    Environmental Epidemiology of Intestinal Schistosomiasis in Uganda: Population Dynamics of Biomphalaria (Gastropoda: Planorbidae) in Lake Albert and Lake Victoria with Observations on Natural Infections with Digenetic Trematodes

    This study documented the population dynamics of Biomphalaria and associated natural infections with digenetic trematodes, along the shores of Lake Albert and Lake Victoria, recording local physicochemical factors. Over a two-and-a-half-year study period with monthly sampling, physicochemical factors were measured at 12 survey sites and all freshwater snails were collected. Retained Biomphalaria were subsequently monitored in laboratory aquaria for shedding trematode cercariae, which were classified as either human infective (Schistosoma mansoni) or nonhuman infective. The population dynamics of Biomphalaria differed by location and by lake and had positive relationship with pH (P < 0.001) in both lakes and negative relationship with conductivity (P = 0.04) in Lake Albert. Of the Biomphalaria collected in Lake Albert (N = 6,183), 8.9% were infected with digenetic trematodes of which 15.8% were shedding S. mansoni cercariae and 84.2% with nonhuman infective cercariae. In Lake Victoria, 2.1% of collected Biomphalaria (N = 13,172) were infected with digenetic trematodes with 13.9% shedding S. mansoni cercariae, 85.7% shedding nonhuman infective cercariae, and 0.4% of infected snails shedding both types of cercariae. Upon morphological identification, species of Biomphalaria infected included B. sudanica, B. pfeifferi, and B. stanleyi in Lake Albert and B. sudanica, B. pfeifferi, and B. choanomphala in Lake Victoria. The study found the physicochemical factors that influenced Biomphalaria population and infections. The number and extent of snails shedding S. mansoni cercariae illustrate the high risk of transmission within these lake settings. For better control of this disease, greater effort should be placed on reducing environmental contamination by improvement of local water sanitation and hygiene

    Differentiation of Schistosoma haematobium from related schistosomes by PCR amplifying an inter-repeat sequence.

    Schistosoma haematobium infects nearly 150 million people, primarily in Africa, and is transmitted by select species of local bulinid snails. These snails can host other related trematode species as well, so that effective detection and monitoring of snails infected with S. haematobium requires a successful differentiation between S. haematobium and any closely related schistosome species. To enable differential detection of S. haematobium DNA by simple polymerase chain reaction (PCR), we designed and tested primer pairs from numerous newly identified Schistosoma DNA repeat sequences. However, all pairs tested were found unsuitable for this purpose. Differentiation of S. haematobium from S. bovis, S. mattheei, S. curassoni, and S. intercalatum (but not from S. margrebowiei) was ultimately accomplished by PCR using one primer from a newly identified repeat, Sh110, and a second primer from a known schistosomal splice-leader sequence. For evaluation of residual S. haematobium transmission after control interventions, this differentiation tool will enable accurate monitoring of infected snails in areas where S. haematobium is sympatric with the most prevalent other schistosome species

    Larval trematodes infecting the South-American intertidal mud snail Heleobia australis (Rissooidea: Cochliopidae)

    Larval trematodes infecting the snail Heleobia australis (Cochliopidae) from the Bahía Blanca estuary, Argentina were surveyed for two years. A total of 7,504 snail specimens was dissected and the larval stages of 15 different trematodes were recovered and examined morphologically. These larvae included four species that had previously been reported from H. australis in Brazil, Uruguay, and Argentina: the heterophyid Ascocotyle (Phagicola) longa and the microphallids, Maritrema bonaerense, Maritrema orensense, and Microphallus simillimus. Three other species, the echinostomatid Stephanoprora uruguayensis, the microphallid Levinseniella cruzi, and the psilostomid Psilochasmus oxyurus are reported here for the first time as parasites of H. australis. Eight other trematodes found in H. australis are described and placed in the appropriate superfamilies, families or genera (Cryptogonimidae, Apocreadiidae, Aporocotylidae, Notocotylidae, Haploporidae, Renicolidae, Himasthla, and Renicola). The prevalence of the trematode taxa infecting H. australis in the Bahía Blanca estuary was low (20%). Microphallidae were the richest and the most prevalent family, probably because of the high abundance of crabs — the second-intermediate hosts of certain microphallid species — and the considerable diversity of gulls. Here we compare the parasite assemblage found in the H. australis from Bahía Blanca estuary with other parasite assemblages infecting Heleobia spp. and other rissooids from the rest of the world.Fil: Alda, Maria del Pilar.     Diagnosis of centrocestus formosanus infection in zebrafish (Danio rerio) in Italy: A window to a new globalization-derived invasive microorganism

