    Preparation and determination of polyclonal antibody to thymosin α1

    目的 :制备用于检测转胸腺素基因蓝藻表达产物的特异性抗体。方法 :用提纯的小肽 (2 8个氨基酸 )胸腺素α1与牛血清白蛋白偶联后作为抗原 ,采用皮下多点注射免疫的方法免疫大鼠。经过 3个月的免疫 ,获得抗Tα的多克隆抗体。结果 :用间接酶联免疫吸附 (ELISA)方法 ,测得抗体效价超过 40 96。蛋白印迹 (Westernblot)检测结果显示 ,该抗体能特异性地与胸腺素α1抗原产生明显免疫亲和反应。结论 :所制备的抗体具有很好的灵敏性和特异Objective:To prepare specific antibody against Thymosin α1 Methods:Thymosin α1(Tα1,28 peptide) was conjuncted with BSA as immune antigen,rats were immunized and the polycolonal antibody to Tα1 was obtained.Results:With ELISA detection,the titer of antibody was more than 1:4 096;Western blot analysis showed that the antibody can bind with Tα1 specifically.Conclusion:The prepared antibody against Tα1 has good specificity and reactivity and can be used to detect the expression products of transgenic cyanobacteria which expressed Tα1 gene.国家“863”课题资助!(No 819 0 4 0 3

    电解液在电化学反应过程中起到了重要的支撑作用,是电解、电镀的反应介质,被喻为电池的“血液”。添加剂作为电解液的重要组成部分,在电化学反应过程中起到了修饰、改善形貌及提高物化性能的作用。例如在电解、电镀过程中起到了降低反应温度、整平、细晶等作用,在锂电池电解液中可以起到修饰SEI膜、提高电导率、除水、降酸、阻燃、限压等作用,从而改善电池的倍率、高低温、循环、安全等性能。目前添加剂种类繁多、性能单一、作用机理不明,难以满足研发及生产的需求,因此添加剂的多功能化及作用机理即成为研究热点。本文系统研究了五类添加剂在氯铝酸盐离子液体电解液中对铝沉积层纯度、形貌、结构的优化作用。设计合成了以烟酰胺为代表的新型多功能添加剂,研究其在锂离子电池中的性能及作用机理。得到如下主要结论: (1) 选用[Bmim]Cl/2AlCl3离子液体为低温电沉积铝电解液体系。无添加剂的情况下电沉积铝的过程属于准可逆过程,为受扩散控制的三维瞬时成核机理。分别考察了无机金属氯化物、稀土元素氯化物、小分子有机添加剂、大分子有机添加剂、多种添加剂混合物这五大类添加剂对电沉积过程的影响。其中,无机氯化物添加剂易与铝形成共沉积;有机添加剂对沉积层纯度无影响,同时具有很好的细晶作用;复合添加剂LaCl3-NH4Cl-C6H12可以有效改善沉积层质量,获得纳米级铝沉积层,并可将铝纯度从99.795 %提高至99.95 %,达到精铝级别。 (2) 以玻璃碳为惰性阳极进行离子液体低温电解铝实验。在不使用添加剂时,随着电解电位的增大铝在阴极的成核经历了由连续成核到瞬时成核的过程,电解铝产品粗糙、纯度较低。加入复合添加剂LaCl3-NH4Cl-C6H12后,铝呈层状生长,沉积层具有银白色金属光泽,且纯度可以达到99.994 %的精铝级别。 (3) 通过筛选有机胺类化合物,确定采用可与水形成稳定氢键(O…H-O和N-H…O)、除水效果明显、无副反应的酰胺类添加剂作为锂离子电池电解液稳定剂。此外,酰胺类添加剂具有明显的成膜性能,可以在溶剂分解前形成SEI膜。在循环性能测试中,加入0.5 % N-甲基乙酰胺可将容量保持率由91.67 %提高至95.99 %,同时内阻明显降低。 (4) 研究了烟酰胺、丙烯酰胺、N,N-亚甲基双丙烯酰胺三种新型添加剂的作用机理及性能。实验发现烟酰胺除了具有显著的除水降酸作用,还可以在电极表面形成优质的SEI膜。烟酰胺可使电解液的酸值在50 天内保持不变,明显优于其他添加剂体系。加入0.1 %的烟酰胺即可促进电极表面成膜,100次循环后容量保持率可达95.92 %,提高锰酸锂电池的循环和倍率性能,同时内阻较低。 (5) 根据以上两种电解液的电化学特性,设计并制作了“电化学过程原位研究装置”和多功能原位光电化学池,建立了一种简便高效的电化学过程原位研究与评价平台

    为了克服传统基于聚类的图像分割算法需要指定聚类数目以及依赖初始值等缺点,提出了一种基于动态小生境的人工鱼群算法的图象分割方法。该算法将图像分割问 题转化为根据图像像素特征对像素的自动聚类问题。采用更为简单的个体描述方式,每条人工鱼表示一个分割区域的一个可行解,并对进化过程中的人工鱼进行动态 的划分小生境,每个小生境对应了图像分割问题中一个分割区域。通过对鱼群行为的模拟及种群的动态划分实现了对图像分割问题的分割区域中心和区域数的同时进 化,实现了一种新的聚类算法,并实现了对图像的自动分割。实验结果表明:该算法可以自动地估计分割的区域数,并获得较好的分割性能。In order to overcome the defects in the traditional clustering-based image segmentation algorithm, e.g., it needs to specify the number of clusters, it is sensitive to initial value, and so on, an image segmentation method based on dynamic niche artificial fish-swarm algorithm (DNAF) is presented in this paper. In the new algorithm, the image segmentation problem is transformed into an automatic pixel clustering process based on the pixel features of the image. A simpler representation is adopted, each artificial fish represents a single feasible solution of one segmented area. Moreover, the dynamic identification of the fish niches is performed at each generation to automatically evolve the optimal number of regions. Each fish niche corresponds to one segmentation region in the image segmentation problem. Therefore, the proposed DNAF algorithm implements simultaneous evolution in the center of the segmentation region and the optimal number of regions through simulation on the behaviors of fish swarm and the dynamic division of population. It thereby achieves a new clustering algorithm and automatic segmentation of an image. Experiment results demonstrate that the DNAF algorithm is able to automatically estimate the number of the segmented regions, and an excellent segmentation performance can be attained

    Expression of Thymosin α_1 Gene in Spirulina platensis

    将钝顶螺旋藻 (Spirulinaplatensis)在 2 4℃培养 ,并经 2mmol/LEDTA预处理16~ 2 4h ,以本实验室构建的基因整合平台系统供体质粒 pEUTISI进行超声波转化螺旋藻 ,经筛选获得了具G4 18抗性的转化藻株。通过PCR扩增和Southern杂交证实 ,pEUTISI中目的基因UB Tα1和nptII基因已整合到钝顶螺旋藻染色体上。转化藻株经4 5℃热诱导 4 0min后 ,进行蛋白质SDS PAGE电泳和Westernblot,杂交结果证实 ,外源胸腺素α1基因在螺旋藻中得到有效表达。By ultrasonic transformation, donor plasmid pEUTISI of gene intergration platform system was successfully introduced into Spirulina platensis which had been cultured at 24℃ and treated with 2mmol/L EDTA for 16~24 hours. Screened by G418, the transformants which were integrated the foreign DNA were selected. PCR and southern blotting data show that the foreign DNA including nptII gene and UB-Tα 1 gene has been integrated into Spirulina platensis chromosomal DNA. Further confirmation was tested by SDS-PAGE and western blotting after the transformants were heat shock induced at 45℃ for 40 min.
