Frequency and distribution of AP-1 sites in the human genome
The AP-1-binding sequences are promoter/enhancer elements that play an essential role in the induction of many genes in mammalian cells; however, the number of genes containing AP-1 sites remains unknown. In order to better address the overall effect of AP-1 on expression of genes encoded by the entire genome, a genome-wide analysis of the frequency and distribution of AP-1 sites would be useful; yet to date, no such analysis of AP-1 sites or any other promoter/enhancer elements has been performed. We present here our study of the consensus AP-1 site and two single-bp variants showing that the frequency of AP-1 sites in promoter regions is significantly lower than their average rate of occurrence in the whole genomic sequence, as well as the frequency of a random heptanucleotide suggesting that nature has selected for a decrease in the frequency of AP-1 sites in the regulatory regions of genes. In addition, genes containing multiple AP-1 sites are more prevalent than those containing only one copy of an AP-1 site, which again may have evolved to allow for greater signal amplification or integration in the regulation of AP-1 target genes. However, the number of AP-1-regulated genes identified in various studies is far smaller than the number of genes containing potential AP-1 sites, indicating that not all AP-1 sites are activated in a given cell under a given condition, and is consistent with the prediction by others that cellular context determines which AP-1 sites are targeted by AP-1
The mechanisms of proanthocyanidins Inhibits PGE_2 synthesis in lipopolysaccharide-Induced RAW264.7 cells
目的:观察原花青素对脂多糖诱导rAW264.7细胞PgE2生成的影响及其作用机制。方法:放射免疫法(rIA)检测原花青素对脂多糖诱导的rAW264.7细胞PgE2生成的影响;lPS诱导rAW264.7细胞9H,再加不同浓度原花青素作用30MIn,放射免疫(rIA)法检测原花青素对COX-2酶活性的影响;半定量逆转录-聚合酶联反应(rT-PCr)法检测原花青素对COX-2 MrnA表达的影响;提取核蛋白,蛋白免疫印迹(WESTErn blOT)法检测原花青素对nf-κb/P65蛋白表达的影响;电泳迁移率变动分析(EMSA)法检测原花青素对nf-κb与dnA结合活性的影响。结果:0.8,4和20Mg·l-1原花青素抑制lPS诱导rAW264.7细胞PgE2生成;0.8,4和20Mg·l-1原花青素不影响lPS诱导rAW264.7细胞COX-2酶活性;0.8,4和20Mg·l-1原花青素下调lPS诱导rAW264.7细胞COX-2 MrnA表达;4,20Mg·l-1原花青素下调lPS诱导rAW264.7细胞nf-κb/P65蛋白表达;0.8,4和20Mg·l-1的原花青素可明显降低lPS诱导下rAW264.7细胞的nf-κb活化。结论:原花青素显著抑制lPS诱导rAW264.7细胞PgE2生成的作用与原花青素抑制COX-2 MrnA表达有关,此作用可能是通过抑制nf-κb/P65蛋白表达和抑制nf-κb的dnA结合活性来实现。