Vascular Architecture of the Lactating and Non-Lactating Teat of the Bitch: A Scanning Electron- and Light Microscope Study
Tissues from fourteen mammary glands of eight bitches were prepared for scanning electron microscopy of vascular corrosion casts and for histology to study the vasculature of the lactating and non-lactating teats. The densely-meshed mammary dermal capillary network formed ridges and troughs. The teat ducts were vascularized by a relatively densely-meshed capillary network which drained into veins longitudinally oriented to the ducts. Between eight and fifteen teat duct openings were seen on the tip of the teat, that were sometimes divided by a septum. The inner vascularization of the teat showed that the main papillary arteries divided into undulating secondary papillary arteries which presented numerous semi-constrictions and loops. Their structure may help during erection of the teat. Arteriovenous anastomoses found at different points may participate in blood flow maintenance during suckling, heat regulation and teat erection. Veins freely anastomosed and ran longitudinally to the axis of the teat. They exhibited numerous bicuspid valves. In non-lactating teats, vessels showed the same main architecture and characteristics mentioned above, although these were considerably less marked. The structure of the vascular elements in the teat of the bitch could favor blood flow during suckling and suggest that vessels adapt to the physiological situation

LiteraturverzeichnisIn einer negativ kontrollierten Studie (Studienteil I) wurde der Einfluss des
Vordippens der Zitzen mit einem schaumbildenden, jodophoren Präparat (0,27%ig)
auf die Entwicklung des Jodgehaltes der Milch von Kühen untersucht.
Laktierende Kühe wurden an 12 aufeinanderfolgenden Tagen täglich wechselnd mit
einem schaumbildenden jodhaltigen Dippmittel vorgedippt und konventionell
vorgereinigt. Die Tiere waren so ihre eigene Negativkontrolle. Die Bestimmung
des Jodgehaltes der Einzelgemelke erfolgte mittels einer ionenselektiven
Sonde. Der Jodgehalt der Milch an den Dipptagen betrug 243 ± 51 ppb, während
dieser in der Milch der Kontrolltage bei 212,74 ± 37,5 ppb lag (p<0,01).
Desweiteren beeinflusste das Tier den Jodgehalt der Milch (p<0,001).
Desweiteren bestand eine negative Korrelation der Jodkonzentration der Milch
zur Milchleistung (p<0,001). Der Jodgehalt der Milch lag aber bei der
Anwendung eines Vordippverfahrens mit einem jodophoren Präparat im
physiologischen Bereich.
In einer positiv kontrollierten Feldstudie (Studienteil II) wurde die
Wirksamkeit eines Nachdippverfahrens mittels eines automatischen Sprühbalkens
hinsichtlich der Inzidenz intramammärer Neuinfektionen, der Mastitisinzidenz,
der Zellzahlentwicklung und des Zustandes des Drüsengewebes und der Zitze
untersucht. Die Studie wurde als Tiergruppenvergleich durchgeführt, wobei in
der Versuchsgruppe (126 Tiere) das Zitzendippmittel nach dem Melken
automatisch durch den Sprühbalken appliziert wurde, während die Zitzen in der
Kontrollgruppe (137 Tiere) von Hand gedippt wurden.
Die Inzidenz intramammärer Neuinfektionen unterschied sich bei der Anwendung
des automatischen Sprühverfahrens nicht statistisch absicherbar von der in der
Kontrollgruppe. Die Inzidenz von Neuinfektionen durch KNS war in der
Versuchsgruppe höher (VV: 6,9% vs. KV: 3,5%, p<0,05). Die Mastitisinzidenz war
in beiden Gruppen vergleichbar (0,351 Erkrankungen pro 100 Dipptage bei den
Versuchstieren vs. 0,370 bei den Kontrolltieren). Hinsichtlich der Entwicklung
des Zellgehaltes der Viertelgemelke und des Gesamtgemelkes waren beide Gruppen
vergleichbar. Bei den Versuchstieren traten bei den Untersuchungen des Euters
im Studienverlauf höhere Anteile von Zitzenhautrissen auf (p<0,05).
In einer weiteren positiv kontrollierten Feldstudie (Studienteil III) wurde
die Wirksamkeit eines Nachdippverfahrens mit einem neuartigen,
schaumbildenden, jodophoren Dippmittel hinsichtlich der Inzidenz intramammärer
Neuinfektionen, der Mastitisinzidenz, der Zellzahlentwicklung und des
Zustandes des Drüsengewebes und der Zitze untersucht. Die Studie wurde als
Tiergruppenvergleich durchgeführt, wobei in der Versuchsgruppe (122 Tiere) das
schaumbildende Zitzendippmittel zum Nachdippen eingesetzt wurde. Die Zitzen in
der Kontrollgruppe (121 Tiere) wurden mit einem konventionellen jodophoren
Produkt mit gleicher Gebrauchskonzentration der desinfizierenden Komponente
versorgt.
