    Virtual Alchemies: Can New Learning Technologies Transform Police Training?

    This was a collaborative action research project with three, mixed method evaluation cycles. Its aim was to increase the impact of new learning technologies (NLT) in two Police Forces. The first reconnaissance cycle found a significant absence of uptake for one computer based training system (<1% impact after five years) with only assumptions of ‘technological determinism’ driving the process. A range of individual, work related and cultural factors were illuminated, towards understanding how the Force might become a ‘network’ organisation, where technology fundamentally transforms how work is done (Symon, 2000). Following the qualitative interview study, a quantitative survey was carried out with 164 respondents on the areas of concern for NLT in the Police. Factor analysis of this data reinforced a parsimonious five factor solution, accounting for 55% of the variance and on which to proceed with cycles two and three. In the second cycle, 130 Police Probationers completed a battery of psychometrics to assess individual difference factors associated with successful outcomes in training with NLT. Results showed 27% of the variance on a bespoke NLT learning measure was predicted by computer attitudes. Age, gender, education, motivational and almost all personality measures offered no significant contribution to explaining the data. The bespoke learning criterion was used as a pre-test, post-test and retention measure and showed significant increases in knowledge were gained from NLT: effective and efficient learning was evidenced. Also in the second cycle, a sub-sample of (n=20-34) Probationers participated in two stages of repertory grid interviews rating different elements of Police training methods. From these data an ‘Ideal Probationer Learning Event’ (IPLE) model was proposed. This was stable over time and positioned NLT within a conceptual structure that identified preferred and non-preferred construct poles equating to four dimensions along which Police officers reliably rated any training event. In essence, it highlighted sites of perceived credibility and offered these as levers for creative change. Follow-up work with 120 Probationers using another example of NLT explored how integration of learning from NLT could occur from a learner-centered point of view, using an interpretive concept mapping technique. This illuminated sites of power exchange and the location of ‘ownership’ of learning in Police culture. The IPLE model was used to direct the third cycle which included constructivist and contingency perspectives. It was propsed that by creatively designing an NLT event to increase elements of the preferred constructs discovered in cycle two, integration of an NLT event would enable the transformative properties of NLT as a to manifest themselves. A sample of up to 60 Police Staff used an NLT package alongside a bespoke, collaborative e-learning group. Criterions were developed according to skill breadth, frequency and difficulty and significant increases in learning were not only found, but maintained following completion of NLT when retention was measured. The e-learning group brought some of the credible elements of training to the NLT event, thereby constituting a ‘unique solution’ (Seltzer, 1971). It created peer learning groups, despite geographical separation. Data for each level of Kirkpatrick’s (1959; 1960) ‘best practice’ training evaluation model was collected enabling the conclusion that creative integration is key to a network uptake of NLT

    Lymphocyte reactivity in patients with carcinoma of the breast and large bowel.

    The reactivity of lymphocytes from patients with either carcinoma of the breast or large bowel has been studied using the human to mouse normal lymphocyte transfer (NLT) reaction. It was found that, in the case of breast cancer, there was a direct correlation between the clinical stage and a reduced NLT reaction. Only patients with regional lymph node or generalized metastases showed significantly reduced lymphocyte reactivity. However, in the case of large bowel cancer there was a generalized reduction in NLT reactivity which was independent of the clinical stage. Incubation of lymphocytes from individuals without neoplastic disease in serum or plasma from breast cancer patients, showing reduced NLT reactivity, resulted in a reduced NLT reaction. This appears to be indicative of the presence of circulating "blocking factor" in such patients

    Toxic effects of available therapeutics are major drawbacks for conventional management approaches in parasitic infections. Vaccines have provided a promising opportunity to obviate such unwanted complications. In present study, we examined immune augmenting capacities of an emerging adjuvant, Naltrexone, against Fasciola hepatica infection in BALB/c mice. Seventy BALB/c mice were divided into five experimental groups (14 mice per group) including 1- control (received PBS), 2- vaccine (immunized with F. hepatica E/S antigens), 3- Alum-vaccine (immunized with Alum adjuvant and E/S antigens), 4- NLT-vaccine (immunized with NLT adjuvant and E/S antigens), and 5- Alum-NLT-vaccine (immunized with mixed Alum-NLT adjuvant and E/S antigens). Lymphocyte stimulation index was assessed by MTT assay. Production of IFN-γ, IL-4, IgG2a and IgG1 was assessed by ELISA method. Results showed that NLT, either alone or in combination with alum, can induce immune response toward production of IFN-γ and IgG2a as representatives of Th1 immune response. Also, using this adjuvant in immunization experiment was associated with significantly high proliferative response of splenocytes/lymphocytes. Utilization of mixed Alum-NLT adjuvant revealed the highest protection rate (73.8%) in challenge test of mice infected with F. hepatica. These findings suggest the potential role of NLT as an effective adjuvant in induction of protective cellular and Th1 immune responses against fasciolosis. © 2018 Elsevier Inc

    Design of a model for a professional development programme for a multidisciplinary science subject in the Netherlands.

