    气候变化对中国海洋经济可持续发展的影响

    为评估气候变化及实施减排政策对中国海洋经济的影响,本文构建了中国海洋经济社会经济核算矩阵(marine social accounting matrix,MSAM),并以此为数据基础构建中国海洋经济可计算一般均衡(computable general equilibrium,CGE)模型,并将该模型与气候变化影响模型相结合,从而实现了从气候变化对自然系统的影响到对经济系统影响的转化。论文设计了实施和不实施温室气体减排两种政策时的可能的6种气候变化情景。各情景的模拟分析表明:海平面上升同时考虑天文潮最高潮位、同时考虑最大台风暴潮和最大天文潮耦合影响两种子情景下,中国实施减排政策对沿海GDP的影响要分别比不实施减排政策低0.19%和0.12%;海平面上升叠加天文潮、风暴潮3种子情景下,实施减排政策对海洋经济的影响要分别比不实施减排政策低1.67%、0.72%、0.37%

    Construction of Normalized cDNA Library from Hepatopancreas of Abalone Haliotis diversicolor supertexta Exposure to Tributyltin

    用rdP试剂提取三丁基锡暴露诱导的杂色鲍肝胰腺总rnA,经OlIgOTEX纯化得到MrnA;应用SMArT技术合成双链CdnA,双链特异核酸酶(dSn)进行双链CdnA的均一化,构建了杂色鲍三丁基锡暴露诱导下的均一化CdnA文库。原始文库的库容为4.3x106Cfu/Ml,重组率为97.9%。从文库中随机挑选了3288个克隆进行测序,得到3048个高质量EST序列,其中有370条COnTIgS,2103条SInglETS,unIgEnES共2473条,冗余率为18.86%。以上结果说明该文库质量较好,为进一步筛选相关功能基因打下基础;较低的冗余率说明该文库值得继续使用大规模ESTS测序的方法寻找相关功能基因,并为进一步使用基因芯片技术研究相关功能基因的表达谱提供便利。A normalized cDNA library from hepatopancreas of abalone Haliotis diversicolor supertexta exposure to Tributyltin was constructed.Total RNA were prepared using RDP reagent.Oligotex(QIAGEN)was used to separate the mRNA from total RNA.The first strand cDNA was synthesized by transcription of mRNA with the SMART technique.The LD-PCR was performed using a modified SMART primer as the primer set,and the first-strand cDNA as the template to synthesize double strand cDNA.Double strand cDNA was normalized using Duplex-Specific Nuclease(DSN).The containing capacity of the unamplified cDNA library was 4.3×106 CFU/ml with a recombinant rate of 97.9%.A total of 3 288 clones were random selected to be sequenced,and 3 048 high quality ESTs were generated.After processing,a total of 2 473 unigenes comprising 370 contigs and 2 103 singlets were obtained.The redundancy was 18.86%.These results indicate that the normalized cDNA library is suitable to be used for further cloning and analysis of genes related to tributyltin exposure.厦门市科技计划项目(3502Z20055024);国家自然科学基金(20877034);集美大学创新团队基金(2008A001

    戊型肝炎病毒衣壳蛋白特异性人源单克隆抗体的筛选与鉴定

    目的:建立从外周血快速筛选戊型肝炎病毒(Hepatitis E virus,HEV)衣壳蛋白特异性人源抗体的方法,从疫苗免疫者外周血中筛选出相应抗体并进行鉴定。方法:采用分选型流式细胞仪获得外周血中HEV衣壳蛋白特异性的记忆B细胞,通过单细胞RT-PCR的方法获得抗体序列,并进行重组表达,最后对获得的人源单克隆抗体进行初步性质鉴定。结果:成功筛选到识别HEV衣壳蛋白的6株人源单克隆抗体,6株抗体均具有抗原结合活性,4株抗体具有中和活性。结论:成功获得HEV衣壳蛋白特异性人源单克隆抗体序列,并进行真核表达,对抗体的性质进行初步鉴定,为后期研究疫苗免疫的人体内的抗体演化打下基础。国家自然科学基金(No.81571996);;福建省自然科学基金(No.2016J05201)资

