    利用ISSr分子标记对来自10个不同生态地区的红锥优良种质进行基因组遗传多样性分析.从20条引物中筛选出11条具有扩增多态性的引物用于ISSr-PCr扩增,共扩增出435条带,其中多态性条带325条,占总扩增条带的74.7%,平均每个引物扩增3.95条.ISSr标记分析揭示出红锥10个优良种质的遗传相似系数介于0.473 7--0.808 1之间,平均值为0.728 7.用STATIS统计软件的欧氏可变类平均法构建类型之间的分子系统树,聚类分析结果将10个优良红锥种质分为4个类群.Genetic diversity of fine germplasm resources of Castanopsis hystrix from ten different ecological regions was analyzed by using the inter-simple sequence repeat(ISSR),and 11 effective primers were selected from 20 primers.The result showed as follows: 435 bands were amplified,among which 325 bands were polymorphic,and percentage of polymorphic bands were 74.7 %,an average of approximately 3.95 bands per primer.Genetic similarity indexes of ten germplasms of C.hystrix ranged from 0.473 7 to 0.8081,and the average value was 0.728 7.Static software was used to calculate the Nei's genetic distance and a dendrogram was constructed based on UPGMA cluster analysis.These ten germplasms were divided into 4 major groups,and their genetic relative and the genetic distance were discussed.广西林业科学研究院国家林业局中南速生材繁育重点实验室开放基金课题;兰州理工大学博士基金(SB08200602);甘肃省教育厅研究生导师基金(0703-11

    Study on transformation system of alfalfa and constructed AtPCS1 plant expression vector

    通过对6种不同基因型苜蓿愈伤组织的形成能力、愈伤组织的分化能力的比较发现,不同苜蓿品种间的分化率存在显著差异(P<0.05),其中甘农1号分化率最高(42.9%);对甘农1号叶片、下胚轴及子叶等外植体的分化再生能力研究发现,叶片起源的愈伤组织分化时间短,且丛生芽形成数量高于下胚轴和子叶;再生芽可在B5培养基上成苗,并在蔗糖浓度为10 g/L的1/2 MS培养基上顺利生根.据此认为,苜蓿甘农1号叶片是适于做遗传转化的理想材料.利用RT-PCR方法扩增拟南芥螯合肽合成酶(AtPCS1)基因全序列构建了AtPCS1的植物表达载体pBI121-AtPCS1,为下一步AtPCS1基因转化苜蓿奠定了基础.Six cultivars of alfalfa with different genotypes were compared with each other in their callus induction capacity and plant regeneration ability.The significant differences were observed(P<0.05) in regeneration ability of these genotypes,and the cultivar Medicago sativa'Gannong No.1' has that the highest regeneration ability.Then the callus induction and plant regeneration ability of different explants as leaf,hypocotyls and cotyledon of Gannong No.1 were studied,and found that the calluses coming from leaf differentiated much earlier and had more regeneration buds than others.The in vitro regenerated plants were successfully rooted in 1/2 MS medium and 10 g/L sugar.The results above suggested that the leaves of the cultivar 'Gannong No.1' would be fit for the ideal explants in transgenic researches.Meantime,the full length of AtPCS1 gene was amplified by RT-PCR from Arabidopsis thaliana(ecotype Columbia).Furthermore,the plant expression vector pBI121-AtPCS1 was constructed,and that were as basic for transgenic alfalfa research in future.福建省自然科学基金(D041004);; 甘肃省自然科学基金(3ZS042-B25-011);; 兰州理工大学博士基金(SB08200602)

    Correlation between Outdoor-exposure and Indoor-accelerated Ageing Test for Epoxy Coating

