Relationship between Vitamin and Fetal Development
胎儿在母体宫内发育经过受精卵、胚胎、胎儿三个阶段,任何影响母胎健康的病因都影响宫内受精卵、胚胎和胎儿的发育,严重者甚至导致胚胎停育、胎死宫内。缺乏维生素容易诱发人体各种疾病,孕期母体维生素的需要量增加,如果孕期母体维生素摄入不足、补充不够、未得到及时增加或增加过量,都会影响胎儿器官的正常发育,导致胎儿发育不良。有些严重的胎儿畸形是致死性的,因此孕期要注意筛查及时处理。孕期加强母胎保健,重视优生优育,改善孕妇和儿童营养状况,合理调配膳食,适当补充维生素,了解各种维生素的功效和胎儿发育的相关性,有助于降低新生儿出生缺陷、防治妇女儿童营养缺乏病。aFertilized egg, embryo and fetus are three stages of fetus growth in maternal intrauterine. Any factors caused by the mother or the fetus can affect intrauterine growth of fertilized egg, embryo and fetus. Some serious factors could lead to embryonic growth cease, even fetal death. Lack of vitamin can induce human related diseases, and vitamin requirements are increased in pregnant women. If insufficient of vitamin intake in those pregnant women, fetus could suffer from dysplasia. Some severe fetal abnormalities can cause fetal death. So, it is necessary that pregnant women should have fetal developmental screening, and that those pregnant women with vitamin insufficient should be treated in time. The reasonable supplement of vitamins, and deployment of diet, is benefit for them to reduce neonatal defects, and to prevent the nutrition-related diseases in pregnant women
暹罗鳄多肽粉提高机体免疫力及抗疲劳的效应
为研究暹罗鳄多肽粉对小鼠免疫力及抗疲劳效应的影响,对小鼠的免疫能力和相关血清指标进行了测定,同时也对小鼠的运动能力及疲劳代谢指标进行了测定.实验结果显示:暹罗鳄多肽粉可以促进小鼠胸腺和脾脏的生长,免疫力得到提高.相对于对照组小鼠,暹罗鳄多肽粉组小鼠血清中的总蛋白、血钙、白蛋白含量升高(p<0.05),谷丙转氨酶(ALT)的活力和血糖浓度下降(p<0.05).小鼠爬绳及负重游泳实验发现服用了暹罗鳄多肽粉的小鼠垂直爬绳时间和负重游泳时间均高于对照组的(p<0.01),小鼠的耐力增强.进一步通过建立抗疲劳小鼠力竭游泳模型研究了暹罗鳄多肽粉的抗疲劳动物学功效,结果显示相对于对照组,暹罗鳄多肽粉组小鼠血液中尿素氮(BUN)和乳酸(LA)的含量以及乳酸脱氢酶(LDH)的酶活力均降低(p<0.05),而超氧化物歧化酶(SOD)的酶活力升高(p<0.05).综上结果表明暹罗鳄多肽粉可能有保肝护肝,提高肌体免疫力以及抗疲劳等功效.福建省自然科学基金项目(2016J05105);;广东省自然科学基金博士启动项目(2016A030310374);;厦门大学生命科学学院-福建鼍龙鳄鱼生物制品研发协同创新中心项目(XDHT2017004C
Relevant relationship between IUD and endometrial polypus
目的:研究宫内节育器(InTrAuTEruS dEVICE,Iud)与子宫内膜息肉的相关性。方法:统计2004年5月--2007年5月在厦门大学附属中山医院行宫腔镜取环并经病理检查确诊Iud合并子宫内膜息肉病例,分析Iud与子宫内膜息肉的相关敏感因素。结果:1357例宫腔镜取环者Iud合并子宫内膜息肉285例,占21%,患者对金属皮带扣或金饰物过敏,经阴道分娩和人工流产且中位子宫患者的发病率较高,息肉好发于Iud的周围。放环时间15年以上,Iud变形、下移和长锈斑的比例均较高。结论:放置Iud且有息肉形成高危因素的患者应及时行宫腔镜检查,并行诊刮术或子宫内膜息肉电切术、取环术。Objective:To study relevant relationship between IUD and endometrial polyps(EMP).Methods:The cases who removed IUD and EMP by hysteroscopic operation and diagnosed by pathological examination were analyzed in Xiamen Zhongshan Hoapital from may 2004 to may 2007,and the related factors of IUD and EMP were analyzed.Results:of 1357 cases who removed IUD by hysteroscopic operation,the cases who had EMP was 21%(285/1357).EMP growed around IUD in cases whose uteri were middle position.Vaginal labor and artificial abortion and allergic to metal including the metal buckle and gold ornaments were high risk factors.Conclusion:The cases with IUD who had risk factors of endometrial polypus formation need do hysteroscopic operation and remove IUD
