Chronic hypoxia enhances aggressiveness of MCF-7 breast cancer cells through EMT
目的:探讨慢性缺氧应激对人乳腺癌MCF-7细胞恶性生物学行为的影响及可能机制。方法:将人乳腺癌MCF-7细胞分为缺氧组(1%O2、5%CO2和94%N2)和正常对照组(常氧)进行培养。利用MTT法、CCK-8实验、细胞直接计数法及细胞侵袭和迁移实验对MCF-7细胞活力、增殖及侵袭和迁移能力进行检测;用软琼脂集落形成实验及Matrigel 3D培养技术检测MCF-7细胞非锚定生长能力及极性改变情况;利用MCF-7细胞构建裸鼠皮下种植瘤模型,检测慢性缺氧应激对体内肿瘤生长及肺转移的影响;利用倒置显微镜观察MCF-7细胞形态改变;Western blot检测低氧诱导因子1(hypoxia-inducible factor-1,HIF-1)和磷酸化的糖原合成酶激酶3β(glycogen synthase kinase-3β,GSK-3β)在缺氧环境下表达水平的改变,以及E-钙黏连蛋白(E-cadherin)、N-钙黏连蛋白(Ncadherin)、波形蛋白(vimentin)、基质金属蛋白酶3(matrix metalloproteinase-3,MMP-3)、MMP-9等上皮-间充质转化(epithelial-mesenchymal transition,EMT)相关蛋白的表达水平。结果:与正常对照组相比较,慢性缺氧组MCF-7细胞活力、增殖能力及侵袭迁移能力增强,细胞非锚定生长能力提高且在3D培养系统更容易发生极性改变,呈现侵袭样生长,体内生长及转移能力增强;除了HIF-1被缺氧诱导表达升高外,GSK-3β呈现活化趋势,且上皮样标志物E-cadherin蛋白表达水平明显下降,而间充质样标志物N-cadherin、vimentin、MMP-3和MMP-9蛋白表达水平明显升高。结论:慢性缺氧应激促进了乳腺癌细胞恶性生物学行为,且其机制可能与EMT有关。AIM:To investigate the effects of chronic hypoxia on the aggressiveness of MCF-7, a human breast cancer cell line , and the underlying mechanisms .METHODS: MCF-7 cells were cultured under hypoxia ( 1% O2 , 5%CO2 and 94%N2 ) or control (95%O2 and 5%CO2 ) condition.The viability, proliferation, and invasion and migration abilities of the MCF-7 cells were determined by MTT assay , CCK-8 assay, cell counting, and cell invasion and migration assays.Anchorage-independent growth and the alteration of cellular polarization of the MCF-7 cells were tested by soft agar colony formation assay and Matrigel-3D culture assay, respectively.The effects of chronic hypoxia on the growth and metas-tasis of MCF-7 cells in vivo were investigated by xenograft in nude mice .The morphological changes of the MCF-7 cells were observed under an inverted microscope .Hypoxia-induced alterations in the levels of hypoxia inducible factor-1 ( HIF-1 ) and phosphorylated glycogen synthase kinase-3β( p-GSK-3β) as well as epithelial-mesenchymal transition ( EMT) mol-ecules, such as E-cadherin, N-cadherin, vimentin, matrix metalloproteinase ( MMP)-3 and MMP-9, were determined by Western blot .RESULTS:Chronic hypoxia significantly increased the viability , proliferation , and invasion and migration abilities of MCF-7 cells in vitro, enhanced the anchorage-independent growth , facilitated cellular polarization alteration in Matrigel-3D culture, and promoted cancer metastasis in vivo.Hypoxia up-regulated HIF-1, activated GSK-3β, down-regu-lated E-cadherin and increased the protein levels of N-cadherin, vimentin, MMP-3 and MMP-9.CONCLUSION:Chronic hypoxia enhances the aggressiveness of breast cancer cells probably through EMT .中国博士后科学基金第59批面上资助项目(No.2016M592037);安徽省自然科学基金资助项目(No.1608085MH187