Die Inzidenz intramammärer Neuinfektionen war in beiden Gruppen vergleichbar.
Das galt auch für die Inzidenz klinischer Mastitiden (0,6382 Erkrankungen pro
100 Dipptage bei den Versuchstieren vs. 0,5005 bei den Kontrolltieren).
Hinsichtlich der Entwicklung des Zellgehaltes der Viertelgemelke und des
Gesamtgemelkes waren beide Gruppen vergleichbar. Die Entwicklung des Zustandes
der Zitzenhaut und der Strichkanalöffnung unterlag während des Versuches
starken Schwankungen.
In einer negativ kontrollierten Feldstudie (Studienteil IV) wurde die
Effizienz eines Vordippverfahrens mit einem neuartigen, schaumbildenden,
jodophoren Dippmittel geprüft. Die Parameter zur Einschätzung der Wirksamkeit
waren dieselben wie in den Studienteilen II und III. Die Untersuchung wurde
als Viertelvergleich im Split-Udder-Design durchgeführt. Die rechten Zitzen
wurden vor jedem Melken mit einem schaumbildenden, jodophoren Dippmittel
vorgedippt, während die linken Zitzen als unbehandelte Kontrolle dienten.
Diese wurden konventionell mit einem feuchten individuellen Euterlappen und
trockenem Einmaleuterpapier gereinigt.
Beim Vergleich der Inzidenz intramammärer Neuinfektionen über den
Versuchszeitraum (6,6% bei den gedippten Vierteln vs. 6,95% bei den
Kontrollvierteln), der Inzidenz klinischer Mastitiden (0,0575 Erkrankungen pro
100 Dipptage in gedippten Vierteln vs. 0,0748 in Kontrollviertel) und dem
Mittel der Zellgehalte der Viertelgemelksproben bestand kein statistisch
absicherbarer Unterschied zwischen den Behandlungsgruppen. Das Spektrum der
nachgewiesenen Erreger der beiden Versuchsgruppen war vergleichbar. Der
Zustand des Euterdrüsengewebes und der Zustand der Strichkanalöffnung
entwickelte sich in Versuchs- und Kontrollvierteln vergleichbar, wobei der
Zustand des Drüsengewebes bei den Kontrollvierteln mit Ausnahme der
Abschlussuntersuchung zu allen Untersuchungszeitpunkten geringfügig schlechter
war.A negative controlled field study (part I) was conducted to determine
potential iodine residues in milk after predipping with a foaming teat
disinfectant containing .27 % polyvidone iodine. The teats of 9 lactating
dairy cows were alternately predipped and conventionally cleaned over a period
of 12 days. Due to this study design the animals were there own negative
controls. An iodine selective electrode was used to measure the iodine
concentration in milk of all bucket milk samples in the study period. Iodine
concentration of milk was 243.7 ± 51 ppb on days with predipping and 212.7 ±
37.5 ppb on control days, respectively (p<.001). Iodine concentration was
influenced by the individual cows (p<.001) and was negatively correlated with
milk yield. Iodine concentration was elevated by predipping with a iodophore
depending on milk production. However, iodine concentrations remain within the
physiological range of milk.
The effect of an automatic application of a postmilking teat dip regarding to
incidence of new intramammary infections (IMI), incidence of clinical
mastitis, the influence on somatic cell count (SCC) and the characteristics of
udder tissue and teat was investigated in a positive controlled field study
(part II). Teat dip was either applied automatically in the treatment group
(126 animals) or with a dip cub (control group; n= 137). The incidence of new
IMI did not differ on a statistical significant level. In the treatment group
it was 20% and in the control group (14.9%, p>.05). Incidence of new IMI
caused by CNS was higher in the treatment group (6.9%) than in the control
(3.5%, p<.05). Incidence of clinical mastitis did not differ between groups
(.351 cases per pro 100 days in the treatment group vs. .370 in the control
group). The development of SCC was comparable in both groups. Teats in the
treatment group showed a higher rate of lesions of skin on March 1st, 2000
(treatment group: 16.2% vs. control group: 11.2; p<.05), April 3rd , 2000
(23.1% vs. 7.2%; p<.05) and April 26th , 2000 (17.7 vs. 3.0%; p<.05).
