供外源基因转化水稻的系统.但是这些转化系统主要应用TI质粒衍生的载体,通过T-dnA左右两端的序列将目的基...Many laboratories have set up several foreign gene transformation systems for rice.But these transformation systems were all used T DNA system of Ti plasmid, then the foreign gene integrated into plant genomes DNA by means of the insertion of both ends of T DNA.The integrated target of T DNA in plant genome DNA is random, while the thoroughly random integration would interfere with self stable system of plant genome, which could lead to pernicious mutation.In this study, according to the mechanism of homologous recombination, we had constructed a new donor plasmid for indica rice.It would cause the foreign gene intergated into chromsomal rDNA locus. According to the known sequence of ribosomal DNA, we cloned the 2.5kb fragment of it by PCR, and used the fragment as the integrated homologous sequence.After subcloning foreign DNA including nptII gene and metallothionein gene at the end of homologous fragment, the donor plasmid pURKMT which will integrate into the genomes DNA of indica rice was constructed.Then, donor plasmid pURKMT was introduced into calli of indica rice (Jiahe NO.7) by electroporation.Screened by kanamycin, the transformant calli which was integrated the foreign DNA were selected.Dot blotting data showed that the foreign DNA including nptII gene and MT gene has integrated into rice genome DNA

    Terahertz time-domain spectroscopy for MCT ceramics

    通过固相反应法制备MgTIO3-CATIO3陶瓷,采用太赫兹时域光谱系统测试该陶瓷样品的时域光谱,经过计算得到0.2 THz~2.5 THz频率范围内陶瓷的电容率和吸收系数;介质陶瓷的电容率随烧结温度升高先增大后减小,烧结条件为1 260℃x3 H的陶瓷性能最佳,在1 THz处电容率为18.68,吸收系数为8.52 CM-1,可望应用于太赫兹整机设备中的材料与元器件。Ceramics of MgTiO3-CaTiO3 are prepared via solid-state reaction route.Terahertz time-domain spectroscopy is adopted to obtain permittivity and absorption coefficient of the samples.It is found that the best properties for the ceramics sintered at 1 260 ℃ for 3 h are displayed with permittivity of 18.68 and absorption coefficient of 8.52 cm-1 at 1 THz.This kind of ceramics may be suitable for application of Terahertz devices.国家863计划资助项目(2001AA325100

    由于SI/SIgE异质结构的带阶差主要发生在价带,为实现高效率的发光,本文从理论上设计了在硅基SI1-XgEX虚衬底上外延应变补偿的SI/S1-ygEy(y>X)量子阱的能带结构,将量子阱对电子的限制势垒提高到100MEV以上。在实验上,采用300℃生长的gE量子点插入层,制备出薄的SIgE驰豫缓冲层(虚衬底),表面gE组份达到0.25,表面粗糙度小于2nM,驰豫度接近100%。在我们制备的SIgE缓冲层上外延了应变补偿SIgE/SI多量子阱结构,并初步研究了其发光特性。In this paper,band structures of strain compensated Si/S_(1-y)Ge_y(y>x) quantum well grown on Si_(1-x)Ge_x virtual substrate was design to enlarge the conduction band offset up to 100meV for improving luminescence.The fully strain-relaxed Si_(0.75)Ge_(0.25) virtual substrate was prepared by inserting a low-temperature Ge islands layer in ultra-high vacuum chemical deposition.The root-mean-square surface roughness of the virtual substrate is less than 2nm.The luminescence of the strain compensated Si/SiGe quantum well on the virtual substrate was investigated.国家重点基础研究发展计划资助项目2007CB613400;国家自然科学基金资助项目(60676027;50672079

    Enhanced Visible Light Photoelectrochemical Performances with Nitrogen Doped TiO_2 Nanowire Arrays

    成功制备了氮掺杂锐钛矿TiO2纳米线,并研究了它的光电化学性质.结果表明,与商用P25 TiO2纳米粒子和未掺杂TiO2纳米线相比,氮掺杂TiO2纳米线作为光阳极明显地提高了光电转换效率(IPCE%),在可见光区有明显光吸收;在100 mW/cm2可见光光照下,氮掺杂TiO2纳米线具有最大的光电流密度和能量转换效率.例如,当电压为0.09 V(vs.Ag/AgC l)时最大能量转换效率为0.52%,均高于未掺杂TiO2纳米线和商用P25 TiO2纳米粒子的,充分表现出它优越的光响应和光电化学性能,在光电化学池、太阳能制氢等方面具有广泛的应用前景.Self-organized anodic anatase TiO2nanowire arrays doped with nitrogen have been successfully fabri-cated and their photoelectrochemical(PEC) properties have been characterized and found to be substantially im-proved compared to undoped nanowires or commercial P25 nanoparticles.Photocurrent measured with monochro-matic incident light showed that the incident photon-to-current efficiency(IPCE,%) values of nanowire arrayelectrodes with or without N-doping were obviously higher than that of commercial P25 nanoparticle electrodes,and nitrogen-doped TiO2nanowire arrays(NTNA) had noticeable absorption in the visible region.The NTNAelectrodes showed the highest photocurrent density and power conversion efficiency under 100 mW/cm2visiblelight illumination.A maximumolphotoconversion efficiency of 0.52% was achieved for the NTNA sample at anapplied potential of 0.09 V versus Ag/AgCl(saturated KCl) electrode under visible illumination,much higherthan that of the undoped nanowire and commercial P25 nanoparticle electrodes.These results demonstrate thatNTNA thin films are promising for enhancing the photoresponse and effectively improving PEC performances ofnanostructured TiO2in the visible region for different applications including solar hydrogen generation.作者联系地址:清华大学化学系;美国加利福尼亚大学Santa Cruz分校化学与生物化学系;Author's Address: 1.Department ofChemistry,Tsinghua University,Beijing100084,China;2.Department ofChemistry and Biochemistry,University ofCalifornia,Santa Cruz,California95064,US
