    Hepatitis C virus (HCV) NS5B protein is the viral RNA- dependent RNA polymerase capable of directing RNA synthesis. In this study, an electrophoretic mobility shift assay demonstrated the interaction between a partially purified recombinant NS5B protein and a 3’ viral genomic RNA with or without the conserved 98-nucleotide tail. The NS5B-RNA complexes were specifically competed away by the unlabeled homologous RNA but not by the viral 5’ noncoding region and very poorly by the 3’ conserved 98- nucleotide tail. A 3’coding region with conserved stem-loop structures rather than the 3’ noncoding region of the HCV genome is critical for the specific binding of NS5B. Nevertheless, no direct interaction between the 3’ coding region and the HCV NS5A protein was detected. Furthermore, two independent RNA- binding domains (RBDs) of NS5B were identified, RBD1, from amino acid residues 83 to 194, and RBD2, from residues 196 to 298. Interestingly, the conserved motifs of RNA-dependent RNA polymerase for putative RNA binding (220-DxxxxD-225) and template/primer position (282-S/ TGxxxTxxxNS/T-292) are present in the RBD2. Nevertheless, the RNA-binding activity of RBD2 was abolished when it was linked to the carboxy- terminal half of the NS5B. These results provide some clues to understanding the initiation of HCV replication

    [[abstract]]護理資訊系統有助於提升臨床作業品質及效率,成效評值除了可以了解系統的效益外也可以作為優化的重點指引,由於在臨床作業中,護理資訊系統扮演的是輔助護理照護的功能,協助護理人員完成護理紀錄,進而提升護理紀錄品質向來是護理資訊努力的方向之一。資訊系統成功模式為一種可針對資訊系統進行多構面且具因果關係的評估模式,其亦可呈現各資訊因子與使用者滿意度、淨效益之間的關聯,被廣泛用於各類領域評值資訊系統成效。本研究的目的在於探討護理紀錄系統導入之成效,研究方法乃以資訊系統成功模式為架構,編修「護理紀錄系統成效問卷」探討使用者端效益,並分析研究醫院護理紀錄品質審查結果,探討系統導入前後,護理紀錄品質之差異。「護理紀錄系統成效問卷」以使用系統的護理人員為研究對象,收案143份,結果分析,24題項中有19題得分高於中位數,各構面得分依序為資訊品質(2.68)、淨效益(2.65)、使用者滿意度(2.55)、服務品質(2.51)、系統品質(2.46),使用者滿意度與系統品質(r=0.713**)、資訊品質(r=0.778**)、服務品質(r=0.708**)皆有正相關,淨效益與系統品質(r=0.581** )、資訊品質( r=0.689**)、服務品質(r=0.649**)亦有正相關,而3項資訊因子與使用者滿意度的相關性皆高於與淨效益的相關性,使用者滿意度與淨效益則有高度相關(r=0.827**)。在系統導入前、後護理紀錄品質差異方面,分析結果於護理指導、出院護理指導及出院護理紀錄部分品質明顯提升。故而資訊科技導入時,應提升資訊因子品質以使系統發揮最好效益,亦建議持續執行系統評值,了解使用情形並做為系統優化重點考量。[[abstract]]Improving the quality of nursing records by users has always been one of the directions of the nursing information system. The information system success model is a multi-faceted and causal evaluation model for information systems. It shows the correlation between information factors and user satisfaction and net benefits. It is widely used in various fields of evaluation of technology informatics. The purpose of this study is to evaluate the nursing documentation information system. The research method is based on the successful model of the information system, “Nursing Record System Effectiveness Questionnaire” (CVI 0.9, Cronbach's α 0.82) to explore user benefits, and analyze the difference of nursing records quality between before and after the nursing documentation information system intervention. The “Nursing Record System Effectiveness Questionnaire” collected 143 cases. The scores of all category, high to low sort, were of information quality (2.68) and net benefits (2.65), user satisfaction (2.55), service quality (2.51), system quality (2.46). User satisfaction and system quality (r=0.713**), information quality (r=0.778**), service quality (r=0.708**) is positively correlated. Net benefit is positively correlated with system quality (r=0.581**), information quality (r=0.689**), service quality (r=0.649**), and user satisfaction was highly correlated with net benefit (r=0.827**). The results that difference of nursing records quality between before and after system intervention were significantly different in nursing education、discharge nursing education and discharge nursing record . Therefore, information technology intervene, the quality of information factors should be improved to make the system work best. It is also recommended to continuously implement system evaluation to understand the situation and make it a key consideration for system optimization

    [[abstract]]護理記錄是護理人員對於病人照護過程所做的記載,紀錄若未整合或反覆謄寫容易產生錯誤,且影響病人安全,故運用資訊科技有助於提升護理紀錄的正確性與完整性,進而整合照護資訊、簡化流程,提升照護品質。本研究目的是資訊科技導入護理紀錄之使用者評值成效。本研究方法一為以資訊系統成功模式為架構之護理紀錄系統成效問卷調查表,進行使用者系統評值,二為分析護理紀錄審查結果於護理紀錄系統導入前後的內容差異。本研究問卷收案143份,各構面得分依序為資訊品質(2.68)、淨效益(2.65)、使用者滿意度(2.55)、服務品質(2.51)、系統品質(2.46),資訊因子與使用者滿意度及淨效益皆呈正相關。護理紀錄於護理措施、出院護理指導及出院護理紀錄於系統導入後紀錄品質明顯提升。結果顯示使用者對於護理紀錄系統導入評價趨於正向,故資訊科技導入時,應以提升資訊因子品質以使系統發揮最好效益;系統導入初期,護理紀錄品質差異可能僅呈現於部分項目,須持續追蹤評值
