    Exploring the utility of Brachypodium distachyon as a model pathosystem for the wheat pathogen Zymoseptoria tritici

    peer-reviewedBackground Zymoseptoria tritici, the causative organism of Septoria tritici blotch disease is a prevalent biotic stressor of wheat production, exerting substantial economic constraints on farmers, requiring intensive chemical control to protect yields. A hemibiotrophic pathogen with a long asymptomless phase of up to 11 days post inoculation (dpi) before a rapid switch to necrotrophy; a deficit exists in our understanding of the events occurring within the host during the two phases of infection. Brachypodium distachyon has demonstrated its potential as a model species for the investigation of fungal disease resistance in cereal and grass species. The aim of this study was to assess the physical interaction between Z. tritici (strain IPO323) and B. distachyon and examine its potential as a model pathosystem for Z. tritici. Results Septoria tritici blotch symptoms developed on the wheat cultivar Riband from 12 dpi with pycnidial formation abundant by 20 dpi. Symptoms on B. distachyon ecotype Bd21-1 were visible from 1 dpi: characteristic pale, water soaked lesions which developed into blotch-like lesions by 4 dpi. These lesions then became necrotic with chlorotic regions expanding up to 7 dpi. Sporulation on B. distachyon tissues was not observed and no evidence of fungal penetration could be obtained, indicating that Z. tritici was unable to complete its life cycle within B. distachyon ecotypes. However, observation of host responses to the Z. tritici strain IPO323 in five B. distachyon ecotypes revealed a variation in resistance responses, ranging from immunity to a chlorotic/necrotic phenotype. Conclusions The observed interactions suggest that B. distachyon is an incompatible host for Z. tritici infection, with STB symptom development on B. distachyon comparable to that observed during the early infection stages on the natural host, wheat. However first visible symptoms occurred more rapidly on B. distachyon; from 1 dpi in comparison to 12 dpi in wheat. Consequently, we propose that the interaction between B. distachyon and Z. tritici as observed in this study could serve as a suitable model pathosystem with which to investigate mechanisms underpinning an incompatible host response to Z. tritici.Teagasc Walsh Fellowship Programm

    Response to Teladorsagia circumcincta infection in Scottish Blackface lambs with divergent phenotypes for nematode resistance

    peer-reviewedThe objective of this study was to identify Scottish Blackface lambs that were at the extremes of the spectrum of resistance to gastrointestinal nematodes and characterise their response to an experimental nematode challenge. Lambs (n = 90) were monitored for faecal egg count (FEC) (2 samples from each of 2 independent natural infections). The most resistant (n = 10) and susceptible (n = 10) individuals were selected and challenged with 30,000 Teladorsagia circumcincta larvae (L3) at 9 months of age. Response to infection was monitored by measuring FEC, plasma pepsinogen, serum antibodies against nematode larval antigens and haematology profile, until necropsy at 71 days post infection. Worm burden, worm fecundity and the level of anti-nematode antibodies in abomasal mucosa were determined at necropsy. FEC was consistently higher in susceptible animals (P < 0.05), validating the selection method. Worm fecundity was significantly reduced in resistant animals (P = 0.03). There was also a significant correlation (r = 0.88; P < 0.001) between the number of adult worms and FEC at slaughter. There was no effect of phenotype (resistance/susceptibility) on plasma pepsinogen or on haematology profile. Phenotype had a significant effect on the level of anti-nematode IgA antibodies in serum (P < 0.01), reflecting a higher peak in resistant animals at day 7 post infection. It is concluded that significant variation in the response to gastrointestinal nematode challenge exists within the Scottish Blackface population with resistant animals displaying significantly lower FEC, lower worm fecundity and higher concentration of anti-nematode IgA antibodies in serum.Kathryn McRae was supported by a Teagasc Walsh fellowship and the Allan and Grace Kay Overseas Scholarship

    The climatic impact of food consumption in a representative sample of Irish adults and implications for food and nutrition policy

    peer-reviewedTo evaluate the greenhouse gas emissions (GHGE) associated with the diet of Irish adults. GHGE were estimated by applying conversion factors to habitual food consumption data taken from the National Adult Nutrition Survey, which was representative of the population. Descriptive analyses were undertaken for GHGE for the total population, as well as accounting for energy misreporting and across categories of sociodemographic and socio-economic factors and tertiles of emissions. Republic of Ireland. Adults aged 18–87 years (n 1500). The GHGE derived from daily dietary intakes was estimated as 6·5 kg of CO2 equivalents (CO2eq) per person. Males, younger consumers, those with secondary education and student employment status were associated with significantly higher GHGE. Red meat was the highest contributor to GHGE with 1646 g CO2eq arising from a mean intake of 47 g/d. Dairy and starchy staples were the next largest dietary GHGE sources, with mean daily emissions of 732 g CO2eq and 647 g CO2eq, respectively. The lowest emissions were associated with consumption of vegetables, fruits and legumes/pulses/nuts. Based on profiling using actual food consumption data, it is evident that one single measure is not sufficient and a range of evidence-based mitigation measures with potential to lower emissions throughout the food chain should be considered.     Implementing biosecurity measures on dairy farms in Ireland