    2017年厦门金砖会晤期间气象因素与管控措施对空气质量的影响

    2017年厦门金砖会晤期间采取了大气污染临时管控措施,使AQI小时值和日均值均达到了双优的预期目标.本文根据管控措施实施的前、中、后阶段,厦门及周边城市大气污染物浓度的变化,对气象因素和人为因素的影响分别进行分析.结果发现,气态污染物对临时管控措施的敏感性最强,SO2和NO2的降幅(39.9%和25.6%)明显高于PM2.5和PM10的降幅(5.5%和4.8%),台风外围带来的大风和降水可显著改变大气污染物的周期性变化规律.大气PM2.5组成及SO2/NO2、SO【math395z】/NO【math396z】、OC/EC和WSOC/OC等比值变化显示机动车(尤其是柴油货车)是本地区大气污染物的重要来源.控制变量分析显示,厦门金砖会晤期间气象因素对颗粒物和NO2削减的贡献更大(20.3%),而临时管控措施对SO2的削减效果更明显(23.2%),且有一半以上(51%~64%)的大气污染物来自外来源输送.国家重点研发计划(No.2016YFC0502901);;国家自然科学基金(No.41471390);;厦门大学校长基金本科生项目(No.20720162006)~

    Comparison of efficacy between mPCNL with holmium laser and EMS standard tract PCNL in the treatment of complex renal calculi under ultrasound guidance

    目的比较b超引导下微通道经皮肾镜钬激光碎石术(MPCnl)与标准通道经皮肾镜气压弹道联合超声碎石术(标准通道PCnl)治疗复杂性肾结石的临床疗效。方法对78例复杂性肾结石患者分别行MPCnl(35例)和标准通道PCnl(43例)治疗,术后对2组患者的手术时间、一期结石清除率和手术并发症等指标进行比较分析。结果 2组患者分别成功建立16f和24f经皮肾通道并一期行碎石术。MPCnl组手术时间明显长于标准通道PCnl组;一期结石清除率MPCnl组低于标准通道PCnl组,差异均具有统计学意义(P均38.5℃1例。2组并发症差异无统计学意义(P>0.05)。结论 b超引导下MPCnl与标准通道PCnl治疗复杂性肾结石安全有效、并发症发生率低,标准通道PCnl联合应用EMS第4代气压弹道联合超声碎石清石系统碎石清石效率高,较MPCnl钬激光碎石明显有优势,值得推广。Objective To compare the outcomes of the surgical treatment of complex renal calculi with minimally invasive percutaneous nephrolithotomy(mPCNL) and EMS percutaneous nephrolithotomy(standard tract PCNL) under ultrasound guidance.Methods A total of 78 patients with complex renal calculi were allocated to either mPCNL treatment group(mPCNL group,n=35) or EMS(swiss,the fourth generation) standard tract PCNL treatment group(standard tract PCNL group,n=43).The operative time,primary stone-free rate and operative complications in the two groups were compared.Results 16F and 24F percutaneous renal accesses were successfully established in mPCNL group and standard tract PCNL group respectively,and then the first lithotrity-lithiasis cleaning procedure was performed.The operation time in mPCNL group was longer than that in the standard tract PCNL group 〔(102±31) min vs(67±22) min,P38.5 ℃ occurred in one case of mPCNL group.There was no significant difference in operative complications in two groups(P>0.05).Conclusions Both the treatments of complex renal calculi with mPCNL and EMS(the fourth generation) standard tract PCNL under ultrasound guidance are similarly safe,but the standard tract PCNL has the superiority of higher primary stone-free rate and shorter operation time over the holmium Laser mPCNL.Therefore the standard tract PCNL merits propagation for treating complex renal calculi

    By using Western blot and immunofluorescence assays,the recombinant HEV capsid protein p239 was found specifically attached to the HepG2 cell surface and entered to the cytoplasm with the increase of incubation temperature.Pre-mixture of wild-type HEV with p239 blocked the infectivity of the virus on primary cultured human hepatocytes and HepG2 cells,indicating that p239 and HEV competed the same targeting site on these cells.These data provide evidence that p239 has a similar cell surface structure with wild-type HEV
