    Preparation and application of monoclonal antibodies against Herpes simplex virus-1

    目的:制备并筛选HSV-1单抗,建立定量检测HSV-1病毒颗粒抗原的双抗体夹心ElISA方法,用于HSV-1病毒颗粒的质控。方法:以HSV-1免疫bAlb/C小鼠制备单克隆抗体,以筛选的中和单克隆抗体1f6为捕捉抗体,HrP标记的2b1为检测抗体,构建定量检测HSV-1病毒颗粒抗原的双抗体夹心ElISA方法,并对本方法的特异性、灵敏度、精密度、准确性和线性等性能进行验证。用本方法定量检测的病毒量与病毒滴度作回归分析。结果:构建的双抗体夹心定量检测HSV-1病毒颗粒抗原的ElISA方法,线性范围为0.125~2μg/Ml,相关系数为r2=0.995 5,定量限度为0.125μg/Ml,试剂的变异系数CV<10%,抗原回收率介于85.6%~107.1%之间。与HSV-1以外的其他样本无交叉反应。本方法检测与病毒感染滴度具有很好的相关性。结论:成功构建了定量检测HSV-1含量的ElISA方法,为HSV-1病毒颗粒抗原定量检测提供快速手段。Objective: To prepare and screen monoclonal antibodies against Herpes simplex virus-1( HSV-1),and develop a double antibody sandwich quantitative enzyme-linked immunosorbent assay( Q-ELISA) for detection of HSV-1 particle.This method was used to control the quality of viral particle in the developing and manufacturing process of HSV-1.Methods: BALB / c mice was immunized with HSV-1 to prepare monoclonal antibodies.A double antibody sandwich Q-ELISA was developed to determine concentration of HSV-1 particle,which was based on the neutralizing monoclonal antibody 1F6 as capture antibody,and 2B1 as HRP-conjugated antibody.The performance of the reagent was evaluated,including specificity,sensitivity,precision,accuracy and linear.And the relation between the amount of virus detected by this method and the virus titer was analyzed by regression analysis method.Results: The QELISA for HSV-1 particle was developed.The quantitation scope was 0.125- 2 μg / ml,the coefficient correlation was 0.995 5,the limit of detection was 0.125 μg / ml,the recovery was between 85.6% and 107.1%,the variation coefficient was lower than 10%,and the reagent does not react with other samples except HSV-1 antigen.This method has a good correlation with virus titer.Conclusion: The QELISA for HSV-1 particle was successfully developed,which provide a new approach for rapid and quantitative detection of HSV-1 antigen

    MCML电路由于具有高速低摆幅、抗干扰能力强、在高频下比传统CMOS电路功耗更低等优点,越来越受到广泛关注.通过分析二值MCML电路的设计方法,引入与参考电压进行比较的思路,设计了一种结构简单的新型高性能三值D型触发器.采用TSMC 180 nm工艺,使用HSPICE进行模拟.结果表明,所设计的触发器不仅具有正确的逻辑功能,工作频率达到10 GHz,平均D-Q延时和PDP也比传统CMOS三值触发器有明显降低,且随着工作频率的上升,PDP不断下降,适合于高速和高工作频率的应用

    辅酶与酶蛋白组成结合蛋白酶,而结合蛋白酶催化反应的专一性取决于所含的辅酶,因此有必要建立辅酶的灵敏测定方法,并用于结合蛋白酶的催化动力学行为、构象及活性等化学特性的表征.实验表明辅酶II显著猝灭铽钛铁试剂络合物的荧光.本文推导了ΔF与辅酶II初始浓度的线性关系式(ΔF=nk[Q]0 ),并据此建立了辅酶II的高灵敏荧光分析方法.研究了荧光光谱、缓冲液、pH范围、铽离子和钛铁试剂最佳浓度、灵敏度、检测限和干扰等分析化学特性.辅酶II在40. 0nmol/L~1. 0μmol/L浓度范围内呈良好的线性,线性回归方程:ΔF=0. 868C(nmol/L) -13. 1(R2 =0. 994),检测限:1. 63×10-8 mol/L,且牛血清白蛋白及其它生物学中的常见离子在一定范围内均不干扰测定.结果表明,该方法用于测定辅酶II操作简便,测定快速,并预示着该方法可应用于若干含磷酸基的重要辅酶的测定.Conjugated proteases are composed of apoenzymes and coenzymes.The specificity of the enzyme catalysis is dependent on the coenzyme in the conjugated protease.It is necessary to develop a sensitive method for the determination of coenzymes,by which some chemical properties of conjugated proteases,such as its dynamics behavior,construction and activity,can be realized deeply.The experiment shows that NADP could remarkably quench the fluorescence intensity of Tb~(3+)-Tiron complex in Tris-HCl buffer solution of pH=7.60±0.01,and a sensitive fluorescence quenching method was developed for the determination of NADP.The quantitative formula was theoretical induced.It was demonstrated that the ΔF was directly proportional to and the beginning concentration of NADP(ΔF=nk[Q]_0).The analytical characteristics, such as fluorescence spectrum,buffer solution,pH range,optimum concentrations of Tb~(3+) and Tiron,sensitivity,detection limit and reference,etc.,were studied.A linear concentration range for NADP determination was obtained from 40.0 nmol/L to 1.0μmol/L,and its linear regression equation is ΔF=0.868C(nmol/L)-13.1 (R~2=(0.994)).The detection limit is high up to 1.63×10~(-8) mol/L.The bovine serum albumen and frequent ions in biology can’t interfere the determination of NADP within limits.The obtained results implied that the proposal method should find its application to the (determination) of other important coenzymes,which contain phosphates

    目的:探讨婚姻关系对双方均为唐山地震幸存脊髓损伤(SCI)患者夫妇的生活满意度和心理状况的影响。方法:调查1976年唐山地震幸存SCI患者40名,其中,生活在截瘫康复村已婚者20名(男、女各 10 人),生活在疗养院未婚者 20 名(男、女各10人),比较婚姻状况对两组患者的生活满意度、生活质量、焦虑和抑郁情绪状况的影响。结果:已婚组 SCI患者对目前生活的满意度明显高于未婚组( P 0.05); 焦虑平均得分高于未婚组( P 0.05)。结论:婚姻对提高唐山地震幸存SCI患者的生活满意度和心理健康水平有良好作用

    根据城轨列车驱动电机与齿轮箱之间连接的需要,设计了用于城轨列车的一种齿式联轴节。在设计过程中,从齿式联轴节结构、齿部的几何参数、密封结构、齿部侧隙的理论计算、空间运动分析等方面进行了研究;建立了齿式联轴节的三维模型,基于Ansys计算软件对齿式联轴节的3种运行工况进行了有限元分析,并运用ZGCAD齿轮软件进行了齿轮强度计算;设计制造齿式联轴节样机,进行了样机周期性循环试验。研究结果可为其他领域齿式联轴节的设计提供借鉴和参考

    利用147 ̄Sm(19 ̄F,4nγ)162 ̄Lu反应研究了162 ̄Lu的高自旋态。由7个带BGO康普顿抑制的高纯锗探测器和一个小平面探测器进行了在束γ测量,首次建立了双奇核162 ̄Lu转晕带的能级纲图。发现在低转动频率下,162 ̄Lu的转晕带能级发生符号因子反转。对质量数A=160核区双音核的转晕带重新进行了分析和考察,讨论了转晕带能级符号因子反转的系统性。High-spin states in 162Lu have been roduced and studied via 147Sm (19F, 4nγ) 162Lu reaction. lh-beam γ-rays were measured by using planar detector and 7 high purityGe detectors with BGO anti-Compton Shield.
