    玻碳电极表面复合配位银电结晶机理研究

    以具有实际应用价值的复合配位体系无氰镀银电解液为研究对象,运用循环伏安和电位阶跃等实验方法,结合Scharifker-Hill经典理论模型分析,成功获得了Ag在玻碳电极(GCE)表面电沉积的成核机理及成核动力学参数,并分析了温度对成核方式及成核动力学参数的影响.结果表明,该体系下Ag在GCE表面的电沉积是由扩散控制的不可逆过程,遵循三维瞬时成核生长机理.随着阶跃电位从-750 m V负移至-825 m V,峰值还原电流Im逐渐增大,达到峰值还原电流所需时间tm逐渐缩短,扩散系数D变化不大,基本稳定在(7.61±0.34)×10-5cm2·s-1,成核密度数N0则从3.26×105cm-2提高至10.2×105cm-2.银沉积初期的形貌观察,验证了其三维瞬时成核生长机理.提高温度可以显著改善电解液中具备活性的银配位离子的扩散能力,缩短成核时间,提升成核密度数N0.国家自然科学基金项目(No.21621091)资

    不同季节垃圾填埋场周围重金属污染特征及评价

    采集漳州市某生活垃圾填埋场周围冬夏两季的地表水(包括渗滤液)、土壤以及植物样品,分析9种重金属总量的季节性变化.结果表明,冬季地表水中Zn和As,土壤和植物中的Cd和As的含量均大于夏季,而夏季波动性强于冬季;夏季渗滤液中总Hg的含量明显高于冬季,其余变化不明显.地表水重金属含量均低于地表水环境质量Ⅴ类标准限值,渗滤液中所有重金属含量均低于污水排放二级标准限值.土壤中重金属Cd、As和Pb的污染最重,已达到重度污染,冬季的污染明显高于夏季.冬季植物富集能力强于夏季,细叶芒对Cd的富集能力较强,可作为重金属污染修复的先锋植物.环保公益性行业科研专项(201509054)资助~

    目的研究石决明提取物对体外培养的晶状体氧化应激性白内障形成的作用及机制。方法离体培养小鼠晶状体,应用不同浓度的石决明提取物预孵育晶状体24h后,加入1mm过氧化氢,继续培养3小时后恢复正常培养,72小时后观察小鼠晶状体混浊程度,测定晶状体组织培养液中的乳酸脱氢酶(LDH)含量,晶状体组织中还原型谷胱甘肽(GSH)含量及超氧化物歧化酶(SOD)活力。结果石决明提取物在1~2 mg/ml浓度范围内减轻氧化应激造成的晶状体混浊,减少晶状体LDH的释放,提高组织内GSH含量和SOD活力。结论石决明提取物可减轻氧化应激白内障的形成,其作用主要与石决明提取物提高内源性抗氧化系统有关。Objective To study the effect of the abalone shell extract on oxidative stress induced cataract formation and its mechanism in cultured mouse lens in vitro. Methods The cultured mouse lens were pretreated with the abalone shell extract in different concentrations for24 hours,and then 1mm hydrogen peroxide was added and continued incubating for 3 hours,and they were changed to normal culture media.After 72 hours,the opacity of each lens was observed under an anatomical microscope,the content of lactate dehydrogenase( LDH)leakages,the content of the reduced glutathione( GSH) and activity of superoxide dismutase( SOD) in lens tissue were assayed. Results Abalone shell extract in the concentration range of 1 ~ 2 mg / ml reduced the lens opacity caused by oxidative stress,alleviated the release of LDH,and increased GSH content and SOD activity in cultured lens.     Study on the free radical scavenging activity of polyphenolic compounds by on-line high-flux screening method

    目的:构建植物化学物在线清除自由基活性高通量筛选技术,创建植物化学物抗氧化活性研究新方法。方法:以附带2个检测器的高效液相色谱(HPlC)装置和流动注射技术为手段,引入二苯基三硝基苯肼(dPPH)有机自由基分别与经HPlC分离的植物化学物发生在线清除反应,优化流速、反应管尺寸等条件,比较反应前后dPPH吸收信号的变化。结果:确定和验证了两类植物多酚化学物茶多酚和丹酚酸在线清除自由基活性HPlC-dPPH表征方法。反应环聚醚醚酮(PEEk)盘管规格为长10M、内径0.254MM,dPPH·浓度为1.0x10-5MOl/l,流速为0.7~0.8Ml/MIn。结论:HPlC-dPPH流动注射在线表征技术能够实现对多组分植物化学物抗氧化能力的高通量筛选。Objective:To construct a high-flux screening procedure by on-line free radical scavenging activity method of phyto- chemicals, and create a new evaluation system of antioxidant activity of natural plant materials.Methods:A HPLC system with two detectors and flow injection techniques was employed.An organic free radicals DPPH was induced on-line HPLC-DPPH and reaction with the single phytochemical components isolated by HPLC.To optimize reactive conditions including flow rate and the size of reaction coil, and detect the DPPH signals before and after reaction.Results:A new on-line HPLC-DPPH method was established for a high-flux screening of several free radical scavenging components exampled as Tea polyphenols and Salvianolic acids.The reaction coil of PEEK was 10 m length and inner diameter 0.254 mm, the concentration and flow velocity of DPPH was 1.0 × 10-5mol/L and 0.7~0.8 ml/ min, respectively.Conclusion:The PHLC-DPPH flow injection hyphenated technique could realize high-flux screening analysis for antioxidant of phytochemicals.江苏省高校自然科学研究计划资助项目(05KJB330083

    Determination of Salvianolic acid B by High Performance Liquid Chromatography with Fluorescence Detection

    建立了丹酚酸B的高效液相色谱-荧光检测含量测定方法。色谱条件为:BACKMANODS柱( 4 6mmi.d .×2 5 0mm ,5 μm) ,以甲醇-0 5 %冰醋酸水溶液为流动相,流速为1 0mL/min ;荧光检测波长λEX=3 3 8nm ,λEM=44 3nm ;进样量5 μL。丹酚酸B进样量为0 12~1 2 μg时,其质量与峰面积呈良好的线性关系(r=0 9998) ;加样回收率为98 6%~10 8% ;最低检测限为0 0 13 μg/mL。该法具有更高的灵敏度,为丹酚酸B的药代动力学研究提供基础A new method for the determination of Salvianolic acid B by high performance liquid chromatography with fluorescence detection has been established.The column used was BACKMAN C 18 (4.6mm i.d.×250mm,5μm);The mobile phase was methanol-0.5% acetic acid solution with gradient condition at a flow rate of 1.0mL/min;The fluorescence detector was operated at λ EX =338nm and λ EM =443nm with the injection volume of 5μL.The result proved that Salvianolic acid B has a good linearity in the range of 0.12μg to 1.2μg with the correlation coefficient of 0.9998;The recoveries were 98.6%~108%;The limit of detection is 0.013μg/mL.This method has a higher sensitivity and can be used in the study of pharmic metabolize.国家自然科学基金重点项目(2 0 2 350 2 0