    Schools are increasingly integrating multidisciplinary education into their programmes. The Minister of Education, Culture and Science has introduced a new, integrated science subject in secondary education in the Netherlands, called Nature, Life and Technology (NLT). This research note describes the design of a generic model for a professional development programme to prepare teachers with and assist them during the implementation of a multidisciplinary science module. Three sources were used for this design: subject features including school practices, the curriculum design phases, and professional development characteristics, in combination with three factors influencing the quality of the professional development: context, process and content. These sources and factors have been translated into a generic model. Three experts evaluated this generic model: a teacher trainer who was also involved in the development of NLT modules; a biology and NLT teacher who was also involved in a regional NLT information centre; and a chemistry and NLT teacher. Overall, the experts were positive. They judged 12 out of 13 aspects of the model as sufficient or better. They were, however, critical about the issue of whether the model offers sufficient possibilities for participants to familiarise themselves with new subject content knowledge

    Homeostatic NF-κB Signaling in Steady-State Migratory Dendritic Cells Regulates Immune Homeostasis and Tolerance

    SummaryMigratory non-lymphoid tissue dendritic cells (NLT-DCs) transport antigens to lymph nodes (LNs) and are required for protective immune responses in the context of inflammation and to promote tolerance to self-antigens in steady-state. However, the molecular mechanisms that elicit steady-state NLT-DC maturation and migration are unknown. By comparing the transcriptome of NLT-DCs in the skin with their migratory counterparts in draining LNs, we have identified a novel NF-κB-regulated gene network specific to migratory DCs. We show that targeted deletion of IKKβ in DCs, a major activator of NF-κB, prevents NLT-DC accumulation in LNs and compromises regulatory T cell conversion in vivo. This was associated with impaired tolerance and autoimmunity. NF-κB is generally considered the prototypical pro-inflammatory transcription factor, but this study describes a role for NF-κB signaling in DCs for immune homeostasis and tolerance that could have implications in autoimmune diseases and immunity

    The Whites Mill Tract, approximately 100 acres in size, was purchased by Salford Township in 1998 as open space. It is located in a forested landscape comprising about 10,000 acres that stretches across upper Montgomery and Bucks Counties. Because of the extensive contiguous forested area, this region is the target of several efforts to protect the natural resources and its ecological integrity (see Figure 1). The Natural Lands Trust (NLT) and the Montgomery County Conservancy have collaborated to secure conservation easements on the 1700 acre Musser Scout Reservation in the Unami Creek Valley. NLT is also working with Milford Township, Bucks County to extend protection efforts upstream along the Unami and Ridge Valley Creeks. Marlborough Township, Montgomery County recently purchased the Camp Skymount property, which is located about three-quarters mile upstream from Whites Mill pond. NLT\u27s Fulshaw-Craeg Preserve is located approximately one mile downstream. In addition NLT holds conservation easements on a number of private properties in the area

    Dynamical effects of self-generated magnetic fields in cosmic ray modified shocks

    Recent observations of greatly amplified magnetic fields (δB/B∼100\delta B/B\sim 100) around supernova shocks are consistent with the predictions of the non-linear theory of particle acceleration (NLT), if the field is generated upstream of the shock by cosmic ray induced streaming instability. The high acceleration efficiencies and large shock modifications predicted by NLT need however to be mitigated to confront observations, and this is usually assumed to be accomplished by some form of turbulent heating. We show here that magnetic fields with the strength inferred from observations have an important dynamical role on the shock, and imply a shock modification substantially reduced with respect to the naive unmagnetized case. The effect appears as soon as the pressure in the turbulent magnetic field becomes comparable with the pressure of the thermal gas. The relative importance of this unavoidable effect and of the poorly known turbulent heating is assessed. More specifically we conclude that even in the cases in which turbulent heating may be of some importance, the dynamical reaction of the field cannot be neglected, as instead is usually done in most current calculations.Comment: 4 pages, 1 figure, accepted for publication in ApJ Letter

    Identifying Hard Noise in Long-Tailed Sample Distribution

    Conventional de-noising methods rely on the assumption that all samples are independent and identically distributed, so the resultant classifier, though disturbed by noise, can still easily identify the noises as the outliers of training distribution. However, the assumption is unrealistic in large-scale data that is inevitably long-tailed. Such imbalanced training data makes a classifier less discriminative for the tail classes, whose previously "easy" noises are now turned into "hard" ones -- they are almost as outliers as the clean tail samples. We introduce this new challenge as Noisy Long-Tailed Classification (NLT). Not surprisingly, we find that most de-noising methods fail to identify the hard noises, resulting in significant performance drop on the three proposed NLT benchmarks: ImageNet-NLT, Animal10-NLT, and Food101-NLT. To this end, we design an iterative noisy learning framework called Hard-to-Easy (H2E). Our bootstrapping philosophy is to first learn a classifier as noise identifier invariant to the class and context distributional changes, reducing "hard" noises to "easy" ones, whose removal further improves the invariance. Experimental results show that our H2E outperforms state-of-the-art de-noising methods and their ablations on long-tailed settings while maintaining a stable performance on the conventional balanced settings.     Single-Cell Transcriptomics of Regulatory T Cells Reveals Trajectories of Tissue Adaptation.

    Non-lymphoid tissues (NLTs) harbor a pool of adaptive immune cells with largely unexplored phenotype and development. We used single-cell RNA-seq to characterize 35,000 CD4+ regulatory (Treg) and memory (Tmem) T cells in mouse skin and colon, their respective draining lymph nodes (LNs) and spleen. In these tissues, we identified Treg cell subpopulations with distinct degrees of NLT phenotype. Subpopulation pseudotime ordering and gene kinetics were consistent in recruitment to skin and colon, yet the initial NLT-priming in LNs and the final stages of NLT functional adaptation reflected tissue-specific differences. Predicted kinetics were recapitulated using an in vivo melanoma-induction model, validating key regulators and receptors. Finally, we profiled human blood and NLT Treg and Tmem cells, and identified cross-mammalian conserved tissue signatures.
