    Biomedical and Pharmacological Applications of Marine Collagen

    Biomimetic polymers and materials have been widely used in a variety of biomedical and pharmacological applications. Particularly, collagen-based biomaterials have been extensively applied in various biomedical fields, such as scaffolds in tissue engineering. However, there are many challenges associated with the use of mammalian collagen, including the issues of religious constrains, allergic or autoimmune reactions, and the spread of animal diseases. Over the past few decades, marine collagen (MC) has emerged as a promising biomaterial for biomedical and pharmacological applications. Marine organisms are a rich source of structurally novel and biologically active compounds, and to date, many biological components have been isolated from various marine resources. MC offers advantages over mammalian collagen due to its water solubility, low immunogenicity, safety, biocompatibility, antimicrobial activity, functionality, and low production costs. Due to its characteristics and physicobiochemical properties, it has tremendous potential for use as a scaffold biomaterial in tissue engineering and regenerative medicine, in drug delivery systems, and as a therapeutic. In this Special Issue, we encourage submissions related to the recent developments, advancements, trends, challenges, and future perspectives in this new research field. We expect to receive contributions from different areas of multidisciplinary research, including—but not restricted to—extraction, purification, characterization, fabrication, and experimentation of MC, with a particular focus on their biotechnological, biomedical and pharmacological uses

    Managing risks and harms associated with the use of anabolic steroids

    Background: People using AAS may adopt a range of strategies to prevent and treat adverse health conditions potentially associated with the use of these substances (AAS-HC). These strategies include seeking support from physicians, using the needle and syringe exchange programme (NSP) and seeking support from informal sources such as coaches and online forums. The process of identifying risks and harms, adopting and modifying health-related strategies is similar to the methods of risk-management employed in other fields of human activity. This approach recognises the importance of the informal body of knowledge produced by decades of AAS-related folk-pharmacology and seeks to understand harm-reduction from the users’ perspective.Objectives: The primary objective of this thesis is to investigate the strategies adopted by people using AAS to prevent and treat AAS-HC. Secondary objectives include to explore the factors associated with the adoption of health strategies and the occurrence of AAS-HC, as well as the barriers and facilitators experienced by AAS users when accessing health services and other sources of support.Methods: To achieve the objectives above, three work packages (WP) were produced as part of a mixed-methods research design. WP1 is a systematic review and meta-analysis of the prevalence of AAS users seeking support from physicians. WP2 is a cross-sectional online survey that identified AAS-HC, risk factors and health-related strategies adopted by AAS users in the UK. WP3 is a qualitative study based on in-depth interviews to discuss the experiences of AAS users and their risk-management strategies (RMS).Results: The estimated overall prevalence of AAS users seeking support from physicians is 37.1%. Higher prevalence rates were observed in studies from Australia (67.3%) and amongst clients of the NSP (54.1%), whilst the lowest was observed among adolescents (17.3%). The health conditions most commonly reported by the 883 participants of the online survey were insomnia (33.3%) and anxiety (32.2%). Most participants adopted preventive strategies such as having blood tests in the last 12 months (86.2%) and seeking a GP to treat AAS-HC (55.0%). Those who sought a GP for AAS-related information were 76% less likely to report an AAS-HC in the last 12 months. The interviews described AAS users’ RMS as a continuous process of awareness and behavioural changes. Participants described an extensive use of private health services and other sources of support to bypass the barriers experienced by AAS users engaging with the public health system.Conclusion: A large number of AAS users refrain from seeking support from physicians. Health professionals should be trained to recognise and manage the most common AAS-HC and help users improve their RMS. Further studies should investigate the efficacy of AAS-related RMS and the subpopulations of AAS users more likely to experience AAS-HC and less likely to engage with health services.<br/

    Beam scanning by liquid-crystal biasing in a modified SIW structure

    A fixed-frequency beam-scanning 1D antenna based on Liquid Crystals (LCs) is designed for application in 2D scanning with lateral alignment. The 2D array environment imposes full decoupling of adjacent 1D antennas, which often conflicts with the LC requirement of DC biasing: the proposed design accommodates both. The LC medium is placed inside a Substrate Integrated Waveguide (SIW) modified to work as a Groove Gap Waveguide, with radiating slots etched on the upper broad wall, that radiates as a Leaky-Wave Antenna (LWA). This allows effective application of the DC bias voltage needed for tuning the LCs. At the same time, the RF field remains laterally confined, enabling the possibility to lay several antennas in parallel and achieve 2D beam scanning. The design is validated by simulation employing the actual properties of a commercial LC medium

    Management strategies and contributory factors for resistance exercise-induced muscle damage: an exploration of dietary protein, exercise load, and sex

