Identificación de Microorganismos Descomponedores de la Materia Orgánica en el Centro Académico de Investigación y Ecoturismo Biodiversidad de la UNSM – T
El objetivo general de la presente investigación fue identificar microorganismos descomponedores de la materia orgánica en suelos del Centro Académico de Investigación y Ecoturismo–Biodiversidad UNSM-T, ubicado dentro del ACR-CE. A través del aislamiento, cultivo y caracterización a nivel de género. La localización geográfica y delimitación de las áreas de interés de muestreo se realizaron teniendo en cuenta las características de vegetación, altitud y tipo de suelo, se identificaron dos zonas de muestreo (parcelas de 100x100 m) y mediante la técnica de transectos cuadrados se recolectaron 10 sub muestras de suelo con barrenos de tipo espiral cada una, las mismas que se dividieron en dos partes (análisis físico-químico y análisis biológico). Los resultados fueron la identificación taxonómica de los microorganismos descomponedores a nivel de género, a través del aislamiento de microorganismos en los medios de cultivo de los diferentes grupos de interés, el cual permitió encontrar una diversidad y abundancia de microorganismos asociados a la materia orgánica, con un valor aproximado de la población de 1,75 x 107, entre ellos Clostridium sp., Penicillium sp. y Saccharomyces sp., concluyendo que los suelos estudiados, albergan microorganismos que ayudan a la descomposición de la materia orgánica.The main objective of this research work was to identify decomposing microorganisms existing in the Academic, Research and Ecotourism - Biodiversity Center of the UNSM-T, located within the ACR-CE. The characteristics of the study area were more or less homogeneous in the forest configuration, species and density, ecological conditions and topography. The delimitation of the sampling area was done by applying simple random sampling, using the technique of square transects, for which two sampling areas (plots of 100x100 m) were identified and 10 sub samples of soil with holes were collected each , the same that were divided into two parts (physical-chemical analysis and biological analysis).
The results were the taxonomic identification of the decomposing microorganisms at the gender level, through the isolation of microorganisms in the culture media of the different interest groups, which allowed to find a diversity and abundance of microorganisms associated with organic material, with an approximate population value of 1.75 x 107, including Clostridium sp., Penicillium sp. and Saccharomyces sp., concluding that the soils studied, harbor microorganisms that help to the organic material decomposition.Trabajo de investigaciónAp
Compatibilidad de los bioplaguicidas clibadium sylvestre y trichoderma harzianum como alternativa en el control ecológico de plagas
Clibadium sylvestre is a plant that in some areas of the Amazon is used in artisanal fishing. Trichoderma harzianum is a biocontroller of fungi and phytopathogenic nematodes. The objective of this research was to evaluate the effect of the ethanolic extract of C. sylvestre seeds on the growth of T. harzianum to determine the compatibility between both biopesticides. The seeds were obtained from C. sylvestre from the department of San Martín, Peru, dried at 40°C for 48 h of exposure, pulverized, and subjected to extraction with 70% ethanol, and finally the product obtained was decanted and filtered. The bioassays with T. harzianum were carried out on Sabouraud Dextrose Agar (SDA) with the ethanolic extract of C. sylvestre at 125 to 1000 mg•L-1. T. harzianum mycelium was seeded in the central part of each Petri dish with SDA, incubated at room temperature, measuring the diameter of the colony every 12 h. The parameters evaluated in T. harzianum were colony diameter (mm) up to 84 h and growth speed (mm•h-1) up to 100 mm in diameter. The colony diameter and growth speed were different from the control, indicating that the ethanolic extract of C. sylvestre acts as an inhibitor of T. harzianum. It is concluded that both biopesticides are not compatible to be used together in ecological pest management.Clibadium sylvestre es una planta que en algunas zonas de la Amazonía es empleada en la pesca artesanal. Trichoderma harzianum es un biocontrolador de hongos y de nematodos fitopatógenos. El objetivo de esta investigación fue evaluar el efecto del extracto etanólico de semillas de C. sylvestre sobre el crecimiento de T. harzianum para determinar la compatibilidad entre ambos bioplaguicidas. De C. sylvestre procedente del departamento de San Martín, Perú se obtuvieron las semillas, fueron secadas a 40°C por 48 h de exposición, pulverizadas, y sometidas a extracción con etanol al 70%, y finalmente el producto obtenido fue decantado y filtrado. Los bioensayos con T. harzianum se realizaron en Agar Sabouraud Dextrosa (ASD) con el extracto etanólico de C. sylvestre a 125 a 1000 mg•L-1. Se sembró el micelio de T. harzianum en la parte central de cada placa de Petri con ASD, se incubó a temperatura ambiente realizando mediciones del diámetro de la colonia cada 12 h. Los parámetros evaluados en T. harzianum fueron el diámetro de la colonia (mm) hasta 84 h y la velocidad de crecimiento (mm•h-1) hasta de 100 mm de diámetro. El diámetro de la colonia y la velocidad de crecimiento fueron diferentes al control, lo que indica que el extracto etanólico de C. sylvestre actúa como un inhibidor de T. harzianum. Alternativas de manejo de la pudrición basal causada por el hongo Fusarium spp. en el cultivo de pitahaya (Hylocereus undatus), en el cantón Palora – Morona Santiago
