    在我们周围的世界中,手性是普遍存在的现象。手性化合物(单一对映体)在药物、天然产物、农业化学和液晶材料等许多领域具有广泛的应用。除旋光性外,对映异构体具有基本相同的物理和化学性质。这一特殊性增加了对映体分离和分析的困难和复杂性,也使得手性分离分析成为当今分离科学和分析化学中的一个研究热点。手性高效液相色谱法是目前公认的一种可靠、准确的对映体分析方法,其中以手性固定相法最为简便。但是现有的手性固定相缺乏通用性,大多手性色谱分析基于实际经验探索。种类繁多且价格昂贵的手性色谱柱限制了手性色谱技术的应用。如何从色谱学、谱学和计算化学等方面深入了解色谱拆分过程的手性识别机理,从理论上指导实际的手性分离分...Chirality widely exists in our world.     人红细胞凝集型单抗的制备与鉴定

    目的制备人红细胞凝集型单克隆抗体并进行鉴定。方法以3种方式处理人血红细胞,获得红细胞免疫原,分别免疫Balb/c小鼠。应用杂交瘤技术,取免疫小鼠的脾细胞与Sp2/0骨髓瘤细胞融合,以凝集实验筛选分泌凝集型抗体的杂交瘤细胞,继而用有限稀释法克隆3次,并对杂交瘤细胞的稳定性进行测定。通过腹水法制备抗体,亲和层析法纯化抗体,对抗体的特性进行研究。结果 3种方式获得的免疫原均能建立有效的免疫反应,低渗加超声破碎法所获得的小鼠血清抗体效价最高。通过融合获得8株可稳定分泌抗体的杂交瘤细胞,其中,4A1分泌的抗体凝集效价最高。抗体可使4种人血型的红细胞凝集,无种属交叉凝集反应。结论有效制备了针对人红细胞的凝集性单抗,为其之后的应用研究奠定了基础。福建省省属公益类项目(2015R1036-3,2016R1034-2,2017R1036-3);;福建省科技计划(2014Y0082,2014Y0083

    采用Chiralcel OD H和Chiralpak AD H手性柱,研究5种新近合成的外消旋双芳基硒代甘油硫醚和 4 种芳基硒代甘油烷氧醚对映体的高效液相色谱拆分方法.对比这两种手性固定相、异丙醇和乙醇流动相调节剂、以及样品结构对硒代甘油醚色谱拆分的影响.除样品(1c)外,其他8个外消旋硒代甘油醚样品均可达到基线拆分.The enantiomer separation of nine pairs of racemic 1-arylthio-3-arylseleno -2-propanols and 1-alkoxy-3-arylseleno-2-propanols was achieved with high performance liquid chromatography by using cellulose tris(3,5-dimethylphenyl carbamate) (Chiralcel OD-H) and amylose tris(3,5-dimethylphenylcarbamate) (Chiralpak AD-H) as chiral stationary phases and hexane-2-propanol or hexane-ethanol as mobile phases in various percentages.Amylose tris(3,5-dimethylphenylcarbamate) exhibited higher chiral recognition ability compared to cellulose tris(3,5-dimethylphenylcarbamate).The effects of alcohol in the mobile phase and solute structures on the retention and resolution were investigated.On the Chiralcel OD-H column,solutes elute faster with ethanol than with 2-propanol modifier as expected from the higher polarity of ethanol. However,on Chiralpak AD-H column the retention factors of all the enatiomers increase,the enantioseparation factors and resolutions dramatically change on changing 2-propanol to ethanol.The explanation may be that ethanol changes the higher order structure of amylose.The enantioseparation on the Chiralpak AD-H column is much more influenced by the kind of alcohol in the mobile phase than the Chiralcel OD-H.Under optimized conditions,baseline enantioseparation of eight pairs of racemic 1-substituted 3-arylseleno-2-propanols were obtained

    显微CT技术是一种非破坏性的三维成像技术,在医学、地质学、材料学等方面有着广泛的应用,在植物成像方面也是一种前景很好的技术手段。但植物组织主要由轻元素构成,X射线成像对比度很低,其应用受到很大限制。本文在前人的研究基础上,使用不同浓度的磷钨酸处理不同植物样品,比较不同处理时间、不同植物结构的差异,经CO_2临界点干燥后再扫描等前处理方法对显微CT成像效果的影响,探索出增强植物材料成像对比度的处理流程,使其更适用于显微CT扫描

    开展了低浓度氢氟酸体系中MIBK萃取分离钽铌的研究,考察了萃取时间、萃取相比、氢氟酸浓度、钽铌浓度、钽与铌质量比对萃取分离过程的影响。结果表明,在始氢氟酸浓度0.5mol/L、钽铌总浓度60g/L、萃取相比(O/A)为5、钽与铌质量比2的条件下萃取5min可获得最佳的钽铌分离效果,钽铌分离系数高达50 000