The effect of postmilking teat dipping with a foaming iodophor agent regarding
to IMI, incidence of clinical mastitis, SCC and the characteristics of udder
tissue and teat was investigated in a further positivly controlled field study
(part III).
We compared two groups of animals. Teats were dipped with the new foaming
iodophor in the treatment group (122 animals) while teats in the control group
(121 animals) were dipped with a conventional iodophor teat dip with the same
iodine content. The incidence of new IMI and clinical mastitis did not differ
between the groups. We detected .6382 cases per pro 100 days in the treatment
group vs. .5005 in the control group. The development of SCC was comparable in
both groups.The teat skin an teat duct condition showed intense fluctuation
during study period.
In a fourth study we investigated the efficacy of premilking teat dipping with
a foaming iodophor teat dip in a negative controlled field study. We used the
same parameters as in part II and III to evaluate clinical efficacy of the
method. Predipping was compared with a negative control using a split udder
experimental design. Al teats were dipped after milking with the same dip.
There was no difference between treated and control quarters in incidence of
new IMI during study period (treated quarters: 6.6% vs. untreated: 6.95%),
incidence of clinical mastitis (.0575 cases per pro 100 days in the treatment
group vs. .0748 in the control group) and geometric mean of SCC of quarter
milk samples. Spectrum of detected pathogens was comparable. Condition of
udder tissue and teat duct did not differ between treated and control
quarters

SelbständigkeitserklärungEs wurden eine Studie, die aus zwei Studienteilen bestand, zur Wirksamkeit des
internen Zitzenversieglers OrbeSeal® durchgeführt. In dem ersten Studienteil A
wurde die klinische Wirksamkeit der zusätzlichen Applikation des internen
Zitzenversieglers OrbeSeal® zum konventionellen Antibiotikum (Orbenin extra®)
überprüft. Die Studie wurde im Splitted-Udder-Design durchgeführt. Jedem Tier
wurden je zwei Kontroll- und zwei Versuchsviertel zugeordnet. Die
Versuchsviertel (VV) wurden mit der Kombination aus Orbenin extra® und
OrbeSeal® behandelt, während die Kontrollviertel (KV) nur Orbenin extra®
erhielten. Es wurden im Zeitraum April bis Juli 2003 insgesamt 359 Tiere einer
kommerziell arbeitenden Milchviehanlage in Brandenburg in diese Studie
aufgenommen. Die Kontrollund Versuchsviertel waren in Hinblick auf die
Entwicklung der Zellzahl, des Drüsengewebsscores und der Hyperkeratose im
Bereich des Strichkanals während des Untersuchungszeitraums vergleichbar. Die
bakteriologischen Untersuchungen der Proben unmittelbar sowie fünf bis acht
Tage nach der Abkalbung weisen für die Versuchsviertel (VV =
Kombinationsbehandlung) einen höheren Anteil an bakteriologisch negativen
Ergebnissen auf als die Kontrollviertel (KV = nur Antibiotikum) (zum Zeitpunkt
der Abkalbung: KV 87,8 % vs. VV 91,0 % und zum Zeitpunkt 5 - 8 d p.p.: KV 82,1
% vs. VV 86,4 %). In den drei Auswertungszeiträumen für die Neuinfektionsraten
kamen in den Versuchsvierteln weniger Neuinfektionen vor als in den
Kontrollvierteln (Trockenstellen bis Abkalbung: KV 6,2 % vs. VV 4,6 %,
Trockenstellen bis 5 - 8 Tage p.p.: KV 16,1 % vs. VV 11,3 % und Abkalbung bis
5 - 8 Tage p.p.: KV 12,3 % vs. VV 10,5 %). In dem Zeitraum von der Abkalbung
bis zum Trockenstellen war die Wahrscheinlichkeit einer Neuinfektion für die
Versuchsviertel 27,1 % geringer als für die Kontrollviertel. Bei Eutervierteln
mit einer Prädisposition für eine intramammäre Infektion, wie hohe
Milchleistung vor dem Trockenstellen oder mittelgradig bis stark ausgeprägter
Hyperkeratose im Bereich des Strichkanals, konnte mit der zusätzlichen
Applikation von OrbeSeal® die Inzidenz von Neuinfektionen und klinischer
Mastitiden in den ersten 100 Tagen nach der Abkalbung reduziert werden. In dem
weiteren Studienteil B auf einem zweiten Betrieb in Brandenburg wurde eine
Kombinationsbehandlung aus OrbeSeal® und Orbenin extra® (Gruppe A) mit einer
ausschließlichen OrbeSeal® Behandlung (Gruppe B) und einem
