    Study on Functional Properties of Pumpkin Powder

    本实验用中性蛋白酶酶解制得的南瓜粉,通过测定该南瓜粉对ACE和ALP活力的影响,研究了南瓜粉对血管紧张素转化酶(angiotension converting enzyme,ACE)和碱性磷酸酶(alkaline phosphatase,ALP)活性的影响,考察其降血压作用及潜在的抗肿瘤作用;同时提取并初步纯化南瓜多糖,通过fenton反应产生羟基自由基(·OH),测定南瓜粉清除·OH能力,以及通过FRAP法测定其总还原能力,利用纸层析法分析南瓜多糖的单Z糖组成。结果表明,该南瓜粉对ACE和ALP均有抑制作用,具有一定的降血压作用和潜在的抗肿瘤作用;南瓜多糖是由半乳糖、葡萄糖、阿拉伯糖和鼠李糖四种单糖组成,具有良好的抗氧化活性。Pumpkin powder was produced by using neutral proteinase to hydrolyze pumpkin pulp and then freeze-drying,and pumpkin polysaccharides were extracted. In order to understand the hypotensive effect and potential antitumor effect of the pumpkin powder,its effects on the activities of ACE (angiotensin converting enzyme) and ALP (alkaline phosphatase) were investigated. The scavenging effect of the pumpkin powder was determined on hydroxl radical (·H),which is produced by Feton reaction,and its total reducing power was determined by FRAP method. The monosaccharide composition of the pumpkin polysaccharides was analyzed by paper chromatography method. The results showed that the pumpkin powder can inhibit the activities of ACE and ALP and has hypotensive effect and potential antitumor effect.     The effects of LaCl 3 on membrane potential and transmembrane proton gradient for rice ( Oryza sativa ) seedling roots were studied. Highly purified plasma membranes were isolated by aqueous two phase partitioning method. Both the gradient of transmembrane proton and membrane potential were stimulated by certain low concentration of LaCl 3 and depressed by high concentration of LaCl 3.     Purification and characterization of hydrogenase from Synechococcus sp. PCC 7942

    报道了室温、空气环境下聚球藻Synechococcus sp.PCC 7942氢酶的分离纯化。经过超声破碎、超速离心、离子交换层析、疏水层析及凝胶层析等步骤,氢酶被纯化了218倍,得率为6.5%,比活为1.46 U·mg-1蛋白。纯化氢酶的SDS-PAGE图显示五条蛋白带,分子量约为83kDa,60kDa,47kDa,30kDa和27kDa。该氢酶为可溶性的双向氢酶,其催化放氢的最佳电子供体为还原态的甲基紫精,最适温度50℃,最适pH 8.0。Hydrogenase from Synechococcus sp.PCC 7942 was purified to close homogeneity aerobically at room temperature.The hydrogenase-containing crude extract was collected after ultrasonic disruption and removal of cell debris by ultracentrifugation.Subsequently,three steps of column chromatographies(anion exchange,hydrophobic interaction and gel filtration)were performed.Hydrogenase was purified about 218-fold with a yield of 6.5% finally.The purified enzyme has a specific activity for hydrogen evolution of 1.46 U.mg-1 protein.SDS-PAGE gel of the purified enzyme revealed five predominant protein bands with estimated molecular weights of 83,60,47,30 and 27 kDa,respectively.The enzyme is a soluble bidirectional hydrogenase and shows maximum activity while using reduced methyl viologen as an electron donor.The optimum temperature and pH value for hydrogen evolution catalyzed by the purified hydrogenase are 50 ℃ and pH 8.0.福建省自然科学基金(C0410002)资

    分析了真空冷冻干燥机水汽凝结器存在容易造成"冰塞"的原因,并提出了安装气流扩散器的改进方法,对改进前后的凝结器捕水性能进行了比较。由于气流扩散器的安装,捕水周期得以延长,捕水能力及工作效率显著提高,实用性较好,而且扩散器的制作费用低,性价比高,有推广价值。This paper analyzes the reason for ice-block formation in water vapor condenser used in vacuum freezing dryer system,proposes a good method by the installation of an air current diffuser and compares the water capturing ability of the modified condenser with the original one.Due to the installation of the air current diffuser,the water capturing cycle is prolonged and the water capturing ability significantly improved.The new air current condenser has the advantage of improved efficiency,low cost,good performance versus price ratio and therefore is of value for practical application