脱氧胆酸改性普鲁兰多糖纳米粒子制备与表征
通过酯化反应将脱氧胆酸偶联于普鲁兰多糖骨架形成具有两亲性的普鲁兰多糖衍生物(DP),采用纳米沉淀法制备纳米粒子(DPNs),考察制备条件对纳米粒子性质影响,为进一步将其作为药物载体的研究提供基础。衍生物DP结构通过FT-IR和1 H NMR表征,DPNs经透射电镜、动态光散射仪和zeta电位仪表征检测。获得不同取代度脱氧胆酸改性普鲁兰多糖衍生物,制备得到的纳米粒子呈球形,表面光滑规整,平均粒径100~300nm,zeta电位在-20mV左右。脱氧胆酸改性普鲁兰多糖衍生物通过纳米沉淀法能制备出纳米粒子,颗粒性质受制备条件影响
系统性红斑狼疮患者高表达的CD154促进自身抗体分泌
目的】探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)CD154表达水平、及CD40/CD154对SLE患者分泌自身抗体的影响和地塞米松(Dex)对此的作用。【方法】选择SLE活动期患者(10例)和正常对照者(11例),分离PBMC,检测其CD154的表达水平;培养PBMC,应用CD40 mAb和Dex进行干预,使用ELISA法检测培养液上清抗dsDNA水平。【结果】①活动期组PBMC CD154表达水平明显高于对照组6.4%±2.1%vs 1.5%±1.2%,(P〈0.05);②活动期组培养液上清抗dsDNA抗体水平明显高于对照组[(11.8±6.8)U/mL vs(5.6±2.3)U/mL(RPMI 1640),(11.4±7.0)U/mL vs(6.3±2.1)U/mL(IgG),P均〈0.05];③CD40 mAb使两组培养液上清抗dsDNA抗体水平明显增高[活动期组:(18.6±7.7)U/mL vs(11.8±6.9)U/mL,对照组:(8.3±4.4)U/mL sv(5.6±2.3)U/mL,P均〈0.05]。④在活动期组,Dex明显抑制CD40 mAb引起的PBMC培养液上清抗dsDNA抗体水平的增高[(8.8±5.2)U/mL vs(18.6±7.7)U/mL,P〈0.05],使其低于非特异性处理时水平[(8.8±5.2)U/mL vs(11.8±6.8)U/mL,P〈0.05];Dex不影响对照组。【结论】活动期SLE患者PBMC表达的CD154水平升高,此能够促进抗dsDNA抗体的分泌,而Dex能够抑制其作用
Hippo Signaling Suppresses Cell Ploidy and Tumorigenesis through Skp2
大多数真核生物的体细胞是二倍体,即仅含有两组染色体,分别遗传自父本和母本。而一些特定组织如心脏、肝脏等就含有多倍体细胞,特别是肝脏组织含有较高比例的四、八倍体等多倍体细胞。肝脏是人体的重要解毒器官,同时酒精、肝炎病毒等毒性物质或毒性代谢物容易诱发肝细胞的基因突变,多倍体被认为有利于提供代偿性的正常基因来维持肝脏稳态。然而肝脏受损后,多倍体细胞将会受胁迫进行增殖,再生修复受损的肝组织。因此研究机体调控多倍体细胞产生及多倍体细胞进行细胞分裂的调控机理对于理解肝癌的发病机理和肝癌的治疗至关重要。Hippo信号通路在调节组织成体干细胞的分化和增殖,调控器官再生与尺寸大小中具有重要作用。深入研究发现, Hippo信号通路下游效应分子YAP通过AKT-SKP2信号促进二倍体细胞向多倍体转化及多倍体细胞的生长增殖。本项研究阐明了Hippo缺失及YAP激活促进多倍体细胞产生及增殖作为肝癌发生发展中的一个重要机制,为肝癌诊疗提供了新的策略。
周大旺,博士,厦门大学生命科学学院教授、副院长、国家杰出青年基金获得者。【Abstract】Polyploidy can lead to aneuploidy and tumorigenesis. Here, we report that the Hippo pathway effector Yap promotes the diploid-polyploid conversion and polyploid cell growth through the Akt-Skp2 axis. Yap strongly induces the acetyltransferase p300-mediated acetylation of the E3 ligase Skp2 via Akt signaling. Acetylated Skp2 is exclusively localized to the cytosol, which causes hyper-accumulation of the cyclin-dependent kinase inhibitor p27, leading to mitotic arrest and subsequently cell polyploidy. In addition, the pro-apoptotic factors FoxO1/3 are overly degraded by acetylated Skp2, resulting in polyploid cell division, genomic instability, and oncogenesis. importantly, the depletion or inactivation of Akt or Skp2 abrogated Hippo signal deficiency-induced liver tumorigenesis, indicating their epistatic interaction. Antitumor effects of a novel chloroquinoline derivative as EGFR inhibitor on human esophageal carcinoma
