Proof-of-Principle Study Suggesting Potential Anti-Inflammatory Activity of Butyrate and Propionate in Periodontal Cells.
Short-chain fatty acids (SCFAs) are potent immune modulators present in the gingival crevicular fluid. It is therefore likely that SCFAs exert a role in periodontal health and disease. To better understand how SCFAs can module inflammation, we screened acetic acid, propionic acid, and butyric acid for their potential ability to lower the inflammatory response of macrophages, gingival fibroblasts, and oral epithelial cells in vitro. To this end, RAW 264.7 and primary macrophages were exposed to LPSs from Porphyromonas gingivalis (P. gingivalis) with and without the SCFAs. Moreover, gingival fibroblasts and HSC2 oral epithelial cells were exposed to IL1β and TNFα with and without the SCFAs. We report here that butyrate was effective in reducing the lipopolysaccharide (LPS)-induced expression of IL6 and chemokine (C-X-C motif) ligand 2 (CXCL2) in the RAW 264.7 and primary macrophages. Butyrate also reduced the IL1β and TNFα-induced expression of IL8, chemokine (C-X-C motif) ligand 1 (CXCL1), and CXCL2 in gingival fibroblasts. Likewise, butyrate lowered the induced expression of CXCL1 and CXCL2, but not IL8, in HSC2 cells. Butyrate further caused a reduction of p65 nuclear translocation in RAW 264.7 macrophages, gingival fibroblasts, and HSC2 cells. Propionate and acetate partially lowered the inflammatory response in vitro but did not reach the level of significance. These findings suggest that not only macrophages, but also gingival fibroblasts and oral epithelial cells are susceptive to the anti-inflammatory activity of butyrate
Rank Protein Immunolabeling during Bone-Implant Interface Healing Process
The purpose of this paper was to evaluate the expression of RANK protein during bone-healing process around machined surface implants. Twenty male Wistar rats, 90 days old, after having had a 2 mm diameter and 6 mm long implant inserted in their right tibias, were evaluated at 7, 14, 21, and 42 days after healing. After obtaining the histological samples, slides were subjected to RANK immunostaining reaction. Results were quantitatively evaluated. Results. Immunolabeling analysis showed expressions of RANK in osteoclast and osteoblast lineage cells. The statistical analysis showed an increase in the expression of RANK in osteoblasts at 7 postoperative days and a gradual decrease during the chronology of the healing process demonstrated by mild cellular activity in the final stage (P < .05). Conclusion. RANK immunolabeling was observed especially in osteoclast and osteoblast cells in primary bone during the initial periods of bone-healing/implant interface
O MÚLTIPLO E O DIVERSO EM "VÍCIOS E VIRTUDES"
The Helder Macedo's narrative presents us the writer who meets a friend, also writer, and listens from him the story of his loves. The text inside the text leads us, in a spiral movement, from the author to the character, from the present to the past, from the contingent to the history. The present work aims to reflect on the games of masks formulated by Macedo from the overlapping of registers and narrative plans that is presented in his novel.
Na comparação estatística dos valores obtidos...Purpose: The aim of this study was to compare the biologic and mechanical behavior of bone tissue around implants with commercially available surfaces, modified by acid etching (SA), and machined surfaces (SU), with the experimental surfaces modified by laser (SL), and modified by laser with silica deposition (SS), using biomechanical, topographic and histometric analyses. Materials and Methods: Thirty rabbits received 120 implants measuring 3,75x10mm in their right and left tibias, being two implants of each type of surface per tibia. The implant located in the uppermost position, inserted in the tibial metaphysis, was used for the biomechanical test, described in Chapter I; and the implant located below this, inserted in the tibial epiphysis, was used for the histometric study, described in Chapter II. The morphology of the implant surfaces was analyzed by scanning electron microscopy (SEM) and by energy dispersive X-ray spectrometry (EDS) before and after 4 weeks of implantation and removal by reverse torque. Mean roughness and cross-sectional measurements were obtained. After periods of 4, 8, and 12 weeks the biomechanical test was performed, and after this the animals were euthanized. The parts of the implant located in the tibial epiphysis were taken for cutting nondecalcified on a microtome. Histometric analysis was performed using an optical microscope. The bone interface contact (BIC), and the bone area (AO) were measured in percentages. A slide of each surface in the period of 4 weeks was metalized in order to analyze the element mapping by means of SEM-EDS. Results: The topographic characterization showed differences between the analyzed surfaces, and the mean roughness values of SL and SS were statistically higher (p<0,05) than those of SA. In the statistical comparison of the values obtained by the biomechanical test, it was... Management of accidental swallowing in implant dentistry
This report describes a protocol for managing the accidental swallowing of dental instruments in implant dentistry, illustrated by a patient who accidentally swallowed a hexagonal wrench. The first step was to refer the patient to the medical emergency hospital service for radiographic and clinical evaluation. The hexagonal wrench was located in the stomach and was immediately removed with an endoscopic procedure. The gastric mucosa was sampled via biopsy and the sample submitted to the urease test, which was positive for Helicobacter pylori. Triple treatment was instituted for gastritis caused by H pylori to avoid exposing the patient to unnecessary risk. Removal of a foreign body by means of an endoscopic procedure constitutes a safe and effective treatment
