FUS-CHOP promotes invasion in myxoid liposarcoma through a SRC/FAK/RHO/ROCK-dependent pathway

Deregulated SRC/FAK signaling leads to enhanced migration and invasion in many types of tumors. In myxoid and round cell liposarcoma (MRCLS), an adipocytic tumor characterized by the expression of the fusion oncogene FUS-CHOP, SRC have been found as one of the most activated kinases. Here we used a cell-of-origin model of MRCLS and an MRCLS cell line to thoroughly characterize the mechanisms of cell invasion induced by FUS-CHOP using in vitro (3D spheroid invasion assays) and in vivo (chicken chorioallantoic membrane model) approaches. FUS-CHOP expression activated SRC-FAK signaling and increased the invasive ability of MRCLS cells. In addition, FAK expression was found to significantly correlate with tumor aggressiveness in sarcoma patient samples. The involvement of SRC/FAK activation in FUS-CHOP–mediated invasion was further confirmed using the SRC inhibitor dasatinib, the specific FAK inhibitor PF-573228, and FAK siRNA. Notably, dasatinib and PF573228 could also efficiently block the invasion of cancer stem cell subpopulations. Downstream of SRC/FAK signaling, we found that FUS-CHOP expression increases the levels of the RHO/ROCK downstream effector phospho-MLC2 (T18/S19) and that this activation was prevented by dasatinib or PF573228. Moreover, the ROCK inhibitor RKI-1447 was able to completely abolish invasion in FUS-CHOP–expressing cells. Child-parent Interactions in American and Turkish Families: Examining Measurement Invariance Analysis of Child-parent Relationship Scale

The parent-child relationship is a cornerstone of early childhood development and one-way early childhood programs can have a positive influence on early development is to adopt programmatic features to enhance this relationship. Research supports these conclusions in both U.S. and cross-cultural contexts, even though assumptions about parenting and the parent-child relationship may differ across cultures. However, for true understanding of cultural differences, it is important to have comparable measures across cultures. The purpose of the study is to assess measurement invariance of the two constructs of the Child-parent Relationship Scale using data gathered in programs serving low-income preschool children in the U.S.(n = 4,450) and Turkey (n = 592) from 2014 to 2015. Using Single-group Confirmatory Factor Analysis, the original factor structures of the Turkish and the English versions were tested. Besides, Multigroup Confirmatory Factor Analysis provided evidence for configural, metric, scalar invariance, strict factorial invariance or error variance invariance and construct level invariance across the two versions. Only configural invariance was established, which showed an agreement for the existence of an underlying theoretical construct for each subscale (Conflict and Closeness) of the Turkish and the English versions. However, item CPRS 4 was a non-significant item for Conflict in the Turkish version that affected the possibility to conduct further analyses. Findings encourage researchers to propose and assess cultural and linguistic adaptations for the Child-parent Relationship Scale before cross-cultural comparisons related to family relationships

ANALYSIS OF GENDER RELATIONS IN THE INDUSTRIAL COMMUNITY OF AGUIRRE, PUERTO RICO

This thesis is a study of the gender relations of the residents of Aguirre, Puerto Rico, between 1940 and 1991. The primary goal of the project was to explore how gender roles and relations in the Aguirre community were impacted by the social class system introduced by the Aguirre Sugar Company. This project was based on the interpretation of the past and present situation of the Aguirre community using oral history, by conducting a series of interviews among its residents. The interviews resulted in three main themes. First, the concepts of `normal and natural\u27 were used to distinguish gender roles. Second, Aguirreños identified `family as community\u27, since through the family individuals built their gender identity and learned the basic rules of coexistence within the social hierarchy of the community. Third, although the gender and class roles were clear in the community, `resistance and negotiation\u27 occurred in the home and at the Company between those of different gender and social classes. The Aguirre Sugar Company was one of the principal influences on the construction of the Aguirreños identity, and left a mark on the past, present and future generations

Towards a Sustainable FTAA: DOes Latin America Meet the Necessary Financial Preconditions?

This paper focuses on identifying preconditions that will ensure the sustainability of a Free Trade Area of the Americas (FTAA). It argues that the macro, micro, and political conditions advanced in the literature to measure a country's ability to compete internationally, while necessary, are not sufficient to ensure the success and permanence of a free trade agreement. Instead, two additional financial conditions are needed. The first is that each partner in the free trade area needs to have sustainable public debts as determined by the achievement of credible and sustainable structural fiscal balances. The second is that exchange rate regimes across trading partners should be compatible in the sense that adverse shocks in one country do not generate a policy dilemma in other partners between abandoning their exchange rate system or the free trade area. A preliminary analysis of the evidence in the Latin American and Caribbean region shows the importance of these two preconditions. An analysis of debt sustainability reveals that there are a number of countries in the region that need to deal with potential solvency problems before reaching the status of credible partners in a regional trade arrangement. Argentina is already deemed insolvent, and countries such as Ecuador and Venezuela rank high on the list of countries where the issue of debt sustainability can become a serious problem. Not resolving this before reaching a regional trade agreement can threaten its long-term stability. The examination of the compatibility of exchange rate systems across trading partners is also very revealing. Part of the success of NAFTA since the late 1990s and the "impasse" of Mercosur during 1999-2001 had to do with the choices of exchange rate regimes. In both trade areas the share of trade among the partners is very high, and in NAFTA, this includes significant financial transactions. While Mexico was able to use the flexibility of the exchange rate to improve competitiveness following the sharp decline of portfolio flows from US investors into Mexico following the Asian and Russian crises, Argentina had no mechanisms to deal with an adverse shock from Brazil (such as a depreciation of the real in 1999). From this perspective, the recent move of Argentina towards a more flexible exchange rate system is good news for a sustainable free trade area.