La pitahaya amarilla (Hylocereus undatus) es un cultivo de gran importancia en nuestro país,
ya que últimamente se ha constituido como un cultivo muy rentable económicamente para los
productores del Cantón Palora. Sin embargo, la transición de planta silvestre a cultivo ha
traído consigo varios problemas agronómicos, entre ellos los fitosanitarios que reducen
significativamente la producción de la planta. Entre ellos los causados por hongos del género
Fusarium, que para su control el método más utilizado ha sido el químico, sin embargo, el uso
de agroquímicos afecta directamente a la calidad del suelo y agua, generando limitaciones
para los productores y el medio ambiente, es por esto que se han buscado alternativas más
eficientes y rentables como el uso de microrganismos antagónicos para prevenir y erradicar
enfermedades fúngicas. Por lo que en la presente investigación se experimentó con controles
alternativos y un tradicional. La investigación constó de cuatro tratamientos, cada tratamiento
con 5 repeticiones; dando un total de veinte unidades experimentales, cada una de ellas con
5 pencas de pitahaya amarilla con un total de cien. Los tratamientos consistieron en
aplicaciones tipo drench de disoluciones inoculadas con Trichoderma y Bacillus subtilis más
un tratamiento químico Carboxina (Vitavax) y un tratamiento control, en plantas infectadas
por Fusarium. El tratamiento químico resulto ser el más eficiente en el control de Fusarium,
ya que muestra un porcentaje menor en incidencia y severidad del patógeno, seguido de los
controles alternativos Trichoderma y Bacillus subtilis respectivamente, estas mostraron ser
más económicas, pero no más efectivas.The yellow pitahaya (Hylocereus undatus) is a crop of great importance in our country, since
it has recently become a very profitable crop economically for the producers of Cantón Palora.
However, the transition from wild plant to cultivation has brought with it several agronomic
problems, including phytosanitary problems that significantly reduce the production of the
plant. Among them are those caused by fungi of the genus Fusarium, which for their control
the most used method has been chemical, however, the use of agrochemicals directly affects
the quality of soil and water, generating limitations for producers and the environment, which
is why more efficient and profitable alternatives have been sought, such as the use of
antagonistic microorganisms to prevent and eradicate fungal diseases. For this reason, in the
present research we experimented with alternative controls and a traditional one. The
research consisted of four treatments, each treatment with 5 replications; giving a total of
twenty experimental units, each with 5 yellow pitahaya stalks for a total of one hundred. The
treatments consisted of drench type applications of solutions inoculated with Trichoderma and
Bacillus subtilis plus a chemical treatment Carboxin (Vitavax) and a control treatment, on
plants infected by Fusarium. The chemical treatment proved to be the most efficient in the
control of Fusarium, since it showed a lower percentage in incidence and severity of the
pathogen, followed by the alternative controls Trichoderma and Bacillus subtilis respectively,

Efectos de Trichoderma (in vitro) en los microorganismos no patógenos descomponedores de la materia orgánica de un suelo oxisol clase IV del piedemonte llanero
Titulo en ingles: Effects of Trichoderma (in vitro) in the microorganisms not patógenos in process of the organic matter of a floor oxisol class IV of the piedemonte llanero.RESUMEN:Los microorganismos son componentes importantes del suelo constituyéndosen en su parte vital y son los responsables de la dinámica de transformación y desarrollo de los procesos bioquímicos. La diversa cantidad de microorganismos que se encuentran en una fracción del suelo cumplen funciones determinantes en la transformación de los componentes orgánicos e inorgánicos. En el suelo existe un equilibrio microbiológico en donde naturalmente las poblaciones se autorregulan.En los últimos años se ha venido utilizando en la agricultura el uso del hongo heterótrofo del género Trichoderma, como controlador de organismos del suelo, dando buenos resultados antagonista, que puede alterar el equilibrio microbiológico que existe en el suelo.Por lo anterior se hizo necesario realizar pruebas de confrontación o pruebas antagónicas ( in vitro) en el laboratorio de Microbiología Vegetal de la Universidad de los Llanos, con el fin de determinar el tipo de interacción de dos cepas de Trichoderma harzianum y Trichoderma viride, con aislamientos de los hongos Aspergillus fumigatus, Aspergillus flavus, Aspergillus niger, Aspergillus repens, Mucor petrinsularis, Penicillium parvum, Rhizopus cohnii y las Bacterias, Pseudomonas sp y Bacillus sp, microorganismos benéficos descomponedores de la materia orgánica de un suelo