Optical System for Asymmetrical Crossed Czerny-Turner Raman Spectrometer with High Luminous Flux
为克服分立式便携拉曼光谱仪光通量低的缺点,设计了一种集拉曼探头光路与分光系统于一体的光学系统。探头光路采用大数值孔径的非球面透镜实现样品的有效激发和信号的高效收集,通过胶合透镜组缩小会聚光路尺寸、消除轴向色差。分光系统基于交叉非对称Czerny-Turner结构,为获得期望的光谱分辨率和光谱范围,建立了分光系统光谱分辨率及光谱范围与交叉非对称Czerny-Turner结构参数的关系。由测得的汞灯谱图可知,分光系统的光谱分辨率优于6 cm~(-1)(0.37 nm),光谱范围为790~950 nm(200~2000 cm~(-1))。将设计的光学系统对CCL4进行测试,实验结果表明在相同积分时间内由这种整体式的光学系统检测到的CCL4光谱谱峰强度是用商业探头通过光纤连接分光系统检测到的近3倍,验证了光学系统设计的合理性。To overcome the low luminous flux in separated portable Raman spectrometer, an optical system embodies the probe of Raman spectrometer and monochromator is designed. An aspherical lens with high numerical aperture is used to obtain the effective excitation of sample and the efficient collection of signal in the optical system of the probe. A cemented lens group is used to minish the size of converging path and eliminate chromatic aberration.The monochromator is based on asymmetrical crossed Czerny-Turner structure. For the purpose of achieving desired spectrum resolution and spectrum range, the relationship between the spectrum resolution and spectrum range of the monochromator and the structural parameters of the asymmetrical crossed Czerny-Turner system is established. According to the tested spectrogram of mercury lamp, the spectrum resolution of the monochromator is better than 6 cm~(-1)(0.37 nm), and spectrum range is 790~950 nm(200~2000 cm~(-1)). A sample of CCL4 is introduced to examine the performance of the optical system, the experimented results demonstrate that the peak intensity of spectrogram of CCL4 measured by this incorporative optical system is almost three times as high as that using a commercial probe couples to the monochromator, which verifies the rationality of the design of the optical system.国家自然科学基金(21503171);; 中央高校基本科研业务费项目(20720150091、20720150094);; 福建省高端装备制造协同创新中心资
卷积神经网络模型在儿科疾病预测中的应用
目的:针对儿童看病需求量大导致的儿科诊疗服务效率和准确率偏低等问题,利用自然语言处理和深度学习技术,从儿科历史病历数据中自动\"学习\"专家医生诊断模式,形成智能辅助诊断模型,从而对新的儿科病历数据输出疾病诊断决策。结果:基于深度卷积神经网络的七分类疾病智能诊断模型的正确率为84.26%,F1-score为84.33%,基本达到可投入实际应用的级别。结论:智能诊断决策作为预诊信息提供给医生进行确诊参考,对提升医生诊断速度效果明显。国家自然科学基金面上项目(编号:71571056);;福建省自然科学基金面上项目(编号:2012J01274)~
Effects of mixed-culture densities of Spartina alterniflora on eco-physiological characteristics of Kandelia candel seedling
按照不同密度将互花米草与秋茄进行混种,对比研究胁迫条件下秋茄幼苗生长、光合特性及其渗透调节物质变化规律。结果表明,低密度互花米草促进秋茄的茎长以及各部分生物量,高密度则起抑制作用。随着互花米草密度的增大,秋茄幼苗叶片光合速率、气孔导度、蒸腾速率、水分利用率和蛋白含量均下降;相反,胞间CO2浓度、可溶性糖、淀粉、脯氨酸含量却上升,这些变化有利于对抗互花米草带来的不利影响。

Development of TOF-MS Used as the Detector of HPLC

基于表面增强拉曼光谱的合成色素专利蓝V的快速检测
食品安全问题一直是社会和广大群众关注的焦点问题,食品安全现状较为严峻,因此实现食品中有害物质的快速检测具有重要的实际意义。合成色素是一种常见的食品添加剂,然而合成色素的超标添加和非法添加依旧是食品安全中的重要问题之一,极大地危害人民群众的身体健康和食品工业的健康发展。常见的合成色素检测方法,均存在耗时长、费用高等缺点,不适应于合成色素的实时监测和快速筛查。为克服传统方法的缺点,提出利用表面增强拉曼光谱检测技术对合成色素进行检测,该方法具有检测速度快、检测灵敏度高等优点,能够达到现场实时检测的目的。此外,由于拉曼检测方法往往依赖于复杂的样品前处理操作,而常见的固相萃取技术一般依赖于人工操作,过程复杂且耗时较长,严重影响食品快速检测效率。因此,开发了一种全自动固相萃取装置,通过设计嵌入式硬件电路系统及其软件,精确控制蠕动泵流速和多路阀门开关实现了活化、上样、淋洗、洗脱四个步骤的全自动操作和参数控制,从而达到食品样品的全自动快速固相萃取。在实验部分,配制不同专利蓝V浓度的果汁饮料,然后利用该装置对果汁中的专利蓝V进行前处理,对萃取柱填料和萃取中各个步骤的时间和试剂进行了合理的选择,利用表面增强拉曼光谱检测技术成功地检测了合成色素中的专利蓝V。实验结果表明,所研制的自动固相萃取装置对比传统手工萃取,每个样品节省了近一半的萃取时间(10 min降为5 min)且能够同时处理5个样品,萃取时间稳定不易受人为因素影响,从而极大地提高了萃取效率和稳定性。此外,通过自动萃取获得的样品,对比手工萃取操作,因其受外界干扰相对较小,能够得到更强的拉曼光谱信号(约增强50%),获得了满意的萃取效果。对不同浓度的专利蓝V样品的结果显示,该方法能够实现检出质量浓度在0.5 mg·L-1水平,可有效满足现场监测需求。具有快速、方便、灵敏度高等特点。国家自然科学基金项目(21874113)资