    运用细胞生物学、生物化学等手段 ,观察鉴定分离提取的牡蛎天然活性肽BPO 1对人胃腺癌细胞的生物学效应 .实验结果表明 ,BPO 1能有效抑制人胃腺癌BGC 82 3细胞增殖活动 ,细胞生长受到抑制 ,分裂指数下降 .细胞周期检测出现凋亡峰 .光镜观察显示经BPO 1处理后的BGC 82 3细胞失去原有的恶性表型 ,表现出明显不同于对照组细胞 .与癌细胞诱导凋亡物相似的抗肿瘤效果BPO-1, eluted from oyster cells (Saccostrea Cucullata), treat the human Gastric Adenocarcinoma BGC-823 cells in vitro for investigating the biological effects by the methods of cell biology, biochemistry and molecular biology. The results revealed that the proliferation of BGC-823 cells were inhibited apparently after being treated with BPO-1, the growth of cells were inhibited, the cell mitotic index was declined obviously. Flow cytometry analysis showed BPO-1 could induce the emergence of the peak of apoptosis. Light microscopy showed that BGC-823 cells, which induced with BPO-1, had lost their original malignant phenotype which were different from those of untreated cells but were similar to those of apoptotic cells, and the abilities of AKPase were declined. The results showed that BPO-1 could inhibit the proliferation of the human gastric adenocarcinoma BGC-823 cells and act as a apoptosis inducer in its antitumor effects.福建省自然科学基金 (F0 10 0 0 10 )资助项

    Effects of Oyster Low Molecular Weight Bioactive Substance on the Human Lung Adenocarcinoma A549 Cells

    以新鲜牡蛎为材料 ,经特酸抽提和分子筛凝胶层析 ,获得牡蛎低分子活性物质 ,SDS PAGE凝胶电泳分析鉴定为该组分主要由 8种蛋白组成 .应用该物质处理人肺腺癌A5 49细胞 .实验结果显示 ,经 0 .1mg/mL牡蛎低分子活性物质处理的人肺腺癌A5 49细胞 ,细胞生长抑制率达 6 2 .2 % ,细胞分裂指数下降 2 8.95 % .光镜观察显示经牡蛎低分子活性物质处理的细胞形态发生明显改变 .结果表明 ,牡蛎低分子活性物质能有效的抑制肺癌细胞的增殖In this study, we isolated and purified low molecular weigh bioactive substance from oyster (Saccostrea Cucullata) acid extracts. SDS-PAGE electrophoresis identified they were mainly composed of eight kinds of proteins. Using 0.1 mg/mL to treat the human lung adenocarinoma A549 cells, the result showed that the rate of cell growth inhibitory was 62% and the mitotic index decreased to 27‰. Moreover, light microscopy assays revealed that the cell morphography obviously changed in the cells treated oyster low molecular weight bioactive substance.     应用超离心和柱层析结合聚丙烯酰胺凝胶电泳(PAgE)及电镜技术,研究中国鲎血蓝蛋白的结构特点.结果表明,经SEPHAdEXg-100纯化的血蓝蛋白在PAgE电泳中出现4条带.纯化的血蓝蛋白再经dEAE-32纤维素柱层析得到5个洗脱峰,每个峰在PAgE电泳下可分辨出4条带.在电镜下,血蓝蛋白分子出现环形、五角形、十字形和蝴蝶结形等构型并与其他解离的中间的构型同时存在.By using ultracentriFugation,chromatography,polyacrylamide gel electron phoresis (PAGE) and electron microscopy,the structural characteristics of hemocyanin of T.tridentatus have been investigated.The results show that there are Four bands in PAGE For hemocyanin puriFied by Sephadex G 100 chromatography.PuriFied hemocyanin can be separated by means of DEAE 32 cellulose chromatography into Five peaks, marked as I~V.Four bands can be distinguished by PAGE For each peak.Under electron microscope,Five typical view of T.tridentatus hemocyanin molecule can be observed according to diFFerent project angle.They look like ring,pentagon,cross,and bowtie in shape respectively.There are some of intermediate dissociating conFiguration of hemocyanin that can be seen in the same Field.国家和福建省自然科学基

