    Quantifying the Differences in Binding Affinity of Reduced Glutathione for Glutathione S-Transferase at pH 6.5 and 8.5 Using Isothermal Titration Calorimetry

    The binding affinity between an enzyme and its substrate is often dependent on the pH of the local environment. Knowing the pH at which reduced glutathione (GSH) binds with the highest affinity to the enzyme glutathione S-transferase (GST) is useful for determining the optimal pH for purification of GST-fusion proteins during GST-affinity chromatography. In this study, GST of the species Schistosoma japonicum was purified, quantified, and utilized to study its binding interaction with GSH at pH 6.5 and 8.5 via isothermal titration calorimetry (ITC). After protein expression, extraction, and purification, the GST concentration was quantified using QubitTM fluorometry. Thermodynamic properties and a dissociation constant (KD) for each experiment were obtained utilizing the MicroCal PEAQ-ITC Analysis Software for the binding of GSH to GST at pH 6.5 and 8.5. Statistical analysis of the technical replicate data was performed to obtain an average and standard deviation of the KD at each pH point.     A time series analysis for sales of chicken based food product

    This study provides a time series analysis and interpretation of the output for forecast sales of chicken based food product of weekly sales data. These data were collected directly from the outlet shop of one factory in Malacca started from January 2015 to December 2016. Methods of forecasting include autoregressive (AR) method and simple exponential smoothing (SES) method. The accuracy for both methods will be compared using mean squared error (MSE), mean absolute percentage error (MAPE) and mean absolute deviation (MAD). There will be 1 period ahead of predictions for AR method and 1 period ahead for SES method. This analysis found that AR method with AR (1) model is more accurate than SES method and can be used for the future prediction of chicken based food product of weekly sales data. Recommendations for future study is trying out other method to analyse this sales of chicken based food product and using R software to analyse the dataset

    Yarn irregularity parameterisation using optical sensors

    This paper presents an original system to measure yarn diameter using coherent optical signal processing to eliminate the influence of hairiness over the output signal. The system consists of optical hardware to produce an image which characterises the yarn diameter and electronic hardware that converts the optical yarn diameter image into a proportional voltage. LabVIEW customised software was used to acquire and process the output voltage interfaced through a Data Acquisition Board. The system determines traditional commercial yarn irregularity parameters including Ud (Mean Deviation), CVd (Coefficient of Variation), SDd (Standard Deviation), types of irregularities, frequency diagrams and spectral analysis based on FFT (Fast Fourier Transform). Moreover, several other parameters were also obtained, such as DRd (Deviation Rate), IDRd (Integral Deviation Rate), spectral analysis based on FWHT (Fast Walsh Hadamard Transform) and FDFI (Fast Impulse Frequency Determination), and length of irregularities classified for an overall variation range.     A Comparison of Selected Algorithms of Form Deviation Calculation

    The article concerns the verification of accuracy of selected algorithms of form deviation evaluation. The paper presents the results of experimental investigations concerning coordinate measurements of products composed of free-form surfaces, which are characterized by various degrees of geometric complexity. The experimental investigations were conducted with the use of the method of form deviation estimation which is based on the Lagrange interpolation of measured points and two additional algorithms available in the Calypso software, which cooperates with a coordinate measuring machine. Moreover, the experimental research was performed by using of the coordinate measuring machine ACCURA II equipped with the scanning probe VAST XT. Based on the analysis of gained results of coordinate measurements the method of form deviation calculation which uses the Lagrange interpolation of measured points in order to calculate an actual curve achieved better results than one of the analysed algorithms of the Calypso inspection software

    Mathematical approach to the validation of surface texture filtration software

    A novel method for the validation of surface texture filtration software is introduced. Mathematically traceable reference pairs for linear Gaussian filtration are developed, utilising Fourier series surface definitions in conjunction with the frequency dependent transmission characteristic of the linear Gaussian filter. The novel method is demonstrated using a library of reference pairs to validate the performance of five surface texture analysis software packages. Investigations into the effects of different surface properties are made in relation to the deviation of the software-obtained results from the traceable reference values. Analysis of variance tests are used to verify the statistical significance of the results

    Thematic Mapper image quality: Preliminary results

    Based on images analyzed so far, the band to band registration accuracy of the thematic mapper is very good. For bands within the same focal plane, the mean misregistrations are well within the specification, 0.2 pixels. For bands between the cooled and uncooled focal planes, there is a consistent mean misregistration of 0.5 pixels along-scan and 0.2-0.3 pixels across-scan. It exceeds the permitted 0.3 pixels for registration of bands between focal planes. If the mean misregistrations were removed by the data processing software, an analysis of the standard deviation of the misregistration indicates all band combinations would meet the registration specifications except for those including the thermal band. Analysis of the periodic noise in one image indicates a noise component in band 1 with a spatial frequency equivalent to 3.2 pixels in the along-scan direction

    A brief analysis of the existing softwares for computer processing of microstructure photographs is presented. The descriptions of the the software package developed by the author are demonstrated. This software product is intended for quantitative metallographic analysis of digital photographs of the microstructure of materials. It allows calculating the volume fraction and the average size of particles of the structure by several hundred secants (depending on the photographs resolution) in one vision field. Besides, a special module is built in the software allowing assessing the degree of deviation of the shape of different particles and impurities from the spherical one. The article presents the main algorithms, used during the creation of the software product, and formulae according to which the software calculates the parameters of the microstructure. It is shown that the reliability of calculations depends on the quality of preparation of the microstructure

    This research aims to examine the effect of characteristics, incumbent status and political party affiliation of local head on local expenditure deviation. The independent variables are age, education level, professional background, incumbent status and political party affiliation of local heads. The dependent variable is local expenditure deviation. The population is all local heads in Indonesia in 2018. The study samples are 157 Local heads. The sampling method used is purposive sampling technique. The data analysis method is multiple regression facilitated by SPSS version 26 software. The research results show that age, education level, professional background, and incumbent status of local head do not affect on local expenditure deviation. The political party affiliation of local head has a positive effect on local expenditure deviation.
