    独活寄生汤水提物对退变大鼠椎间盘软骨细胞功能的影响

    目的探究独活寄生汤水提物对经IL-1β诱导退变的大鼠椎间盘软骨细胞中Wnt4、GSK-3β、β-catenin、DKK-1的影响。方法采用机械-酶消化法分离大鼠椎间盘软骨组织,进行软骨细胞体外培养、镜下观察与鉴定,分为正常组、模型组(经10 ng/m L浓度IL-1β造模)、实验1组(独活寄生汤水提物组200μg/m L干预24 h)、实验2组(独活寄生汤水提物组200μg/m L干预48 h)。观察4组大鼠椎间盘软骨细胞中Wnt4、GSK-3β、β-catenin、DKK-1 m RNA与蛋白的表达及上清液中Sox 9表达。结果 (1)第2代大鼠椎间盘软骨细胞增殖速度快,呈现多边形,胞核清晰,且含有1~2个核仁,融合后出现\"铺路石\"状,经Ⅱ型胶原法染色后,阳性对照组胞浆区域浸染为棕黄色;(2)与正常组比较,模型组中软骨细胞Wnt 4、GSK-3β、β-catenin m RNA与蛋白表达及上清液Sox 9含量明显提高(P<0.05),DKK-1 m RNA与蛋白表达明显降低(P<0.05);与模型组比较,实验1组、实验2组中软骨细胞Wnt 4、GSK-3β、β-catenin m RNA与蛋白表达及上清液Sox 9含量明显降低(P<0.05),DKK-1m RNA与蛋白表达明显提高(P<0.05),以实验2组变化最为显著(P<0.05)。结论独活寄生汤水提物组可调控退变椎间盘软骨细胞的功能,下调Wnt 4、GSK-3β、β-catenin和上调DKK-1 m RNA与蛋白表达,进而延缓大鼠椎间盘软骨细胞的退变。福建省自然科学基金项目(2017J01224);;福建中医药大学校管课题资助(X2015034

    目的:从Wnt/β-catenin信号通路探讨独活寄生汤水提物对大鼠椎间盘退变软骨细胞功能的影响。方法:(1)用水提加热回流法制备独活寄生汤水提物成分;(2)选取4周龄健康雄性SD大鼠30只,采用机械-酶消化法分离大鼠椎间盘软骨组织,建立软骨细胞体外培养体系并进行鉴定;(3)RT-PCR、We stern blot法分别检测经DKK-1抑制剂干预及(或)经白细胞介素-1β诱导的椎间盘软骨细胞Wnt4、GSK-3β、β-catenin mRNA与蛋白含量的表达。结果:(1)椎间盘软骨细胞经Ⅱ型胶原法染色后,阳性对照组胞浆区域浸染为棕黄色;(2)RT-PCR、Western blot检测结果示,与正常组比较,模型组Wnt4、GSK-3β、β-catenin mRNA与蛋白含量表达显著升高(P<0.01);与模型组比较,独活寄生汤水提物组(100,200,400μg·mL-1)的Wnt4、GSK-3β、β-catenin mRNA与蛋白含量表达显著降低(P<0.01或P<0.05),其中以200μg·mL-1组的表达量最低(P<0.01)。结论:独活寄生汤水提物组可通过调控Wnt/β-catenin信号通路,下调大鼠椎间盘退变关节软骨中Wnt4、GSK-3β、β-catenin mRNA与蛋白含量表达,从而延缓椎间盘软骨细胞退变。福建省自然科学基金(2017J01224);;福建中医药大学校管课题(X2015034