    The World Health Organisation recommends that resistance exercise be performed at least twice per week to benefit general health and wellbeing. However, resistance exercise is associated with acute muscle damage that potentially can dampen muscle adaptations promoted by chronic resistance training. The extent to which muscle is damaged by exercise is influenced by various factors, including age, training status, exercise type, and – notable to this thesis – sex. To this end, establishing sex-specific management strategies for exercise-induced muscle damage (EIMD) is important to optimise the benefits of exercise. Two EIMD management strategies were focussed on in this thesis: dietary protein supplementation and exercise load manipulation. It was identified in this thesis that research into the impact both of protein supplementation and exercise load on EIMD heavily underrepresent female populations (chapters 3 and 5), despite well-documented sex differences in EIMD responses. Therefore, future research priority should be placed on bridging the sex data gap by conducting high-quality studies centralising around female-focussed and sex-comparative methodological designs. Both peri-exercise protein supplementation and exercise load manipulation in favour of lighter loads were revealed to be effective management strategies for resistance EIMD in males through systematic and scoping review of the current literature (chapters 3 and 5, respectively). Due to a lack of data from females, it is only appropriate for these strategies to be recommended for males at present. To decipher whether protein supplementation and lower exercise loads are beneficial for managing EIMD in females, a randomised controlled trial (RCT) (chapter 4) and a protocol for an RCT (chapter 6) involving male and female participants are presented in this thesis. The incorporation of ecologically-valid resistance exercise in the RCT in chapter 4 highlighted that even mild muscle damage is attenuated in females, reflected in diminished increases in post-exercise creatine kinase concentration and muscle soreness compared with males; however, the reason for this difference requires further investigation. This study, while supporting sex differences, contrasted previous studies, as neither males nor females experienced an attenuation of EIMD during milk protein supplementation. This difference likely owed to the lower severity of muscle damage induced in the current study relative to previous studies, and accordingly, future research should seek to discover alternative management strategies for mild EIMD. A protocol for an RCT examining the impact of exercise load on EIMD in untrained males and females is described in Chapter 6 of this thesis and may be used as guidance for researchers developing similar, sex-comparative studies. It was hypothesised that females will experience attenuated muscle damage relative to males and low-load exercise will induce less muscle damage than high-load exercise in both sexes. A lack of methodological consistency among EIMD studies was a recurring finding throughout this thesis, which posed an issue when attempting to compare between-study outcomes and reach a consensus. Achieving greater uniformity in study designs by adopting comparable methods relating to EIMD markers and time-points of assessment would help improve understanding of the factors influencing the magnitude of EIMD and effective management strategies. While there are limitations with several EIMD markers – for example the variability of biomarkers and subjectivity of perceptual assessments – once the optimal markers are determined, these should be consistently used moving forward. Overall, this thesis has contributed to the current body of knowledge by demonstrating that milk protein ingestion is not an effective management strategy for muscle damage following ecologically-valid resistance exercise; therefore, alternative strategies to mitigate mild muscle damage should be investigated. Further, this work supported previous reports of sex differences in EIMD and indicated that the attenuation of EIMD in females relative to males was not attributed to sex differences in body composition; thus, the aetiology of such differences necessitates further exploration by means of high-quality sex comparative research. Finally, this thesis reached the consensus recommendation that lower exercise loads can be utilised to reduce muscle damage in males; nonetheless, supporting evidence for the application of this recommendation to females is required

    Out of sight, out of mind: accessibility for people with hidden disabilities in museums and heritage sites

    As of 2020, an estimated 14.1 million residents of the United Kingdom reported a disability (DWP 2020). Within this population, approximately 6.1 million people have a hidden disability (Buhalis and Michopoulou 2011). These hidden disabilities range widely, from neurodiverse conditions like autism and dyslexia to long term chronic conditions such as fibromyalgia and arthritis. Due to the wide range of disabilities and their impact on a disabled person’s life, they have generally been underrepresented in accessibility studies. This thesis uncovers the accessibility needs of people with hidden disabilities, specifically in museums and heritage sites where they have heretofore mostly been overlooked. I utilise semi-structured interviews and correspondence with people with hidden disabilities, as well as participant-led experiences through three case study sites in Northern England, to understand the barriers they face. Their experiences help me expose the importance of passive accessibility – accessibility measures built directly into an exhibition design, such as adequate lighting and personal interpretation boards. Additionally, this thesis aims to understand the cultural forces that prevent or support accessibility-related improvements to such sites from taking place. By studying the cultural make-up of each case study organisation through ethnographic observations of the staff at these sites, institutional roadblocks to enacting accessibility-related adjustments are revealed. Specifically, the lack of communication at these sites presents a significant barrier to enacting accessibility suggestions from disabled visitors. Tying together the themes of active/passive accessibility and lack of communication is the theme of gaps in disability awareness, by which I mean that heritage organisations do not wilfully create these barriers to inclusion, and yet they create them still because they simply do not realise these things. Filling these gaps opens up countless possibilities for improving accessibility not only for people with hidden disabilities but for all visitors and staff at museums and heritage sites