    通过室外海洋大气暴晒和室内紫外线照射对环氧涂层进行了老化试验,并运用光泽、颜色、粉化率、SEM以及电化学多重动电位扫描(MCPDP)等手段测试了涂层在室外大气暴晒和室内紫外加速老化下的表面形貌和对金属保护性能的变化,初步探讨室外海洋大气暴晒和室内紫外人工老化试验的相关性。发现对于环氧涂层室内紫外人工老化降解的效果强于室外海洋大气天然老化,基于光泽测试的加速因子约为5;基于颜色和粉化率测试的加速因子都大于12。The outdoor-exposure test and indoor-accelerated ageing test were carried out,and a series of performance evaluations such as gloss,color,the degree of chalking,SEM and MCPDP were applied to test the transformation of the surface topography and protection properties of epoxy anticorrosion paint.The correlation between indoor-accelerated ageing test and outdoor-exposure test was discussed preliminary.It indicates that the results of degradation in the indoor-accelerated ageing test are greater than those in the outdoor-exposure test,the accelerating factor according gloss is about 5 and the accelerating factors according color and the degree of chalking are above 12

    A New Mathematics Model of the Temperature Programmed Analysis Technology

    以表面作用包括表面吸/脱附以及表面反应本征速率方程为基础,提出新的程序升温分析技术(TPAT)的数学模型。与经典的理论模型相比,这种新的TPAT理论模型更接近实际的程序升温分析实验过程。设计和进行特定催化剂的TPd、TPr及TPO实验,得到相应的程序升温谱图,采用新的TPAT理论模型模拟上述谱图,计算出相应的表面作用活化能等重要热力学参数。结果表明,新的理论模型具有良好的模拟性能,平均相对误差(Ard)小于1%。Based on the surface effect including surface adsorption/desorption and intrinsic kinetics rate equation,a new mathematical model of the temperature programmed analysis technology was proposed.This model is different from the classical TPAT theory models,which indicates much more coincidence with the actual reactions than other models.TPAT(TPD,TPR and TPO) experiments were designed and carried out to receive their profiles.Based on these profiles of TPAT and the simulated experiments,the novel theory model was designed and the thermodynamics parameters(such as activation energy,etc.) were deduced and calculated by this model.The results show that this model has excellent simulation with the actual experiments,and the average relative errors are easily controlled less than 1%

    与南海西边界流有关的区域海洋学进展

    从动力学角度,回顾了与南海西部主流系及其涡旋研究有关的南海物理海洋学的进展.主要内容涉及南海西边界流漂流浮标观测、西边界流源区涡旋遥感观测、西边界流源区跨陆架交换、涡流相互作用、西边界流年际变化、西边界流区海气相互作用、南海贯穿流等方面的进展.西边界流是一个强流系,其与季节和年际变化相关的结构特征、变率及跟某些动力过程的关联有待研究.在西边界流变率、涡旋环流相互作用、海气过程以及南海贯穿流取得了以下成果:(1)利用漂流浮标观测样本对南海西边界环流进行分析,指出南海西边界表层环流在到达越南中部沿岸后伴随着流幅变窄的同时流速加强;探讨了南海北部环流变化机理,定量诊断南海西边界流北支冬季逆风流产生的动力机制;利用航次数据对18°n断面的经向地转流进行诊断,表明南海西边界流的经向输送年际变化明显;(2)结合航次观测数据,对2003/2004年冬季南海北部2个反气旋涡旋和2007年夏季18°n附近的3个反气旋涡旋进行研究,指出冬季2个涡旋产生后以罗斯贝(rOSSby)波速度(约0.1M/S)沿北部陆坡向西南方向传播,并初步揭示了南海西边界环流与夏季3个涡旋的相互作用;南海北部陆架区涡旋西南向传播最大(最大为0.09M/S),而越南以东海域涡动能(EkE)最大,这都说明涡旋活动与南海西边界流存在强的相互作用;(3)发现南海西边界流附近海表面温度(SST)强的季节内振荡特征,进一步研究表明此区域冬季SST季节内变化会使潜热季节内信号减弱20%;(4)探讨了南海贯穿流的长期变化特征以及与整个太平洋环流系统的相互关联.国家自然科学基金重点项目(40830851); 国家重点基础研究发展计划(2011CB403504); 中国科学院近海海洋观测研究网络——西沙南沙海洋观测研究站建设项目(KZCX2-EW-Y040)资
