    吡非尼酮药物缓释载体PTMAc-PEG-PTMAc水凝胶制备与体外实验

    【目的】以PTMAc-PEG-PTMAc三嵌段(PPP)水凝胶为吡非尼酮(PFD)药物缓释载体,评价其药物缓释性能。观察PFD载药水凝胶对人眼Tenon′s囊成纤维细胞(HTF)增殖抑制作用。【方法】制备PPP水凝胶;研究其溶胀率及其影响因素;光学显微镜观察水凝胶(载药与空载)中人眼Tenon′s囊成纤维细胞(HTF)的形态学改变;直接接触法进行PPP载药水凝胶进行体外释药实验。通过测定浓度、计算累积药物释放量,绘制出两种水凝胶体外药物释放曲线。CCK-8法检测不同浓度不同时间点水凝胶材料对细胞活力的影响及载药水凝胶体外抑制HTF增殖。【结果】PPP水凝胶成胶稳定。扫描电镜显示水凝胶呈现三维立体网孔状结构。水凝胶具有溶胀特性。溶胀率随水凝胶材料浓度增加而减少。在体外释药试验显示在前4d有明显的药物突释,此后缓慢释药直至第14天达到平衡。增加载药浓度,减少凝胶体积将明显增加药物累积缓释量;显微镜观察PPP水凝胶有抑制HTF细胞黏附作用;PPP水凝胶与HTF相互作用24、48、72、96h观察细胞活力分别为85.7%、93.0%、82.0%、81.6%,水凝胶不同方位HTF细胞形态不尽相同;水凝胶对细胞有抑制细胞黏附作用。两组不同载药浓度(1mg/mL和2mg/mL)载药水凝胶在24、48、72、96h这4个时间点细胞增殖抑制率分别为25.8%、21.8%、55.4%、25.6%;44.6%、35.9%、55.5%、31.4%。而1mg/mLPFD溶液在24、48、72、96h这4个时间点细胞增殖抑制率分别为28.9%、29.7%,7.8%、7.7%。【结论】PPP水凝胶具备良好药物缓释性能,载吡非尼酮水凝胶能明显抑制HTF增殖且较含等量药物溶液长效

    利用透射电镜研究了九龙江口现代潮间带表层沉积物、九龙江地区中全新世剖面和东屿中-晚全新世剖面的微型硅藻(细胞大小为2~20μm)的组成和丰度.共计硅藻12属26种(包括变种).对每个种的生态分布作了描述,并提供了相应的壳面电镜照片.Thalassionemanitzschioides是九龙江口现代潮间带表层沉积物中的优势种;九龙江中全新世地层的优势种为Naviculaclimacospheniae,Thalassionemanitzschioides;东屿中-晚全新世地层中,Cyclotellastriata,Thalassionemanitzschioides是优势种.微型硅藻在各个样品中所占比例都相对较高,最高可达86.7%,表明微型硅藻在沉积硅藻中占有重要的地位. 【英文摘要】 The species composition and cell density of nanodiatoms(cell size smaller than 20 μm) were investigated with TEM for the sedimental samples from different sedimental sections at modern intertidal zones and Middle Holocene in the Jiulong River, and in Dongyu Island in late Middle Holocene and Late Holocene.     少根紫萍(Landoltia punctata)是一种浮水生、多功能、高环境适应能力的能源植物,为了解其生理特性的分子机制,利用少根紫萍转录因子预测数据与拟南芥、水稻、玉米数据库数据进行宏观比较,并结合转录组测序技术对少根紫萍营养胁迫后转录因子表达分析.结果显示,少根紫萍有1 076个转录因子,分属于66个家族,其中在bZIP、WRKY、AP2/ERFERF、MYB、NAC、MADS-box等家族基因数明显减少,一定程度上解释了浮萍高黄铜低木质素、难开花的生理特性;少根紫萍在营养胁迫下偏向上调AP2/ERF-ERF、MYB、bHLH家族特定基因和下调AP2/ERF-ERF、WRKY家族特定基因来响应营养胁迫,尤其是AP2/ERF-ERF、WRKY两个家族中特定基因表达下调在响应水体营养胁迫中有非常重要的作用.本研究在转录因子层面对浮萍的生理特性,特别针对营养胁迫下转录因子的表达进行了探究,可为建立浮萍水生模式植物系统、深入探讨少根紫萍响应营养胁迫机制以及改造成耐受胁迫的高效淀粉积累能源植物提供理论指导
