    The Atmospheric Mercury Pollution Survey During Autumn and Winter Seasons in Xiamen

    文章采用两次金汞齐-冷原子荧光光谱法对厦门地区秋冬季节大气汞的污染状况作了初步调查分析,结果表明厦门地区大气中汞的浓度平均为11.037 ng/M3,其含量高于北半球大气气态总汞含量的背景值(1.5--2.0 ng/M3),表明该地区已受到一定程度的大气汞污染;在一定的采样区域,大气汞浓度与温度、太阳辐射量、大气压、空气湿度成正相关,与风速成负相关;气态汞的含量在80%左右,这与该地区的人为活动多有关,与其他地区报道的类似区域情况的汞污染状况相符;工业区,学校实验室周边区域,生产过程中以汞为原料的工厂等附近区域,大气中汞的含量较高,开阔区域的海边含量较低,交通繁忙区域最低(所选采样点与交通要道有一定的距离),这种浓度分布说明了大气中汞的存在具有区域性特征。In this paper,the atmospheric mercury pollution during autumn and winter seasons in Xiamen was analyzed with the method of two-stage gold amalgamation-cold vapor atomic fluorescence spectroscopy.The results show that the average concentration of atmospheric mercury in Xiamen is 11.037ng/m3,which is higher than the background value(1.5ng/m3--2.0ng/m3) of the atmospheric mercury in the northern hemisphere.It indicates that there is a certain degree of mercury pollution in Xiamen.there is a positive relationship between mercury concentration and temperature、solar radiation、atmospheric pressure and humidity,while there is a negative relationship between mercury concentration and wind speed.The content of gaseous mercury in Xiamen is about 80%,which shows analogous level compared to the mercury concentration in other areas.The high content of gaseous mercury in Xiamen maybe related to the human activities.The mercury concentration is high in some regions such as industrial zone,areas around school laboratories,factories which use mercury as raw materials,while the mercury concentration is a little lower in the open area of the seaside,and the mercury concentration is the lowest in traffic busy areas(there is some distance between sampling locations and traffic busy in this study).The distribution of mercury concentration indicates that there is a regional character of atmospheric mercury in Xiamen

    目的探讨姜黄素对人食管癌EC9706细胞凋亡的诱导作用,热休克蛋白70(HSP70)在肿瘤细胞凋亡过程中在核基质上的变化及其与凋亡调控相关蛋白的关系。方法用细胞计数和流式细胞仪检测姜黄素对人食管癌EC9706细胞的增殖抑制作用,以光学显微镜和透射电镜观察姜黄素诱导人食管癌EC9706细胞凋亡前后的细胞结构变化,琼脂糖凝胶电泳观察人食管癌EC9706细胞凋亡前后的dnA结构变化。双向凝胶电泳和质谱鉴定分析HSP70在核基质中的存在与变化;并以WESTErn blOTTIng进行确证;激光扫描共焦显微镜观察HSP70在EC9706细胞凋亡过程中的定位及其与bAX、bCl-2等基因产物的共定位关系。结果姜黄素能显著抑制人食管癌EC9706细胞增殖并诱导人食管癌EC9706细胞凋亡,双向凝胶电泳、质谱鉴定和结果发现并证实,HSP70在姜黄素处理前后的EC9706细胞核基质蛋白中的存在及其表达下调变化。激光扫描共焦显微镜观察结果显示,HSP70在EC9706细胞凋亡过程中与bAX、bCl-2等基因产物具有共定位关系,且其共定位区域发生了变化。结论姜黄素对人食管癌EC9706细胞具有显著的凋亡诱导作用;HSP70作为一种新发现的核基质蛋白,在姜黄素诱导人食管癌EC9706凋亡过程中的表达与分布发生了显著变化。HSP70与凋亡相关基因的关系对EC9706细胞凋亡具有重要影响。Objective To investigate the effect of curcumin on human esophageal cancer EC9706 cells and explore the role of heat shock protein(HSP70) in cell apoptosis by examining changes in the nuclear matrix and its relationship with apoptosis-related proteins.Methods Cell counting and flow cytometry were performed to probe the inhibitory effect of curcumin on cellular proliferation.Transmission electron microscopy and optical microscopy were used to observe the structural changes in EC9706 cells before and after apoptosis.Agarose gel electrophoresis was conducted to investigate the DNA structure of EC9706 cells before and after apoptosis.Two-dimensional polyacrylamide gel electrophoresis(2-D PAGE) and mass spectrometry(MS) analysis were performed to investigate the presence and changes of HSP70 in the nuclear matrix of EC9706 cells before and after curcumin treatment,which was further corroborated by Western blotting assay.Laser confocal scanning microscopy was used to observe the colocalization of HSP70 with Bax and Bcl-2 during apoptosis.Results The results indicated that curcumin could markedly inhibited EC9706 cell proliferation and finally induced apoptosis.Data from 2-D PAGE,MS,and Western blotting showed that HSP70 was involved in the nuclear matrix proteins and expression of HSP70 was downregulated after curcumin treatment.Laser confocal microscopy showed that HSP70 colocalized with Bax and Bcl-2,and the colocalized regions were altered by the curcumin treatment.Conclusion Our work proves that curcumin could definitely induce EC9706 cells into apoptosis.As a new found nuclear matrix protein,the expression and distribution of HSP70 are altered during the apoptosis of EC9706 cells.The colocalization of HSP70 with apoptosis-related genes evidently affects the apoptosis of EC9706 cells.福建省自然科学基金资助项目(2011J01256); 中央高校基本科研业务费专项资金(2011121061