目的:评价Egfr抑制剂4-取代-7氯喹啉衍生物(TW9183)的体内外抗肿瘤生物活性。方法:采用分子对接技术探讨其与Egfr之间的相互作用模式,蛋白免疫印迹法评价在蛋白水平的直接作用,体外细胞生物学及体内裸鼠移植瘤模型探讨TW9183的活性。结果:Egfr受体的氨基酸残基MET769,PrO770,lyS721可与TW9183间形成氢键,蛋白印免疫迹法表明化合物抑制Egfr的磷酸化;体外细胞实验显示TW9183有明显抑制食管癌细胞增殖、周期阻滞于g2/M期及促进凋亡的效果;裸鼠食管癌移植瘤模型验证其在体内同样具有抑制肿瘤生长促进凋亡的作用。结论:TW9183在体内外实验中显示具有潜在的抗食管癌作用。Objective: To explore the underlying mechanism of 4-substituent-7-chloroquinoline derivative(TW9183),which has a selective anti-tumor effect in vitro and in vivo.Methods: Docking simulation was performed to position compounds into the EGFR structure active site to determine a possible binding model.We also used Western blot assay to directly evaluate the interaction.Cell cultures and tumor xenografts were utilized to test the effect of TW9183.Results: TW9183 was nice bound to the EGFR with the amino hydrogens of MET769, PRO770 and LYS721,forming optimal H-bonds interaction.Western blot assay showed that the compound inhibited EGFR phosphorylation.TW9183 treatment increased typical apoptosis and the number of cells in the G2/ M phase in ECA-109 cells.In ECA-109 xenograft model in nude mice,TW9183 inhibited tumor growth and promoted apoptosis.Conclusion: TW9183 has potential antitumor activity in vitro and in vivo.泉州市科技计划重点项目(2013Z35
宫腔镜在体外受精一胚胎移植失败病例中的应用价值
【目的】探讨官腔镜在体外受精-胚胎移植(IVF-ET)失败病例诊断中的应用价值。【方法】收集从1996到2004年在中山大学附属第一和第二医院生殖中心行体外受精-胚胎移植失败患者107例,在我院进行了宫腔镜检查,其中83例进行了内膜活检,总结分析该资料。【结果】官腔镜发现正常官腔74例(69.16%),异常33例(30.84%),其中包括宫腔粘连6例、内膜薄6例、宫腔息肉6例、内膜肥厚5例、黏膜下子宫肌瘤4例、宫颈管狭窄3例、内膜炎2例、子宫纵隔1例。83例子宫内膜病理活检则发现正常内膜54例(65.06%),其中增生期18例、分泌期36例;内膜异常29例(34.94%),其中黄体功能不全(LDP)12例、轻度增生过长7例(其中有一例同时伴有官腔息肉),内膜炎症5例、官腔息肉3例、子宫内膜结核2例。【结论】对体外受精-胚胎移植失败病例,进行宫腔镜检查有助于明确病因
Expression of Fusion Protein of Parathyroid Hormone and Transferrin N-terminal Half-molecule in Pichia pastoris
利用重叠PCR技术将PTH(parathyroidhormone,甲状旁腺激素)基因与TFN(transferrinN_terminalhalf_molecule,转铁蛋白N端半分子)基因在体外融合,融合基因克隆至真核表达载体pPIC9中,转化毕赤酵母GS115。转化子经甲醇诱导后,融合蛋白得到了表达并分泌到发酵上清液中。经SPSepharoseFF阳离子交换层析、PhenylSepharoseFastFlow疏水层析纯化获得了纯度大于95%的PTH_TFN样品。Westernblot分析及腺苷酸环化酶实验证明融合蛋白中的PTH具有与抗PTH抗体结合能力及刺激腺苷酸环化酶的活性,铁饱和实验证明融合蛋白中的TFN和单独的TFN具有相同铁结合能力。因而TFN可望作为PTH的天然运输载体。The fused gene (PTH_TFN) of parathyroid hormone (PTH) gene and transferring N_terminal half_molecule (TFN) gene was amplified by multiple PCR and inserted into pPIC9 vector. The recombinant plasmid pPIC9_PTH_TFN was transformed into Pichia pastoris GS115 by PEG. After methanol induction, the target protein was expressed in fermentation supernatant at high level.The fused protein PTH_TFN with purity being higher than 95% was finally obtained after purification through two_step chromatography : SP Sepharose Fast Flow and Phenyl Sepharose Fast Flow.Western blot analysis and adenylate cyclase assay proved that the fused protein exhibited the bioactivity to stimulate cAMP synthesis and the ability to bind Fe ~3+ in the Fe ~3+ saturation study as the recombinant TFN did indicating that TFN could be used as the transcellar carrier of PTH.国家高技术研究与发展项目基金资助(No.2004AA215172)。~