viertelindividuellen Behandlungskonzept (Gruppe C) auf Basis des Ergebnisses
im California-Mastitis-Test (CMT-Test) verglichen. Der CMT-Test ist eine
Methode zur semiquantitativen Bestimmung des Milchzellgehaltes. Es wurden nur
Tiere aufgenommen, die einen arithmetischen Mittelwert des Zellgehaltes in den
letzten drei Milchleistungsprüfungen vor dem Trockenstellen von unter 200.000
Zellen/ml aufwiesen. Zusätzlich musste auf mindestens einem Euterviertel ein
Ergebnis von ++ (>600.000 Zellen/ml) oder mehr im CMT vorliegen. In der
viertelindividuell behandelten Gruppe wurden die Euterviertel mit einem
negativen oder einfach positiven CMT-Ergebnis mit OrbeSeal® behandelt. Viertel
mit zweifach oder mehr positiven CMT-Ergebnis wurden mit einer Kombination aus
Orbenin extra® und OrbeSeal® behandelt. Die Neuinfektionsraten waren für alle
drei Auswertungszeiträume in der Behandlungsgruppe B (Trockenstellen bis
Abkalbung: 7,9 %, Trockenstellen bis 7 - 14 d p.p.: 9,5 %, Abkalbung bis 7 -
14 d p.p.: 9,2 %) am höchsten (p<0,05). Während sie zwischen den Gruppen A
(Trockenstellen bis Abkalbung: 3,7 %, Trockenstellen bis 7-14 d p.p.: 6,4 %,
Abkalbung bis 7 - 14 d p.p.: 5,5 %) und C (Trockenstellen bis Abkalbung: 4,7
%, Trockenstellen bis 7.14 d p.p.: 6,9 %, Abkalbung bis 7 - 14 d p.p.: 6,9 %)
in allen Zeiträumen vergleichbar waren. Die Wahrscheinlichkeit einer
Neuinfektion war im Zeitraum vom Trockenstellen bis zur Abkalbung in der
Gruppe B 55,0 % höher als in der Gruppe A und 50,4 % höher als in der Gruppe
C. Die somatischen Zellgehalte (geometrische Mittelwerte in den Gruppen A, B,
C: 81,6 vs. 109,8 vs. 95,0) und die Inzidenz klinischer Mastitiden (Gruppe A,
B, C: 12,6 % vs. 11,2 % vs. 11,9 %) waren in den drei Behandlungsgruppen
vergleichbar. Allerdings traten in der Gruppe A keine, in der Gruppe B zwei
und in der Gruppe C eine klinische Mastitis während der Trockenstehphase auf.
In der viertelindividuell behandelten Gruppe C wurden im Vergleich zum
kombinierten Trockenstellen 63,8 % Antibiotika ohne eine negative
Beeinflussung der Eutergesundheit eingespart. Es kann festgestellt werden,
dass eine zusätzliche Behandlung mit einem internen Zitzenversieglers
insbesondere bei Tieren mit einer hohen Milchleistung oder einer Hyperkeratose
im Bereich des Strichkanals eine bessere Prophylaxe gegen intramammäre
Infektionen bei trockenstehenden Kühen gewährleistet als das antibiotische
Trockenstellen. Um eine Reduzierung des Einsatzes von Antibiotika zu
erreichen, ist eine Selektion zur Auffindung eutergesunder Viertel auf
Viertelebene der auf Kuhebene vorzuziehen. Das viertelindividuelle
Trockenstellen auf der Basis des Ergebnisses im CMT-Test ist in seiner
Wirksamkeit mit dem kombinierten Trockenstellen gleichzusetzen.The efficacy of the teat sealer OrbeSeal® was evaluated in two independent
field studies. In a first study to 359 cows of a dairy farm in Brandenburg was
applied to compare the combined use of a conventional dry cow antibiotic
(Orbenin extra®) with the teat sealer OrbeSeal® against the use of the
antibiotic (Orbenin extra®) alone in the Splitted-Udder-Design. The control
and treated quarters were comparable concerning the somatic cell count,
glandular tissue and teat duct hyperkeratosis. The bacterial examination of
samples immediately post partum as well as five to eight days post partum
revealed that the treated (Orbenin extra® + OrbeSeal®) quarters had a higher
rate of non-infected quarters (at calving: control 87,8 % vs. treated 91,0 %
and 5 - 8 d p.p.: control 82,1 % vs. treated 86,4 %). Moreover the probability
of a new infection was 27,1 % lower in the period from dry off to calving in
the treated quarters. In cows with a predisposition of intra mammary
infections according to high milk yield before dry period or teat duct
hyperkeratosis the protective effect of the teat sealer was pronounced. In the
second study 301 cows with an average cell count under 200.000 cells/ml in the
last three month before dry off, as well as more than a clear (++) positive
result of the California-Mastitis-Test (CMT) in one quarter were included.
