    Beta-thalassemia mutation types and the relationship with the demographic factors in Sanliurfa, Turkey

    Introduction: Beta-thalassemia is an autosomal recessive disease that occurs as a result of a disorder in the (β-globin chains synthesis), and the gold standard method for diagnosis is genetic mutation analysis. It is important to know the distribution of mutations according to regions and races. The aim of this study is to document the mutations in the beta-globin gene of beta-thalassemia major and intermedia patients who were followed and treated in Sanliurfa province, and to examine the relationships between these mutations by defining them according to gender, nationality, consanguineous marriage, history of disease in siblings and blood type.Methods: The files of 272 patients diagnosed with beta-thalassemia major and intermedia followed up in the Pediatric Hematology-Oncology outpatient clinic of Sanliurfa Training and Research Hospital between August 2016 and August 2017 were retrospectively reviewed and mutation analyzes were documented. Coding exons and exon-intron junction regions of beta globin-HBB gene were amplified by PCR method and then DNA sequencing was performed. Gender, nationality, consanguineous marriage, sibling history and blood type information were recorded.Results: Out of 272 patients, 94.1% were thalassemia major and the others were thalassemia intermedia. Approximately one third of the patients (30.1%) were foreign nationals. A total of 27 different mutations in the beta-globin gene were detected. The most common mutation is IVS-I-110 c.93-21 (G gt; A) (23.1%), which is followed by IVS-I-1 c.92 + 1 (G gt; A) (15.8%) and Codon 39 c.118 (C gt; T) (11.5%). One hundred and forty-two individuals (52.2%) had no sibling history, while 103 (37.9%) had one sibling and 27 (9.9%) had two siblings with thalassemia disorder. First-degree, second degree and third degree consanguineous marriages were present in 42.6% (n = 116), 8.1% (n = 22), 11% (n = 30) of parents, respectively.Conclusions: Beta-thalassemia disease is a common hematological condition in Sanliurfa. Approximately one-third of the patients who apply are foreign nationals. Correct identification of beta-globin gene mutations will guide genetic counseling and preventing prenatal disease. This data can contribute to the national thalassemia prevention program in Turkey

    Antiproliferative and apoptotic effects of onobrychis albiflora extract on HCT-116 cells

    In this study, the cytotoxic effects of the extracts of methanol: water (80:20) prepared from the above-ground parts of the varieties of Onobrychis albiflora Hub.-Mor., Onobrychis argyrea Boiss. subsp. argyrea, Onobrychis galegifolia Boiss. and Onobrychis tournefortii (Willd.) Desv. species to HCT-116 cells were investigated. With cytotoxicity analysis that with these species inhibitor concentrations (IC50) which resulted in a 50% reduction in the proliferation of HCT-116 cells were identified. In continuation of the study; the antiproliferative and apoptotic effects of Onobrychis albiflora extract on HCT-116 cells were evaluated by Caspase 3, Annexin V / PI Apoptosis / Necrosis analysis, Apopxin Green and 7-AAD Apoptosis / Necrosis analysis.     Management of Foreign Body Aspiration in Children: Insights From 664 Cases in the Operating Room

    Tracheal or bronchial foreign body aspiration is an important emergency o childhood requiring bronchoscopy under general anesthesia. In this retrospective study, bronchoscopy were performed for foreign body aspiration; anesthesia methods, risks, and postoperative complications were evaluated. Children who underwent bronchoscopy with suspicion of a foreign body in the pediatric surgery operating room between January 2010 and December 2021 were included in the study. Six hundred and sixty four children were evaluated. General anesthesia was applied to all bronchoscopy. Demographic characteristics of the patients, type of foreign body, localization of foreign body, distribution of foreign body according to age groups, complications related to anesthesia and surgical process, length of stay in the postoperative intensive care unit, and duration of hospital stay were evaluated. The patients who aspirated foreign bodies most frequently were children aged 1-3 years (73.6%). During bronchoscopy, organic matter (seeds, hazelnuts, peanuts, chickpeas) was the most frequently removed foreign body in children (559 patients). It was observed that foreign bodies were mostly located in the right main bronchus at a rate of 52.4%, the left main bronchus at 28%. The most common anesthesia-related complications were desaturation in 400 patients, hypercarbia in 200 patients, bronchospasm in 108 patients, and respiratory arrest in 2 patients. Many problems may be encountered in foreign body inhalation, and bronchoscopy of these patients should be performed by an experienced anesthesiologist and surgeon at the right time and in the safest way in terms of anesthesia and surgery