    中药保留灌肠治疗溃疡性结肠炎疗效的荟萃分析

    目的:系统评价中药保留灌肠治疗溃疡性结肠炎(ulcerative colitis,UC)的有效性和安全性。方法:检索CochraneLibrary、PubMed、中国期刊全文数据库(CNKI)、中国生物医学文献数据库(CBM)、万方数据库(Wanfang Data)、中文科技期刊全文数据库(VIP)建库至2017年7月的所有文献。按照Cochrane系统评价方法查找中药保留灌肠治疗UC的临床随机对照试验(RCTs),进行数据提取和质量评价后,采用RevMan 5.3软件进行荟萃(meta)分析。结果:共纳入36篇文献。Meta分析结果显示:治疗组的总有效率优于对照组[RR=1.20,95%CI(1.15,1.25),P<0.000 01];治疗后治疗组中医证候积分低于对照组[SMD=-1.55,95%CI(-2.03,-1.07),P<0.000 01];治疗后治疗组内镜积分低于对照组[SMD=-1.63,95%CI(-2.45,-0.81),P<0.000 1];治疗组复发率低于对照组[RR=0.36,95%CI(0.25,0.52),P<0.000 01];治疗组药品不良反应(ADRs)发生率低于对照组[RR=0.23,95%CI(0.12,0.44),P<0.000 1]。结论:中药保留灌肠治疗可以提高UC的总有效率,减轻相关症状,同时有效降低UC的复发率,减少ADRs发生率。解放军第一七五医院2016年青年苗圃课题(16Y025

    Electrodeposition of CIGS Thin Film with Special Structure Using Laser Etching Template

    &nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 本文利用激光刻蚀模板,在水溶液中电沉积制备金属铜薄膜,讨论了温度、电流、硫酸铜浓度对薄膜形貌的影响. 采用SEM对制备的铜薄膜进行表征,结果表明在沉积温度为30 ℃,沉积电流为4 A&middot;dm-2(表观工作电流密度),硫酸铜浓度在20 ~ 50 g&middot;L-1的水溶液中电沉积可以得到中空馒头状和开口碗状结构的铜薄膜. 利用激光刻蚀模板,在离子液体1-丁基-3-甲基咪唑三氟甲磺酸盐([BMI][TfO]) - 30 Vol%丙醇混合电解质中电沉积CIGS薄膜,研究了沉积电势、沉积时间对薄膜形貌的影响. SEM观察发现,在沉积电势为-1.8 V,沉积时间为1.5 h条件下电沉积可以得到近似柱状的簇状花束样的CIGS薄膜, 电沉积铜后再进一步电沉积CIGS,得到了均匀有序的鼓包柱状结构的Cu/CIGS复合薄膜. 用恒电势方波法对制备的薄膜真实表面积进行测试,计算结果表明,与无模板电沉积制备的CIGS薄膜相比,激光刻蚀模板法制备的Cu/CIGS复合薄膜的表面积提高了约8倍.Copper indium gallium selenide (Cu1.00InxGa1-xSe2.00, CIGS) thin film solar cells have a wide application in semiconductor thin film batteries. The CIGS absorbing layer is the key of the thin film solar cell material. Therefore, it is of theoretical and practical significance to carry out researches in CIGS thin films. In this work, the CIGS thin films with special structures were prepared by using laser etching template. The copper thin film was firstly electrodeposited in aqueous solution with Laser etching template. The influences of temperature, current and concentration of copper sulfate on morphology and structure of the thin film were studied. The results showed that a hollow steamed bun shape and open bowl structure was formed at the surface of copper thin film electrodeposited at 30 oC and at the apparent current density of 4 A&middot;dm-2 in the aqueous solution with the concentrations of CuSO4&middot;5H2O being 20 ~ 50 g&middot;L-1. The CIGS thin films were electrodeposited by using laser etching template in the ionic liquid 1-butyl-3-methylimidazolium trifluoromethanesulfonate ([BMIm][TfO]) - 30 Vol% propyl alcohol mixed electrolyte solution at different deposition potentials, substrates and deposition time. It was demonstrated that the clusters of flowers with approximate columnar CIGS thin film could be prepared at 1.8 V for 1.5 h. Furthermore, the electrodeposition of CIGS thin films was done on the previously electrodeposited copper substrate by Laser etching template. A uniform and orderly spheroidal structure of Cu/CIGS composite film was obtained. The apparent surface area of the Cu/CIGS composite film, calculated by constant voltage square wave method, was about 8 times larger than that of the CIGS thin film prepared without using template, which greatly enhanced absorption of light on the thin film and increased the output of the carrier.