    文昌鱼酸性磷酸酶(ACPASE,E.C.3.1.3.2)是一种含有铁离子的金属酶,酶的紫外-可见光谱(200~600nM)扫描,280nM,500nM有最大吸收峰,320nM有肩峰。荧光扫描:在281nM激发下,330nM有发射峰,在480nM激发下,515nM有一发射峰。可见光谱500nM;的吸收峰以及荧光515nM的发射峰为酶分子含铁的特征峰,与酶活力关系密切。不同浓度的乙醇对酶活力有明显的抑制作用,表现为反竞争抑制类型,其抑制常数为16%,测定酶随时间的变化的失活程度。可见光谱500nM吸收值随酶体系中乙醇含量增加而增加,当乙醇浓度达40%时,吸收值明显下降。在乙醇作用下,监测荧光330nM及515nM处的强度随时间变化的过程作为酶构象变化的指标,酶的330nM荧光发射峰值随乙醇浓度的增强而下降,515nM的荧光发射峰值随着乙醇浓度的增强而下降,峰位不变。在不同浓度乙醇作用下,酶的构象变化快于活力变化。The previous studies by our research team have shown that Branchiostoma belcheri Acid Phosphatase (ACPase, E.C.3.1.3.2) was a kind of metalloenzyme which exhibits an absorption peak with the warelength at 500nm in its visible spectra and an emission peak with the warelength at 515nm in its Fluorescence spectra because its containing iron.This two charactal peaks are sensitive to the enzyme activity (reFerance 7--9 For detail).Changes of conFormation and activity at the active site of the ACPase in diFFerent eoncentration(V/V) of ethanol solutions has been investigated by means of Fiuorescence emission and UV-visible spectra.Kinetic analysis of the inhibition shows that the inhibition of ethanol on the enzyme is Found to be of uncompetitive type and the inhibition constants is Found to be 16%.The rate constants of denaturation are Faster than those of inactivation in the presence of ethanol

    应用从中国鲎血细胞中提取的鲎素处理人胃癌 BGC- 82 3细胞 ,研究海洋生物活性物质的抗肿瘤作用 .实验结果显示 ,经 2 .0 μg/m L鲎素处理的 BGC- 82 3细胞生长缓慢 ,倍增时间延长 ,细胞集落形成能力减弱 ,细胞生长抑制率达 62 .67% ,细胞分裂指数下降 4 0 .91% .处理后细胞碱性磷酸酶细胞化学反应减弱、分布改变 ,碱性磷酸酶和乳酸脱氢酶活力分别下降 4 0 .59%和 76.0 5% .结果表明 ,鲎素能有效地抑制胃癌细胞的增殖活动 ,具有与癌细胞诱导分化物相似的抗肿瘤效果 .Tachyplesin, which was isolated from acid extracts of horseshoe crab ( Tachypleus tridentatus ) hemocytes, was used to treat the human gastric carcinoma cell line BGC 823 and to investigate the antitumor effects of marine biological active substances. It was found that the BGC 823 Cells, after treated with 2.0 μg/mL tachyplesin, grew slowly, the doubling time enlonged, the colony formation ability decreased, the growth inhibitory rate amounted to 62.67%, and cellular mitotic index decreased by 40.91%. In the meantime, tachyplesin could change the distribution of AKPase and decline the abilities of AKPase by 40.59% and LDH 76.05% respectively. These results indicated that tachyplesin could effectively inhibite the proliferation of the human gastric adenocarcinoma cell line BGC 823 and act as a differentiational inducer in its antitumor effects.福建省自然科学基金!     研究两种红树植物秋茄和桐花树幼苗根和叶在不同盐度培养下 ,蛋白质、丙二醛及过氧化氢含量的变化情况 ,结果表明 :盐度对秋茄和桐花树幼根和幼叶蛋白质含量都有影响 ,总体上是低盐度稍降低其含量 ,高盐度稍增加其含量 .秋茄幼根和幼叶在 0~ 5 0盐度范围内MDA含量随盐度的升高先降后升 ,但是根和叶上升的折点不同 (根30 ,叶 2 0 ) ,而桐花树幼根MDA含量在盐度 30范围内比较平稳 ,盐度高于 30时则迅速上升 ,而幼叶在盐度 2 0以内下降 ,高于 2 0则上升 .秋茄和桐花树的幼根在低盐情况下H2 O2 含量随盐度的增加而下降 ,而幼叶都表现出先降后升再降的趋势 ,只是上升的折点不同而已 (秋茄 40 ,桐花树 2 0 ) .这些结果表明 ,盐度对秋茄和桐花树幼根和幼叶的H2 O2 含量及膜脂过氧化作用是有一定差异的 . 【英文摘要】 The variations of protein contents, malondialdehyde(MDA) and H 2O 2 density in roots and leaves of K.candel and A. coniculatum seedlings with the changes of salinity were studied. The results showed that salinity had some effects on the protein contents in roots and leaves in both plants, in general, low salinity decreased its contents and high salinity increased its contents.
