    An introduction to double stain normalization technique for brain tumour histopathological images

    Stain normalization is an image pre-processing method extensively used to standardize multiple variances of staining intensity in histopathology image analysis. Staining variations may occur for several reasons, such as unstandardized protocols while preparing the specimens, using dyes from different manufacturers, and varying parameters set while capturing the digital images. In this study, a double stain normalization technique based on immunohistochemical staining is developed to improve the performance of the conventional Reinhard’s algorithm. The proposed approach began with preparing a target image by applying the contrast-limited adaptive histogram equalization (CLAHE) technique to the targeted cells. Later, the colour distribution of the input image will be matched to the colour distribution of the target image through the linear transformation process. In this study, the power-law transformation was applied to address the over-enhancement and contrast degradation issues in the conventional method. Five quality metrics comprised of entropy, tenengrad criterion (TEN), mean square error (MSE), structural similarity index (SSIM) and correlation coefficient were used to measure the performance of the proposed system. The experimental results demonstrate that the proposed method outperformed all conventional techniques. The proposed method achieved the highest average values of 5.59, 3854.11 and 94.65 for entropy, TEN, and MSE analyses

    O objetivo inerente a esta dissertação consiste no desenvolvimento de técnicas de integração de projetor num sistema de projection mapping baseado em manipulador industrial (projector in hand). O sistema em que este trabalho se insere deve ser capaz de projetar imagens a três dimensões em vigas de construção metálicas, utilizando um projetor laser ou DLP, e aplicando técnicas de projection mapping, para uma projeção correta da informação do ponto de vista do operador. Para obter essa finalidade, é necessário aplicar algoritmos para calibrar eficientemente o projetor. Dada a natureza do projection mapping, em que se efetuam projeções em superficies tridimensionais, as questões de profundidade de foco são essenciais para a eficiência do sistema. Aliado a isto, surge a necessidade de avaliar pormenorizadamente em ambiente de simulação robótica a melhor localização para acoplar o projetor no manipulador industrial

    An Investigation of Global and Local Radiomic Features for Customized Self-Assessment Mammographic Test Sets for Radiologists in China in Comparison with Those in Australia

    Self-assessment test sets have demonstrated being effective tools to improve radiologists’ diagnostic skills through immediate error feedback. Current sets use a one-size-fits-all approach in selecting challenging cases, overlooking cohort-specific weaknesses. This thesis assessed feasibility of using a comprehensive set of handcrafted global radiomic features (Stage 1, Chapter 3) as well as handcrafted (Stage 2, Chapter 4) and deep-learning based (Stage 3, Chapter 5) local radiomic features to identify challenging mammographic cases for Chinese and Australian radiologists. In the first stage, global handcrafted radiomic features and Random Forest models analyzed mammography datasets involving 36 radiologists from China and Australia independently assessing 60 dense mammographic cases. The results were used to build and evaluate models’ performance in case difficulty prediction. The second stage focused on local handcrafted radiomic features, utilizing the same dataset but extracting features from error-related local mammographic areas to analyze features linked to diagnostic errors. The final stage introduced deep learning, specifically Convolutional Neural Network (CNN), using an additional test set and radiologists’ readings to identify features linked to false positive errors. Stage 1 found that global radiomic features effectively detected false positive and false negative errors. Notably, Australian radiologists showed less predictable errors than their Chinese counterparts. Feature normalization did not improve model performance. In Stage 2, the model showed varying success rates in predicting false positives and false negatives among the two cohorts, with specific mammographic regions more prone to errors. In Stage 3, the transferred ResNet-50 architecture performed the best for both cohorts. In conclusion, the thesis affirmed the importance of radiomic features in improving curation of cohort-specific self-assessment mammography test sets

    Computational Imaging Approach to Recovery of Target Coordinates Using Orbital Sensor Data

    This dissertation addresses the components necessary for simulation of an image-based recovery of the position of a target using orbital image sensors. Each component is considered in detail, focusing on the effect that design choices and system parameters have on the accuracy of the position estimate. Changes in sensor resolution, varying amounts of blur, differences in image noise level, selection of algorithms used for each component, and lag introduced by excessive processing time all contribute to the accuracy of the result regarding recovery of target coordinates using orbital sensor data. Using physical targets and sensors in this scenario would be cost-prohibitive in the exploratory setting posed, therefore a simulated target path is generated using Bezier curves which approximate representative paths followed by the targets of interest. Orbital trajectories for the sensors are designed on an elliptical model representative of the motion of physical orbital sensors. Images from each sensor are simulated based on the position and orientation of the sensor, the position of the target, and the imaging parameters selected for the experiment (resolution, noise level, blur level, etc.). Post-processing of the simulated imagery seeks to reduce noise and blur and increase resolution. The only information available for calculating the target position by a fully implemented system are the sensor position and orientation vectors and the images from each sensor. From these data we develop a reliable method of recovering the target position and analyze the impact on near-realtime processing. We also discuss the influence of adjustments to system components on overall capabilities and address the potential system size, weight, and power requirements from realistic implementation approaches