    Centrocestus formosanus is a digenetic trematode with a complex life cycle, involving invertebrate and vertebrate hosts, humans included. In particular, it causes gill lesions and mortality in freshwater fish species, and gastrointestinal symptoms in infected humans. Here, we describe the occurrence of C. formosanus infection in zebrafish imported in Italy and propose a newly designed species-specific primer pair to ameliorate the diagnostic investigations for C. formosanus. Gill arches of 30 zebrafish were examined for the presence of encysted metacercariae under a stereomicroscope and processed through molecular analyses targeting the ribosomal internal transcribed sequence 2 (ITS2). Although C. formosanus distribution was originally restricted to Asia, it has been subsequently reported in new countries, revealing itself as an invasive species and raising important concerns for biodiversity, economy, scientific research, as well as animal and public health. Given the crucial role played by the ornamental fish industry in spreading this parasite, there is an urgent need for control measures to prevent the introduction and establishment of C. formosanus in non-endemic areas, including Europe. We also suggest developing new strategies in microbiology and epidemiology to better explore this new globalization-derived invasive species

    Clinostomum marginatum metacercaria: Incidence in Smallmouth Bass from a North Arkansas Stream and in vitro Oxygen Consumption Studies

    Small mouth bass (Micropterus dolomieui) captured from Crooked Creek (Marion Co., Arkansas) in the summers of 1977 and 1987 were found to have a high incidence of infection with the metacercaria of Clinostomum marginatum (yellow grub). Of 41 fish collected in 1977, 32 (78%) were found infected with metacercariae with some fish containing large numbers of parasites. The number of larvae per fish ranged from 1 to 184, with an average of 23.2 ± 38 per smallmouth. Eighty-six percent of the bass collected in 1987 were found positive for C. marginatum. The number of metacercariae per fish ranged from 1 to 227 with an average of 32.7 ± 54 per fish. Fish from both collection groups ranged in size from 12 to 34 cm. No significant correlation could be found between the number of metacercariae per fish and the length of the host. Using metacercariae removed from host tissue, the effect on oxygen consumption by glucose, serotonin and insulin, singularly or in combination, was measured by manometric methods. Glucose alone did not stimulate oxygen utilization, serotonin alone and with glucose was stimulatory, and insulin with glucose also increased oxygen consumption

    Helminth parasites of sheep and goats in Eastern Ethiopia

    A two-year epidemiology study of helminths of small ruminants involved the collection of viscera from 655 sheep and 632 goats from 4 abattoirs in eastern Ethiopia. A further more detailed epidemiology study of gastro-intestinal nematode infections used the Haramaya University (HU) flock of 60 Black Head Ogaden sheep. The parasitological data included numbers of nematode eggs per gram of faeces (EPG), faecal culture L3 larvae, packed red cell volume (PCV), adult worm and early L4 counts, and FAMACHA eye-colour score estimates, along with animal performance (body weight change). There were 13 species of nematodes and 4 species of flukes present in the sheep and goats, with Haemonchus contortus being the most prevalent (65–80%), followed by Trichostrongylus spp. The nematode infection levels of both sheep and goats followed the bi-modal annual rainfall pattern, with the highest worm burdens occurring during the two rain seasons (peaks in May and September). There were significant differences in worm burdens between the 4 geographic locations for both sheep and goats. Similar seasonal but not geographical variations occurred in the prevalence of flukes. There were significant correlations between EPG and PCV, EPG and FAMACHA scores, and PCV and FAMACHA scores. Moreover, H. contortus showed an increased propensity for arrested development during the dry seasons. Faecal egg count reduction tests (FECRT) conducted on the HU flocks, and flocks in surrounding small-holder communities, evaluated the efficacy of commonly used anthelmintics, including albendazole (ABZ), tetramisole (TET), a combination (ABZ + TET) and ivermectin (IVM). Initially, high levels of resistance to all of the anthelmintics were found in the HU goat flock but not in the sheep. In an attempt to restore the anthelmintic efficacy a new management system was applied to the HU goat flock, including: eliminating the existing parasite infections in the goats, exclusion from the traditional goat pastures, and initiation of communal grazing of the goats with the HU sheep and animals of the local small-holder farmers. Subsequent FECRTs revealed high levels of efficacy of all three drugs in the goat and sheep flocks, demonstrating that anthelmintic efficacy can be restored by exploiting refugia. Individual FECRTs were also conducted on 8 sheep and goat flocks owned by neighbouring small-holder farmers, who received breeding stock from the HU. In each FECRT, 50 local breed sheep and goats, 6–9 months old, were divided into 5 treatment groups: ABZ, TET, ABZ + TET, IVM and untreated control. There was no evidence of anthelmintic resistance in the nematodes, indicating that dilution of resistant parasites, which are likely to be imported with introduced breeding goats, and the low selection pressure imposed by the small-holder farmers, had prevented anthelmintic resistance from emerging

    Helminths in the gastrointestinal tract 1 as modulators of immunity and pathology

    Helminth parasites are highly prevalent in many low- and middle-income countries, in which inflammatory bowel disease and other immunopathologies are less frequent than in the developed world. Many of the most common helminths establish in the gastrointestinal tract, and can exert counter-inflammatory influences on the host immune system. For these reasons, interest has arisen in how parasites may ameliorate intestinal inflammation and whether these organisms, or products they release, could offer future therapies for immune disorders. In this review, we discuss interactions between helminth parasites and the mucosal immune system, and progress made towards identifying mechanisms and molecular mediators through which it may be possible to attenuate pathology in the intestinal tract