    Effectiveness of dialectical behavior therapy-online group skills training during the pandemic: A pilot study

    Dialectical Behavior Therapy (DBT) was developed by Linehan for clients with borderline personality disorder. It is based on behaviorist approach, Zen Buddhism, and dialectical philosophy. There are four components: individual session, group skills training, telephone coaching, and consultation team. DBT group skill training consists of four modules in total, including mindfulness, interpersonal effectiveness, stress tolerance, and emotion regulation skills. Many studies have been conducted on the effectiveness of DBT and DBT group skills training. In this study, the effectiveness of 8-week DBT group skill training in university students on depression, anxiety, and stress levels during the pandemic was investigated. In addition, the improvements in emotion regulation, mindfulness, and interpersonal effectiveness skills were observed. A total of 17 women, who were undergraduate students between the ages of 18-24, participated in the study. An online self-evaluation form was sent to the participants and feedback was given to the applicants by phone call. Sociodemographic Information Form, the Five Facet Mindfulness Questionnaire-Short Form, Difficulties in Emotion Regulation Scale-Brief Form, Interpersonal Competence Questionnaire-Short Form, and Depression Anxiety Stress Scale were sent online to the selected participants before participating in the group skill training, after the 4th session, and at the end of the 8th week. According to the results, DBT group skill training was found to be helpful in reducing depression, anxiety, and stress levels.     Bu projede, organik ürün olarak, talebi oldukça fazla Sultani Çekirdeksiz kuru üzüm üretimi ele alınmıştır. Çalışma, iki alt projeden oluşturulmuştur. I. Alt projede organik tarımda istenen minimum toprak işlemesi yanında ülkemizde bulunan ve birçok olumlu özelliklere sahip Zeolit (Klinoptilolitie mineral [Agro-clıno (NMF 900)]) kullanımı da denemede yer almıştır. II. Alt proje de ise Ege Bölgesi bağlarının ana hastalığı olan külleme mücadelesinde NaHCO3’ (içme sodasının) mevcut koşullardaki kullanım olanakları projede ele alınmıştır Bağlardaki ana zararlı da salkım güvesi olup her iki alt projede, salkım güvesi mücadelesinde tuzaklar asılarak zararlının ergin populasyonu bu tuzaklarla ilk ergin çıkışından uçuşlar sona erinceye kadar haftalık olarak izlenmiştir. Uçuş eğrileri ile tahmin-uyarı istasyonlarından elde edilen meteorolojik veriler ışığında gerekli uygulamalar yapılmıştır. Çalışmanın sonucunda, KULTIVATOR ile toprak işleme sisteminin toprağa olumsuz etkisinin de en az düzeyde olduğu da dikkate alınarak önerilebileceği ortaya çıkmıştır. Yeşilyurt yöresi topraklarındaki mikrobiyal aktiviteyi arttırmada malç uygulaması önerilmesine karşın, bu uygulama diğer disiplinler tarafından uygun görülmediğinden alternatif olarak KULTIVATOR uygulaması tercih edilmektedir. Kükürt ve içme sodası uygulanan parsellerde külleme hastalık şiddeti konvansiyonel olan şahit parsele göre istatistiki öneme sahip düzeyde azalma kaydetmiştir. Konvansiyonel koşullardaki hastalığın daha fazla görülmesini uygun koşulları yaratacak yoğun vejetatif gelişmenin yaratmış olduğu düşünülmektedir. Kükürdün zararlıları baskılayıcı etkisi de bilindiğinden organik yetiştiricilikte Külleme mücadelesinde içme sodasına nazaran özellikle kükürdün tercih edilmesi uygun bulunmaktadır. Denemenin başladığı 2003 yılı baz alındığında Zeolit (Klinoptilolitie mineral [Agro-clıno (NMF 900)]) uygulamalarında verim ve kalite kriterlerinde artış olduğu tespit edilmiştir. Toprak işleme uygulamalarından toprağı çizerek işleme salkım sayısı ve verim bakımından önemli bulunurken pulluk+ diskharow ile işleme salkım ağırlığını; malç uygulamasının ise kuru maddeyi ve tane tutumunu arttırdığı görülmektedir. Yaprak alma uygulamalarından, 6 adet yaprak alınması kontrole nazaran verim, salkım sayısı ve salkım ağırlığını nispeten arttırmıştır. Proje konusu çalışma birimleri itibarıyla değerlendirildiğinde toprağın çizilerek işlenmesi (KULTIVATOR sistemi) ön plana çıkmaktadır