    钯纳米粒子及其团聚体特殊红外性能的CO分子探针红外光谱

    以聚乙烯吡咯烷酮(PVP)为稳定剂, 用乙醇还原氯钯酸制备分散的钯纳米粒子(Pdn). 从分散于电极表面的Pdn出发, 以50 mV·s-1的扫描速度, 在 0.25~1.25 V间循环扫描30 min可以制备团聚体 Pdnag. 固/气和固/液界面透射和反射CO分子探针红外光谱研究结果指出, 吸附在Pdn上非对称和对称桥位上的桥式吸附态CO(COB)在1964 和 1905 cm-1给出两个红外吸收谱峰, 但CO吸附在Pdnag上仅在1963 cm-1给出一个方向倒反的异常红外谱峰. 此外, CO谱峰的半峰宽由Pdn上的14 cm-1变为Pdnag上的24 cm-1. Pdn形成团聚体Pdnag后, Pd纳米粒子间相互作用增强, Pdn和Pdnag红外光学性质的显著差别初步归因于纳米粒子之间的相互作用

    Effects of curcumin on anti-proliferation and expression of apoptosis interrelated gene of Human Osteosarcoma MG-63 Cells

    目的:探讨姜黄素对人成骨肉瘤Mg-63细胞增殖抑制及凋亡相关基因表达的影响。方法:用姜黄素处理Mg-63细胞,细胞计数法检测细胞增殖抑制的效果;荧光染色观察细胞的凋亡;流式细胞仪(fCM)进行细胞周期时相分析;免疫细胞化学法和WESTErn blOTTIng免疫法检测细胞凋亡相关基因的表达水平。结果:随着姜黄素浓度的增加及其作用时间延长,对细胞的增殖抑制作用增强,最高抑制率可达89.07%;光镜下可观察到细胞发生染色质浓缩,细胞核凝聚和碎裂等典型的凋亡形态学改变;fCM检测结果显示细胞经姜黄素处理后出现明显的凋亡峰;免疫反应结果显示,凋亡相关基因bCl-2和P53表达水平降低,而bAX和fAS表达水平升高。结论:姜黄素能显著抑制Mg-63细胞增殖并可有效诱导其凋亡,其作用机制可能与bCl-2和bAX二者的比值发生变化从而接受了凋亡刺激信号有关。Objective: To investigate the regulatory effect of curcumin on proliferation inhibition and apoptosis in human osterosarcoma MG-63 cells.Methods: MG-63 cells were treated by curcumin, and the inhibitory effect on cell proliferation was assessed by cell count.The apoptosis was inspected by fluorescent staining.The cell cycle analysis was detected by FCM.The expression level of genes related with apoptosis was observed by immunohistochemistry and western blotting immunoreaction.Results: The results showed that the cells growth was inhibited with the increasing of the concentration of curcumin and the elongation of the treated time,and the highest inhibition rate was 89.07%.Parts of cells presented the typical morphological changes of apoptosis: chromatin concerntrated and agglutinatied, cell nucleus shrinked and cataclased under the microscope.The results of FCM analysis showed that curcumin could in- duce cells into apoptosis, which was proved by the emergence of apoptotic peak.The results of immunoreaction suggested that the expression level of bcl-2 and P53 were down-regulated and the expression level of bax and Fas were upregulated.Conclusions: Curcumin not only inhibited cell proliferation but also induced apoptosis of MG-63 cells, which may be related to the ratio of the expression level of bcl-2 and bax as well as reception of the signal of stimulating apoptosis.国家自然科学基金(30871241

    学籍信息管理是学校教务管理的重要内容,伴随着计算机技术尤其是数据库技术和网络技术的发展,学籍信息管理系统应运而生,并且得到快速发展和应用.当前学籍信息管理系统一般都集中于学籍静态信息的管理,例如学生基本信息,异动信息等,而对于一些动态信息的管理,例如学生学期注册或上课签到等,由于无法提供这些动态信息的有效性和真实性识别能力,目前还是采用传统的书面签字方法.为此,结合厦门大学新型教务管理系统的开发,采用指纹识别技术,构建了针对学籍信息管理系统模块的远程签到系统.引入指纹考勤功能模块后,可以有效地避免代签等问题,对于保证出勤率和教学质量具有积极意义.Based on the complexity and variety of student status information and the dramatic increase in the number of students and the function & performance deficiency of Xiamen University′s old student status information management system,this article discusses the design and development of new student status management system which meets the needs of current teaching reform.It also shows the major work which the author had done during the design and development of this new system.New student status management system supports more complex modules.The innovations in this paper include:the specific functions are separated from common functions;real-time synchronization of students′ status;function of setting up photos;variable field of modifying the authorities;function of signing in with fingerprint identification.国家863计划项目(2006AA01Z129);国家重点基础研究发展计划(973计划)(2007CB311005);厦门大学985二期信息创新平台项
