    THE EFFECT of HIGH AMMONIA CONCENTRATION ON GILL STRUCTURE ALTERNATION AND EXPRESSION of SOD AND HSP90 GENES IN GRASS CARP,CTENOPHARYNGODON IDELLA

    实验通过测定草鱼的24H半致死浓度,鳃的细胞结构以及SOd和HSP90的表达模式研究了草鱼在组织学和分子生物学水平上对高浓度氨氮暴露的响应。经过半致死实验确定的氨氮24H lC50为243 Mg/l试验中草鱼被置于5个浓度的处理组中(50、72、104、151、220 Mg/l),之后取鳃组织进行组织切片分析,取肝脏、肠和鳃来测定SOd和HSP90的表达情况。经过高浓度的氨氮暴露处理,鳃组织的细胞排列和结构产生了明显的变化,并且SOd和HSP90的表达受到了显著的上调。这些结果表明,高浓度的氨氮能够损害鳃部的细胞结构并且诱导应激蛋白的表达。这个结果同样显示出,SOd和HSP90可以作为评估草鱼氨氮暴露水平的良好指标。Histological and molecular responses of grass carp Ctenopharyngodon idella to high concentration ammonia exposure were studied in this study by determining the lethal concentration,gill cellular structure,and expression patterns of sod and hsp90 genes.Grass carps were treated at five ammonia concentrations(50,72,104,151 and 220 mg/L) for 24h,and then the gill tissues were collected for histological analysis.In addition,three tissues(gill,intestine and liver) were collected to measure the expression of sod and hsp90 genes.After exposure to high concentration of ammonia,the arrangement and structures of gill cells changed dramatically.Edema and fusion of secondary lamellae were observed,and the edge of cell and nucleus could not be clearly defined.Meanwhile,the two genes(sod and hsp90) in different tissues were unregulated significantly,indicating that high concentration of ammonia could impair the cellular structures and induce the expression of stress proteins.These results also suggested that sod and hsp90 were suitable biomarkers for ammonia exposure assessment in grass carp.FoundsoftheNationalProgramonKeyBasicResearchProject(2009CB118706

    Synthesis and Characterization of [Ni(Hlact)_2(phen)]·2H_2O and Its Interaction with BSA Studied by Fluorescence Spectroscopy

    利用溶液法合成了配合物[nI(HlACT)2(PHEn)]·2H2O(1),并对该配合物进行了元素分析、红外光谱和X-射线单晶衍射表征。通过荧光光谱法研究了不同温度下配合物1与牛血清白蛋白相互作用时的荧光强度的变化,计算了在不同温度下,配合物1与牛血清白蛋白(bSA)的结合常数、结合位点数以及热力学函数,并进一步讨论了配合物1与bSA相互作用时的作用力类型和两者之间的距离。结果表明,配合物1对牛血清白蛋白的荧光猝灭为静态猝灭过程,它与牛血清白蛋白的相互作用有一个位点,结合常数的平均值5.06x105l·MOl-1,作用距离为2.35 nM,相互作用力表现为氢键和范德华力。The compound [Ni(Hlact)2(phen)]·2H2O is synthesized in the solution and fully characterized by elemental analysis,IR and X-ray structural analyses.The interaction of Bovine Serum Albumin(BSA) and [Ni(Hlact)2(phen)]·2H2O at different temperatures are investigated with fluorescence spectra.The binding constant,binding sites and thermodynamic functions are calculated at different temperatures.The binding force and distance are discussed for BSA with complex 1.In the result,the model of fluorescence quenching is considered as static quenching process based on Stern-Volmer equation.The binding constant and binding sites are calculated for BSA and [Ni(Hlact)2(phen)]·2H2O as 5.06 ×105L·mol-1 and 2.35nm respectively.Parameters of thermodynamic functions are calculated and the interaction is determined as hydrogen bonding and van der Waal interactions between BSA and complex.国家自然科学基金(No.21073150)资助项

    Online Soft-measurement for Thermal Efficiency of Circulating Fluidized Bed Boilers

    提出循环流化床(Cfb)锅炉热效率的在线软测量方法,该方法将效率分为煤燃烧效率和传热效率两部分,根据Cfb锅炉动态机理模型在线计算锅炉未燃烧碳量,在此基础上计算煤燃烧效率,再通过反平衡法和正平衡法计算锅炉传热效率,最后得到锅炉热效率.应用该方法对某440T/H循环流化床锅炉热效率进行了计算,分析了影响该锅炉热效率的主要因素.结果表明:影响锅炉热效率的最主要因素是烟气中的飞灰含量;该软测量方法适用于Cfb锅炉,能有效指导锅炉调控,从而提高锅炉热效率.An online soft-measuring method for thermal efficiency of the circulating fluidized bed boiler(CFB)is being proposed.The method divides the efficiency into two parts,coal combustion efficiency and heat transfer efficiency.On-line calculation for the unburned carbon content of the boiler is carried out according to the dynamic mechanism model of the CFB boiler,after that combustion efficiency of the coal is calculated.Then heat transfer efficiency is calculated by indirect heat balance and direct heat balance,finally the boiler thermal efficiency is obtained.The thermal efficiency of certain 440 t/h CFB was calculated by the method,and the main factors which influence the thermal efficiency of the boiler were analyzed.Results show that the main factor which influences the thermal efficiency of the boiler is the content of fly ash in the flue gas.The soft-measuring method is suitable for CFB boiler,which can effectively direct the control of the boiler,thus improving the thermal efficiency of the boiler.福建省科技基金资助项目(2007J0165

    Experimental Study of the Abalone Shell on Cataract induced by Oxidative Stress

    目的研究石决明提取物对体外培养的晶状体氧化应激性白内障形成的作用及机制。方法离体培养小鼠晶状体,应用不同浓度的石决明提取物预孵育晶状体24h后,加入1mm过氧化氢,继续培养3小时后恢复正常培养,72小时后观察小鼠晶状体混浊程度,测定晶状体组织培养液中的乳酸脱氢酶(LDH)含量,晶状体组织中还原型谷胱甘肽(GSH)含量及超氧化物歧化酶(SOD)活力。结果石决明提取物在1~2 mg/ml浓度范围内减轻氧化应激造成的晶状体混浊,减少晶状体LDH的释放,提高组织内GSH含量和SOD活力。结论石决明提取物可减轻氧化应激白内障的形成,其作用主要与石决明提取物提高内源性抗氧化系统有关。Objective To study the effect of the abalone shell extract on oxidative stress induced cataract formation and its mechanism in cultured mouse lens in vitro. Methods The cultured mouse lens were pretreated with the abalone shell extract in different concentrations for24 hours,and then 1mm hydrogen peroxide was added and continued incubating for 3 hours,and they were changed to normal culture media.After 72 hours,the opacity of each lens was observed under an anatomical microscope,the content of lactate dehydrogenase( LDH)leakages,the content of the reduced glutathione( GSH) and activity of superoxide dismutase( SOD) in lens tissue were assayed. Results Abalone shell extract in the concentration range of 1 ~ 2 mg / ml reduced the lens opacity caused by oxidative stress,alleviated the release of LDH,and increased GSH content and SOD activity in cultured lens.