    Laryngoscopic Examination During the COVID-19 Pandemic: Turkish Voice Speech and Swallowing Disorders Society and Turkish Professional Voice Society Recommendations

    COVID-19 is highly transmissible and spreads rapidly in the population. This increases the occupational risk for health care workers. In otolaryngology clinic practice, patients with upper respiratory tract infection symptoms are common. Also, routine head and neck examinations such as oral cavity examination, nasal/nasopharyngeal examination, or video laryngostroboscopic evaluation are highly risky because of the aerosol formation. To emphasize this issue, two leading otolaryngology organizations in Turkey; 'Voice Speech and Swallowing Disorders Society', and 'Professional Voice Society' gathered a task force. This task force aimed to prepare a consensus report that would provide practical recommendations of the safety measurements during routine clinical care of laryngology patients.     The purpose of this convergent parallel mixed methods design study was to conduct a post-use evaluation of the coursebook New Language Leader Intermediate from the perspectives of students and instructors at a language preparatory school of a public university in Turkey. 202 students provided quantitative data about various aspects of the coursebook through a researcher-developed survey which also contained open-ended questions about the coursebook. 20 instructors completed the instructors’ version of the same instrument which had some additional sections. The instructors’ survey also had some open-ended questions which were used to collect qualitative data. In addition to the survey instrument, semi-structured interview protocols were utilized with students and instructors. Qualitative data were collected to find out what strengths and weaknesses participants attribute to the coursebook and what recommendations they make to increase the effectiveness of the coursebook. The quantitative and qualitative data were analyzed separately, and the combined results were interpreted in the discussion chapter.Thesis (M.S.) -- Graduate School of Social Sciences.     Yüksek Lisans TeziBu araştırma Konya ili Merkez ilçelerinde resmi ilkokul, ortaokul ve liselerde görev yapan öğretmenlerin okul müdürlerinden bekledikleri ve karşılaştıkları etik davranışların neler olduğunu ve öğretmenlerin okul müdürlerinin etik davranışlarına verdikleri tepkileri belirlemeyi amaçlamaktadır. Nitel bir araştırma olan çalışmadaki veriler, Konya il merkezinde bulunan resmi ilkokul, ortaokul ve liselerden; üç sınıf öğretmeni, yedi ortaokul branş öğretmeni ve üç lise branş öğretmeni olmak üzere toplam 13 öğretmen ile yapılan yarı yapılandırılmış iki odak grup görüşmesinden elde edilmiştir. Araştırma verileri, içerik analizi tekniği kullanılarak çözümlenmiştir. The data in the study, which is a qualitative research, was obtained from two semi-structured focus group interviews with three primary school teachers, seven secondary school branch teachers and three high school branch teachers in Konya city center. Research data were analyzed using content analysis technique. In line with the findings obtained, the following results were reached in the study: ‘’Non-Discrimination’’, ‘’Openness’’, ‘’Participation in decisions’’, ‘’Have a vision’’, ‘’Academic emphasis’’, ‘’Reliability ‘’, ‘’Competence ‘’, ‘’Respect’’ categories were determined under the theme of ‘’Ethical Behaviors Expected by Teachers from School Principals’’. The categories of ‘’Discrimination’’, ‘’Openness’’, ‘’Participation in decisions’’, ‘’Have a vision’’, ‘’Academic emphasis’’, ‘’Reliability’’, ‘’Competence’’, ‘’Respect’’ were determined under the theme of ‘’Ethical Behaviors Encountered by Teachers in School Principals’’. It has been determined that teachers are faced with both ethical and unethical principal behavior in these categories. The categories of ‘’Discrimination’’, ‘’Openness’’, ‘’Participation in decisions’’, ‘’Have a vision’’, ‘’Academic emphasis’’, ‘’Reliability’’, ‘’Competence’’, ‘’Respect’’ were determined under the theme of ‘’The reactions of Teachers Against School Principals Ethical Behaviors’’. Suggestions were made for ‘’Practice’’ and ‘’Researchers’’ regarding the solution of unethical problems faced by teachers about the ethical behaviors that teachers expect and encounter from school principals