Objective:To study the effect of proanthocyanidins on PGE2 synthesis in lipopolysaccharide-induced RAW264.7 cells and its mechanisms.Methods: The effect of proanthocyanidins on PGE2 synthesis in lipopolysaccharide-induced RAW264.7 cells was measured by radioimmunoassay(RIA);After being pretreated with different concentrations of proanthocyanidins for 30 min,and then LPS 1 mg·L-1 for 9 h,the effect of proanthocyanidins on the activity of COx-2 enzyme in RAW264.7 cells was analysed by RIA;the expressions of COx-2 mRNA by RT-PCR;the nuclear protein was isolated and the expressions of NF-κB protein by Western blot;and the DNA-binding activity of NF-κB was measured by electrophoretic mobility shift assay(EMSA).Results: PGE2 synthesis was inhibited by proanthocyanidins 0.8 mg·L-1、4 mg·L-1 and 20 mg·L-1 ;the activity of COx-2 enzyme was not inhibited by proanthocyanidins 0.8 mg·L-1、4 mg·L-1 and 20 mg·L-1 (P>0.05,vs LPS group);the expression of COx-2 mRNA was inhibited by proanthocyanidins 0.8 mg·L-1、4 mg·L-1 and 20 mg·L-1;the expression of NF-κB/p65 protein was inhibited by proanthocyanidins 4 mg·L-1 and 20 mg·L-1.The DNA-binding activity of NF-κB was reduced by proanthocyanidins 0.8 mg·L-1、4 mg·L-1 and 20 mg·L-1.Conclusion: The mechanisms of proanthocyanidins inhibited PGE2 synthesis in lipopolysaccharide-induced RAW264.7 cells is related to the inhibition of the expression of COx-2 mRNA ,which is inhibited possibly by suppressing expression of NF-κB/p65 protein and DNA-binding activity of NF-κB.广东省教育厅自然科学基金(Z03045)---
具核梭杆菌诱导葡萄糖转运蛋白4 高表达在食管鳞癌组织中的临床意义及预后价值
【目的】分析食管鳞癌患者癌组织中具核梭杆菌(Fn)对葡萄糖转运蛋白4(GLUT4)的诱导效应与临床病理特征及5 年生存期的相关性,并探讨其临床意义及预后价值。【方法】选择2014 年1 月到2015 年3 月安阳肿瘤医院手术切除的96 例食管鳞癌患者癌组织石蜡包埋标本为研究对象,采用RNAscope 及免疫组织化学方法分别检测食管鳞癌癌组织中 Fn 感染及GLUT4 的表达情况,并分析 Fn 对GLUT4 的诱导效应,及其与临床病理特征相关性;采用 Kaplan-Meier 生存分析法绘制生存曲线并利用 Log-rank 检验方法分析 Fn 对 GLUT4 的诱导效应与生存时间之间相关性。【结果】食管鳞癌中可见癌细胞胞浆出现红色颗粒,为Fn 感染阳性,连续切片癌细胞浆膜出现棕黄色颗粒,为GLUT4 表达阳性;且Fn 感染与其表达具有显著一致性(P < 0.05)。同时Fn 诱导GLUT4 表达阳性组与性别、吸烟、饮酒、分化程度、浸润深度、淋巴结转移及临床分期显著相关(P < 0.05)。且阳性组5 年生存率及中位生存时间均明显低于阴性组(P < 0.05)。【结论】长期吸烟、饮酒会导致恶劣的口腔环境,Fn 更容易在这种环境下感染并定植,从而诱导癌细胞 GLUT4 高表达,增强葡萄糖代谢,促进食管鳞癌恶性进展。有效清除Fn 并抑制癌细胞GLUT4 表达可能为食管鳞癌治疗提供新策略和治疗手段
孔子及《论语》传播学研究:基于文献视角的回顾与前瞻
新时期以来的孔子及《论语》传播学研究,历经起步学习期、探索发展期和走向深化期三个阶段。综括30多年来从传播学层面研究孔子及《论语》的著述活动,有三个比较明显的研究方向:一是孔子及《论语》传播史实研究;二是孔子及《论语》传播思想研究;三是当代新闻传播事业语境下孔子及《论语》的价值转换研究。文章回顾了新时期以来孔子及《论语》传播学研究的进程,在此基础上认真总结取得的成绩和不足,并对开创孔子及《论语》传播学研究新局面提出了一些看法和建议
柚皮素通过诱导血红素加氧酶-1的表达抑制氧化应激和血管钙化
目的研究柚皮素对血管钙化发生的作用及氧化应激机制。方法采用不同浓度的柚皮素(5、 10、 20 μmol/L)处理大鼠血管平滑肌细胞和血管环,研究柚皮素对高磷高钙诱导的血管平滑肌细胞和血管环钙化程度的影响。用茜素红染色法测定细胞和血管环钙化的程度,荧光探针DHE检测活性氧(ROS)水平,qPCR和western blot检测血红素加氧酶-1(HO-1)的表达水平。采用HO-1抑制剂ZnPP9处理血管平滑肌细胞,研究抑制HO-1对平滑肌细胞钙化的影响。结果柚皮素能明显减轻高磷高钙诱导的血管平滑肌细胞和血管环钙化。此外,柚皮素可明显上调HO-1的表达水平,降低ROS的水平;ZnPP9能阻断柚皮素对血管平滑肌细胞钙化的抑制作用。结论柚皮素可通过上调HO-1的表达水平,降低ROS的水平和抑制高磷高钙诱导的血管钙化
基于P2P技术的多媒体网络会议模型
本文提出了一种基于P2P技术的多媒体网络会议设计模型,该模型采用Chord协议组织节点和实现节点路由定位,将服务器的职能分布到P2P Overlay的各节点上,并根据节点的异构性将它们分为普通节点和超级节点,各节点根据自身能力各尽所能,使网络资源得到充分利用,可有效解决C/S模式下中央服务器带来的性能瓶颈问题。仿真实验表明该网络会议系统模型具有负载均衡、易于扩展和高效的资源定位等特点