    peer-reviewedDairy farms in Ireland are expanding in preparation for a new era of unrestricted milk production with the elimination of the European Union (EU) production quotas in 2015. Countries experiencing a changing agricultural demographic, including farm expansion, can benefit from documenting the implementation of on-farm biosecurity. The objectives of this study were to document and describe influences on biosecurity practices and related opinions on dairy farms. A representative response rate of 64% was achieved to a nationwide telesurvey of farmers. A 20% discrepancy was found between self-declared and truly ‘closed’ herds indicating a lack of understanding of the closed herd concept. Although >72% of farmers surveyed considered biosecurity to be important, 53% stated that a lack of information might prevent them from improving their biosecurity. Logistic regression highlighted regional, age, and farm-size related differences in biosecurity practices and opinions towards its implementation. Farmers in the most dairy cattle dense region were three times more likely to quarantine purchased stock than were their equivalents in regions where dairy production was less intense (P = 0.012). Younger farmers in general were over twice as likely as middle-aged farmers to implement biosecurity guidelines (P = 0.026). The owners of large enterprises were almost five times more likely to join a voluntary animal health scheme (P = 0.003), and were over three times more likely to pay a premium price for health accredited animals (P = 0.02) than were those farming small holdings. The baseline data recorded in this survey will form the basis for more detailed sociological and demographic research which will facilitate the targeting of future training of the farming community in biosecurity

    Using Information from Mid Term Evaluations of RDP for the Multicriteria Analysis of Agri-environmental Schemes

    This paper discusses how environmental indicators and multicriteria methodologies can support the ex post evaluation of Agri-Environmental Schemes. The paper is based on information from the Mid term evaluation of the Rural Development Programmes and develops around an example that compares Ireland and Emilia-Romagna. The results show that the application of Agri-Environmental Schemes only partially achieves to local objectives, and the way in which the Agri-Environmental Schemes are implemented can be reasonably improved. However, the tentative analysis emphasizes the scarcity of quantitative data that can be related to effectiveness, the lack of predetermined quantitatively defined target levels of objectives and the difficulty to assess the relative importance of different evaluation criteria.     Impact of Livestock on Hill Environment

    End of Project ReportThe hill and mountain landscape of western Ireland is an area of high scenic value as well as contributing to the livelihood of local farmers mainly through extensive grazing. Measures that were intended to support lamb prices led to increased ewe numbers. Public concern attributed this increase to an apparent deterioration in the status of the semi-natural vegetation and to an increase in the rate of soil erosion. In the absence of an existing databank, Teagasc undertook a research programme to quantify the impact of hill sheep on the semi-natural vegetation, the progress of soil erosion and changes in certain weather elements e.g. ‘driving rain’. A detailed database of the physical background (physiography and soils) of the Teagasc hill sheep farm, Leenaun, was compiled on the basis of a grid, 100 m x 100 m. Changes in the frequency of vegetation on the unimproved hill were monitored, 1995-2002, by point quadrat. Ten permanent exclosures representing the main vegetation types and a controlled grazing experiment on the upper steep slope were established. Sequential aerial photography (1973/7-1996/8) for selected sites in western counties and wind and rain data (1957-2000) for five western synoptic stations were obtained. The classification of the peatland and heathland habitats was considered. Analysis of data highlighted a range of issues among which were the physical complexity and fragility of the landscape, the sustainability of the hill sheep system on the Teagasc hill sheep farm, the beneficial effects of controlled grazing, the progressive erosion of hill soils and an increase in high intensity levels of ‘driving rain’. The classification of the habitats, peatland and heathland, should encompass other forms of biodiversity as well as flora. The sustainable use of this landscape requires a deep knowledge of the of the grazing management system, which increasingly involves part-time farming, and its impact on the soils and semi-natural vegetation

It is particularly prevalent as the primary silage-making system on both beef farms and smaller-sized farms. However, it is also widespread as a second silage-making system on many other farms.     Preliminary study on the use of near infrared hyperspectral imaging for quantitation and localisation of total glucosinolates in freeze-dried broccoli

    peer-reviewedThe use of hyperspectral imaging to (a) quantify and (b) localise total glucosinolates in florets of a single broccoli species has been examined. Two different spectral regions (vis–NIR and NIR), a number of spectral pre-treatments and different mask development strategies were studied to develop the quantitative models. These models were then applied to freeze-dried slices of broccoli to identify regions within individual florets which were rich in glucosinolates. The procedure demonstrates potential for the quantitative screening and localisation of total glucosinolates in broccoli using the 950–1650 nm wavelength range.