Oxisol de terraza alta de piedemonte llanero. La cepa de Trichoderma viride se aisló de los lotes comerciales de la granja Universidad de los Llanos, vereda Barcelona, Villavicencio Meta Colombia. Y la cepa de Trichoderma harzianum, es propiedad del laboratorio de microbiología y fitopatología de la Universidad de los Llanos. Discos de Micelio de cada hongo se colocaron en los extremos de una placa o caja de petri con medio agar, papa, y dextrosa (PDA) (en caso de los hongos). Para todos los enfrentamientos se realizaron tres (3) repeticiones y el material fue incubado en condiciones de laboratorio (26º – 28º C y 12/12 horas. Luz-oscuridad). En las cajas donde se obtuvieron las colonias de bacteriasse colocaron cuatro (4) discos de micelio del hongo Trichoderma, simulando un cuadrado sobre la placa o caja de petri con medio PDA + Agar Topping, (relación 1:1). Las cepas de Trichoderma harzianum y Trichoderma viride en relación con los microorganismos heterotróficos se evaluó: crecimiento micelial(observación macroscópica) e interacciones entre las hifas (observaciones microscópicas). En el caso de las bacterias, interactuando con Trichoderma se llevaron a cabo sólo observaciones macroscópicas. Los resultados indican que las cepas de Trichoderma harzianum y Trichoderma viride, presentaron gran capacidad antagónica: Aspergillus fumigatus, Aspergillus flavus, Aspergillus niger, Aspergillus repens, Mucor petrinsularis, Rhizopus cohnii y las bacterias Pseudomonas sp y Bacillus sp. El crecimiento de Penicillium parvum no fue impedido por ninguna de las dos cepas de Trichoderma.Se concluye que Trichoderma harzianum y Trichoderma viride, son microorganismos con alta velocidad de crecimiento, alta competencia por espacio y por consiguiente pueden alterar el crecimiento de los hongos y bacterias que descomponen la materia orgánica del suelo, reduciendo la población microbial y de esta manera posiblemente disminuyendo los procesos biológicos.PALABRAS CLAVES: Antagonismo: Acción letal. Biorremediación de agua contaminada con arsénico proveniente de la parroquia Toacaso, mediante el uso de Pleurotus ostreatus, Trichoderma harzianum y Pseudomonas aeruginosa
La parroquia de Toacaso ubicada en la provincia de Cotopaxi, presenta una elevada contaminación del agua con arsénico, proveniente de forma natural del Iliniza, llegando a concentraciones de 0.70 ppm, lo que ha causado acumulación del metaloide en los cultivos y enfermedades como el cáncer en sus habitantes. El objetivo de este estudio fue evaluar la eficiencia de Pleurotus ostreatus, Trichoderma harzianum y Pseudomonas aeruginosa en la biorremediación de aguas contaminadas con As. El trabajo se llevó a cabo en dos fases: de campo y de laboratorio. En el diseño experimental, se utilizó un DBCA con 5 tratamientos y 4 repeticiones. T0 (agua contaminada con arsénico); T1 (agua contaminada con arsénico + Pleurotus ostreatus); T2 (agua contaminada con arsénico + Trichoderma harzianum); T3 (agua contaminada con arsénico + Pseudomonas aeruginosa); T4 (agua contaminada con arsénico + Pleurotus ostreatus + Trichoderma harzianum + Pseudomonas aeruginosa. Se utilizó ANOVA con Tuckey del 5% para variables con significancia. El tratamiento con Pleurotus ostreatus resultó el mejor, con una degradación de 0.70 ppm a 0.22 ppm de As, debido a que el hongo realiza biosorción a través de los micelios de rápida expansión y la quitina, siendo un excelente captador de iones metálicos. Con estos resultados, la comunidad de Toacaso podrá implementar el uso de Pleurotus ostreatus en sus ríos para disminuir la bioacumulación del metal en aguas, suelos y cultivos, siendo así la biorremediación una alternativa amigable con el ambiente, de fácil aplicación.The parish of Toacaso located in the province of Cotopaxi, presents a high levels of water pollution with arsenic (As), originated naturally from Iliniza Volcano, reaching concentrations of 0.70 ppm, which has caused metalloid accumulation in crops and diseases such as cancer in their inhabitants. The aim of this study was to evaluate the efficiency of Pleurotus ostreatus, Trichoderma harzianum and Pseudomonas aeruginosa in the bioremediation of water contaminated with As. The research was carried out in two phases: field study and laboratory work. In the experimental design, a DBCA with 5 treatments and 4 repetitions was used. Being T0 (water contaminated with arsenic); T1 (water contaminated with arsenic + Pleurotus ostreatus); T2 (water contaminated with arsenic + Trichoderma harzianum); T3 (water contaminated with arsenic + Pseudomonas aeruginosa); T4 (water contaminated with arsenic + Pleurotus ostreatus + Trichoderma harzianum + Pseudomonas aeruginosa) ANOVA with Tuckey of 5% was used for significant variables. The treatment with Pleurotus ostreatus was the best, with a degradation of 0.70 ppm to 0.22 ppm of As, due to the process of biosorption that the fungus performs through rapidly growing mycelia and chitin, being an excellent metal ion trap. With these results, the Toacaso community will be able to implement the use of Pleurotus ostreatus in its rivers to reduce the bioaccumulation of the metal in water, soil and crops, thus being bioremediation a friendly alternative to the environment, easy to apply
