    Postglacial vegetation history of local phenomena in western Bohemia

    Region západních Čech představuje jednu z nejméně prozkoumaných oblastí z hlediska studia postglaciální historie vegetace v České republice. Právě zde se přitom předpokládá existence jedné z migračních cest dřevin do severnějších oblastí Evropy během Holocénu. Nejvhodnější oblasti pro studium skýtají sudetská pohoří. Jedním z nich je i oblast Slavkovského lesa v Karlovarském kraji. Postglaciální historie vegetace zde byla studována na dvou profilech - ze slatiniště u Číhanských pramenů a z PR Mokřady pod Vlčkem v blízkosti tělesa Mnichovských hadců. S využitím palynologické analýzy byly vytvořeny jak procentuální, tak influxové diagramy vývoje vegetace. Dále byly z České palynologické databáze vybrány datované profily ze západní části České republiky, které umožnily rekonstrukci vývoje migrace jednotlivých druhů hlavních dřevin v tomto regionu. Lokalita Číhaná zachycuje období od konce glaciálu do staršího Atlantiku. Ve staroholocénních lesích představovala silnou dominantu borovice, už během preboreálu se přidávala líska, která zde ale neměla natolik významné postavení. Na počátku boreálu následovaly dřeviny smíšených doubrav. V Atlantiku se rychle stal dominantou smrk. Sedimentace zde byla ukončena zřejmě důsledkem rychlého růstu rašeliny a následného provzdušnění substrátu dřevinami. The most appropriate area for testing of such proposal might be western Sudetes with Slavkovský les protected landscape area in the Karlovy Vary region. Postglacial vegetation history was studied on the basis of two sedimentary profiles - from the fen bog near Číhanské prameny springs and from the vicinity of Mnichovské hadce serpentines from the nature reserve Mokřady pod Vlčkem. Percentual as well as influx diagrams were constructed on the basis of results of standard pollen analysis. Comparison of these results with the data from other suitable profiles enabled reconstruction of migration history of main tree taxa. The Číhaná profile reflects the period from the Older Dryas up to the Atlantic. The oldest Holocene forest was formed by the dominant Pinus, while Corylus was joining already in the Preboreal. Quercetum mixtum folowed in the begining of the Boreal and spruce quickly gained dominance in the Atlantic.     Salicylanilide Diethyl Phosphates as Potential Inhibitors of Some Mycobacterial Enzymes

    Antimycobacterially active salicylanilide diethyl phosphates were evaluated to identify their potential drug target(s) for the inhibition of several mycobacterial enzymes, including isocitrate lyase, L-alanine dehydrogenase (MtAlaDH), lysine ε-aminotransferase, chorismate mutase, and pantothenate synthetase. The enzymes are related to the nongrowing state of Mycobacterium tuberculosis. Salicylanilide diethyl phosphates represent new candidates with significant inhibitory activity especially against L-alanine dehydrogenase. The most active MtAlaDH inhibitor, 5-chloro-2-[(3-chlorophenyl)carbamoyl]phenyl diethyl phosphate, has an IC50 of 4.96 µM and the best docking results. Other mycobacterial enzymes were mostly inhibited by some derivatives but at higher concentrations; isocitrate lyase showed the highest resistance to salicylanilide diethyl phosphates

    Analysis of public discussion on the topic of the position of wemen in decision making processes

    The Abstract The thesis is focused on views on the role of women in decision-making processes. Public and private sector is characterised by a low level of representation of women in decision-making positions. In connection to the directives of the European Commission, which introduce mechanisms supporting an increase the number of representatives of underrepresented sex in supervisory boards, the debate unleashed on the subject of quotas. Opinions on the effectiveness of quota mechanisms in the Czech Republic are reflected in the media space. This paper aims to describe and analyze the opinions of public discussion on the mechanisms of quotas. Through a discourse analysis of media contributions it tries to determine what arguments planes occurring in the public debate on a subject introduction of quotas. Analyzed articles are dated to the period between May 2014 and May 2015. Individual argumentative discourse planes are systematically coded by discursive rules in recurrent categories. Categories are also thematically merged into discursive planes with respect to their intertextuality and intediscursivity. The results of the analysis are then subjected to a comparison to the development of discursive anchoring the topic of quotas in the European environment

    Nové cholinesterázové inhibitory na bázi karbamátů a thiokarbamátů

    Based on the presence of carbamoyl moiety, twenty salicylanilide N,N-disubstituted (thio)carbamates were investigated using Ellman’s method for their ability to inhibit acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). O-Aromatic (thio)carbamates exhibited weak to moderate inhibition of both cholinesterases with IC50 values within the range of 1.60 to 311.0 μM. IC50 values for BChE were mostly lower than those obtained for AChE; four derivatives showed distinct selectivity for BChE. All of the (thio)carbamates produced a stronger inhibition of AChE than rivastigmine, and five of them inhibited BChE more effectively than both established drugs rivastigmine and galantamine. In general, 5-chloro-2-hydroxy-N-[4-(trifluoromethyl)- phenyl]benzamide, 2-hydroxy-N-phenylbenzamide as well as N-methyl-N-phenyl carbamate derivatives led to the more potent inhibition. O-{4-Chloro-2-[(4-chlorophenyl)carbamoyl]phenyl} dimethylcarbamothioate was identified as the most effective AChE inhibitor (IC50 = 38.98 μM), while 2-(phenylcarbamoyl)phenyl diphenylcarbamate produced the lowest IC50 value for BChE (1.60 μM). Results from molecular docking studies suggest that carbamate compounds, especially N,Ndiphenyl substituted representatives with considerable portion of aromatic moieties may work as non-covalent inhibitors displaying many interactions at peripheral anionic sites of both enzymes. Mild cytotoxicity for HepG2 cells and consequent satisfactory calculated selectivity indexes qualify several derivatives for further optimization.Pomocí Ellmanovy metody byla sledována schopnost 20 nových salicylanilidových karbamátů a thiokarbamátů inhibovat acetylcholinesterázu a butyrylcholinesterázu