Cows were split into three groups: One group was treated with a combination of
OrbeSeal® plus Orbenin extra® (group A). One group only received OrbeSeal®
(group B) and the quarters of the third group were individually treated in
dependence of the CMT result (group C). quarters with a negative or slightly
(+) positive CMT-result were treated with OrbeSeal®, and those quarters with a
clear (++) or higher positive result with the combination of Orbenin extra®
and OrbeSeal®. The study showed that the number of new infections was highest
in group B (from dry off to calving: 7,9 %, from dry off to 7 - 14 d p.p.: 9,5
%,from calving to 7 - 14 d p.p.: 9,2 %), while the new infections in group A
(from dry off to calving: 3,7 %, from dry off to 7 - 14 d p.p.: 6,4 %, from
calving to 7 - 14 d p.p.: 5,5 %) and group C (from dry off to calving: 7 - 14
d p.p.: 4,7 %, from dry off to 7 - 14 d p.p.: 6,9 %, from calving to 7 - 14 d
p.p.:.6,9 %) were comparable. The probability of a new infection in the period
between dry off and calving was highest in group B compared to group A (55 %)
and group B (50,4 %). All three groups also were comparable concerning somatic
cell counts (geometric average: group A, B, C: 81,6 vs. 109,8 vs. 95,0) and
the incidence of clinical mastitis (group A, B, C: 12,6 % vs. 11,2 % vs. 11,9
%). However it is noteworthy that two cases of clinical mastitis appeared in
group B as well as one in group C during the dry off period. Furthermore 63,8
% antibiotics could be saved in the quarter individual group C in comparison
to the combined dry off, without a negative effect on udder health. In summary
it can be said that an additional treatment with an internal teat sealer
guarantees a better prophylaxis against intramammary infections over to the
solely application an antibiotic. This was especially pronounced in treating
quarters with a predisposition for intramammary infections. To reach a
reduction in using antibiotics it is necessary to select healthy udder
quarters in an individual way. A remarkable result of this study is that the
efficacy of quarter individual dry off based on results of CMT-tests is equal
to the combined dry off

Bovine mammary secretions during involution and the peripartum period: relationship with mastitis pathogen growth
An in vitro microassay was used to evaluate growth of eight Staphylococcus strains representing five species and five strains of Corynebacterium bovis in skims from mammary secretions collected from quarters of five Holstein cows during the nonlactating and peripartum periods. Significant variation in growth among different strains of Staphylococcus species andCorynebacterium bovis and among cows was observed. All Staphylococcus species evaluated followed similar patterns of growth in mammary secretion skims. Mammary secretion skims obtained at 14 and 28 days of involution were poor media for growth of Staphylococcus species. Conversely, mammary secretion skims collected at cessation of milking, parturition, and during early lactation supported growth of all species evaluated. Staphylococcus hyicus and Staphylococcus chromogenes growth was greatest followed by Staphylococcus epidermidis, Staphylococcus xylosus, and Staphylococcus hominis. Similarly, Corynebacterium bovis grew well in mammary secretion skims obtained at cessation of milking, at parturition, and 14 days after parturition. However, growth of 4 strains in mammary secretion skims obtained at 14 and 28 day of involution was significantly lower compared to the 5th strain. Growth of Staphylococcus species in whey followed a similar pattern, but increased growth was observed in manimary secretions obtained during involution. Variation in growth of Corynebacterium bovis in mammary secretion skims and whey obtained during involution and peripartum period was not observed. However, growth of Corynebacterium bovis was reduced in whey. Mammary secretion skims were passed through S-200 gel filtration column. Four protein peaks were obtained from each skim sample. Peaks were characterized by two-dimensional polyacrylamide gel electrophoresis. The majority of proteins eluted in peak 1 and 2 from S-200 columns. Protein peaks were evaluated with respect to growth of Staphylococcus species and Corynebacterium bovis. In general, growth of Staphylococcus species and Corynebacterium bovis was reduced in peak 1 and 2 of mammary secretion skims collected during involution. These data suggest that mammary secretion skims support growth of Staphylococcus species and Corynebacterium bovis during lactation but inhibit their growth during the nonlactating period. The presence of high molecular weight biochemical components in mammary secretions are inhibitory to the growth of Staphylococcus species and Corynebacterium bovis
