    Principles for Starting a Song-Writing Ministry

    With the approval of Dr. Keith Currie and Dr. David Schmal, I have chosen to complete the Music & Worship Ministry Project as the Final Thesis for the Masters of Art in Music and Worship. The objective of the project is to produce an Extended Play Record (EP) that contains five original worship songs written specifically for this EP. The components of this project include: songwriting, arranging, performing, recording, and producing. This project will achieve two purposes: first, the songs in this EP are meant to be sung in my local church – Koinonia Evangelical Church (KEC); second, this project is the medium through which I complete my research on building a songwriting ministry in KEC. It answers the question: what are some foundational philosophies to starting a local church songwriting ministry? By working through the different stages of producing this record, I was able to identify seven guiding principles on which KEC will build its songwriting ministry. 1. Words Over Music 2.     Fusion pore conductance to determine the effects of mutating the structure of influenza virus hemagglutinin

    Enveloped viruses, such as influenza, infect cells by fusing their viral envelope with the cell membrane. The fusion pore is a macromolecular structure that links two membranes that are fusing. This paper will focus on the fusion pore initiated by the hemagglutinin (HA) protein of influenza virus upon infection of a host cell. Mutations in the HA protein can alter the time-course and structure of the developing fusion pore. While there is a clear relationship between HA's structure and the dynamic opening of the pore, the initial 3D structure of the fusion pore as it first begins to form remains unknown. We have attempted to address this unanswered question by measuring fusion pore conductance - a one dimensional electrophysiological measurement - at millisecond time resolution for both wild type and mutant HA proteins, using an automated patch clamp apparatus. Correlating the entire life history of the fusion pore with the snapshots we get from 3D imaging (cryo-electron tomography) would allow us to capture the initial pore opening, as well as better understand the effect that mutating the structure of HA has on influenza viral infection. At this time, we have not yet been able to observe the fusion event; however, we do believe that future experimentation using fusion pore conductance to investigate the effects of HA's structure on influenza viral infection are both promising and necessary

    Some Notes on the Pantun Storytelling of the Baduy Minority Group Its Written and Audiovisual Documentation

    Baduy pantun stories are part of the larger Sundanese oral tradition of pantun storytelling in west Java. The stories recount the deeds of the nobility of such old Sundanese kingdoms as Pajajaran and Galuh. Although the Baduy still recite the pantun stories in their rituals, in the larger cities to the east of the Baduy village Kanékés pantun recitation almost disappeared. On the basis of short periods of fieldwork in and around Kanékés village between 1976 and 2014, in this essay I shall discuss Baduy pantun storytelling. I shall summarize earlier major publications and analyse some performance aspects of two Baduy pantun stories which I recorded. Although I do not concentrate on the text, I do discuss a few cultural issues arising from the texts. Baduy oral literature also includes children\u27s and women\u27s songs, as well as fables and myths of origin (dongéng) which do not involve music. These will not be discussed here

    Nanoscale-targeted patch-clamp recordings of functional presynaptic ion channels

    Important modulatory roles have been attributed to presynaptic NMDA receptors (NMDARs) located on cerebellar interneuron terminals. Evidence supporting a presynaptic location includes an increase in the frequency of mini events following the application of NMDA and gold particle-labelled NMDA receptor antibody localisation. However, more recent work, using calcium indicators, casts doubt on the idea of presynaptic NMDARs because basket cell varicosities did not show the expected calcium rise following either the local iontophoresis of L-aspartate or the two-photon uncaging of glutamate. (In theory such calcium imaging is sensitive enough to detect the calcium rise from even a single activated receptor.) It has therefore been suggested that the effects of NMDA are mediated via the activation of somatodendritic channels, which subsequently cause a subthreshold depolarization of the axon. Here we report results from a vibrodissociated preparation of cerebellar Purkinje cells, in which the interneuron cell bodies are no longer connected but many of their terminal varicosities remain attached and functional. This preparation can retain both inhibitory and excitatory inputs. We find that the application of NMDA increases the frequency of both types of synaptic event. The characteristics of these events suggest they can originate from interneuron, parallel fiber and even climbing fiber terminals. Interestingly, retrograde signalling seems to activate only the inhibitory terminals. Finally, antibody staining of these cells shows NMDAR-like immunoreactivity co-localised with synaptic markers. Since the Purkinje cells show no evidence of postsynaptic NMDAR-mediated currents, we conclude that functional NMDA receptors are located on presynaptic terminals

    Soundwalk as a multifaceted practice

    The soundwalk was invented as part of the initiatives undertaken by the World Soundscape Project group with an acoustic ecology profile, which emphasised the noise pollution that exists in people’s sonic environment and the need to reacquire our ‘lost skill’ of conscious listening. Initially, the practice of soundwalking was used as a method allowing us to ‘hone our hearing’ (to boost our sonological competence), to show the human condition with respect to modern reality. Soon, the soundwalk became an inspiration for many artistic undertakings that made use of the sonic properties of the environment and employed various listening strategies. This article is designed to present the idea of soundwalking since its theory and practices began to form. By presenting selected works by Hildegard Westerkamp, I intend to show the motivations behind the practice of soundwalking, which encompass the complex issues of perceiving and assessing city sounds. I refer these to Tim Ingold’s proposition to understand sound as a medium of experience. Soundwalking, as a practice of conscious listening by focusing attention on aural sensations, paradoxically seems to reveal the multi‑sensory structure of our relationship with the world, and the mediatory function of sound in our experience of being‑in‑the‑world

    Long-term continuous monitoring of the preterm brain with diffuse optical tomography and electroencephalography: A technical note on cap manufacturing

    open12noDiffuse optical tomography (DOT) has recently proved useful for detecting whole-brain oxygenation changes in preterm and term newborns' brains. The data recording phase in prior explorations was limited up to a maximum of a couple of hours, a time dictated by the need to minimize skin damage caused by the protracted contact with optode holders and interference with concomitant clinical/nursing procedures. In an attempt to extend the data recording phase, we developed a new custom-made cap for multimodal DOT and electroencephalography acquisitions for the neonatal population. The cap was tested on a preterm neonate (28 weeks gestation) for a 7-day continuous monitoring period. The cap was well tolerated by the neonate, who did not suffer any evident discomfort and/or skin damage. Montage and data acquisition using our cap was operated by an attending nurse with no difficulty.
