Severe phoma stem canker symptoms have been observed on winter oilseed rape in China but the seed yield loss caused by this disease remains unknown. In May 2012 and May 2013, 17 and 13 crops were surveyed, respectively, in seven counties of Hubei Province, central China. Stems with phoma stem canker disease symptoms were sampled for pathogen isolation and identification. Only L. biglobosa was identified by culture morphology and species-specific PCR; no L. maculans was found. To evaluate the yield losses, yield components (number of branches per plant, number of pods per plant, 1000-seed weight, number of seeds per pod) were assessed on healthy and diseased plants sampled from crops in four counties and on plants from inoculated pot experiments (plants of three cultivars were inoculated at the green bud stage by injecting L. biglobosa conidia into the stem between the first and second leaf scars). Results of the field surveys showed that diseased plants had 14–61% less branches and 32–83% less pods than healthy plants, respectively. The estimated seed yield loss varied from 10% to 21% and from 13% to 37% in 2012 and 2013, respectively. In the pot experiments, there were no differences in numbers of branches or pods but there were differences in number of seeds per pod between inoculated and control plants. For the three cultivars tested, the inoculated plants had yield losses of 29–56% compared with the control.     The cypress species are grown for their timber value, ornamental beauty and shelter. Their existence is threatened by the presence of cypress canker disease caused by fungal pathogens of the genus Seiridium. The long term solution for controlling this disease is to breed for cypress clones that are resistant to cypress canker. Screening for resistance is conducted by artificially inoculating cypress plants with the pathogen's inoculum. This study aimed at developing reliable methods of artificial inoculation that are suitable for New Zealand's climatic conditions. Infection of cypress plants in nature is caused by conidia but mycelial inocula are more commonly used in artificial inoculation. Several methods of inducing sporulation of Seiridium species were investigated Addition of plant substrates was shown to increase sporulation of cultures of Seiridium isolates. Studies comparing the two types of inocula (mycelial plugs and conidial suspensions) showed that mycelium inocula caused a higher percentage of canker lesions than spore inocula. Conidial inocula offer a more consistent pathogenicity. Experiments to determine the effective spore load revealed that the percentage of canker increased with the increase of inoculum load. Pathogenicity varied between species and individual colonies of Seiridium isolates. Infection of cypress in nature is thought to occur through wounds and in this work, wounding was required for infection under both glasshouse and outdoor conditions. Inoculation of the main stem and side branches showed disease symptoms develop more rapidly on side branches than on the main stem. Investigations on in vitro inoculation of tissue cultured plants and excised side shoots showed the possibility of screening cypress ramets under different environmental conditions. Temperature and percentage relative humidity were found to influence the percentage of successful inoculations on cypress plants

This study shows the first report of L. maculans isolates virulent against Rlm7 in the UK. Leptosphaeria maculans isolates virulent against Rlm7 represented 3% of the pathogen population when cultivars with the Rlm7 gene represented 5% of the UK oilseed rape area in 2012/13. However, the Rlm7 gene has been widely used since then, representing >15% of the UK oilseed rape area in 2015/16. Winter oilseed rape field experiments included cultivars with the Rlm7 gene, with the Rlm4 gene or without Rlm genes and took place at five sites in the UK over four cropping seasons. An increase in phoma leaf spotting severity on Rlm7 cultivars in successive seasons was observed. Major resistance genes played a role in preventing severe phoma leaf spotting at the beginning of the cropping season and, in addition, quantitative resistance (QR) in the cultivars examined made an important contribution to control of phoma stem canker development at the end of the cropping season. Deployment of the Rlm7 resistance gene against L. maculans in cultivars with QR in combination with sustainable disease management practices will prolong the use of this gene for effective control of phoma stem canker epidemics.Peer reviewe

    Bacterial canker of kiwifruit, caused by Pseudomonas syringae pv. actinidiae (Psa) biovar 3, was first recorded in New Zealand in November 2010 and quickly made production of the goldfleshed kiwifruit cultivar, ‘Hort16A’, which is highly susceptible to Psa, no longer viable in the Bay of Plenty region. Production of the green-fleshed cultivar, ‘Hayward’ has remained viable but there is uncertainty around its long-term productivity. This thesis investigated aspects of Psa in commercial ‘Hayward’ orchards using observational studies. The aims were to: 1) quantify a change in productivity associated with disease; 2) determine the prevalence of disease in orchards; 3) identify factors that altered the initial development of disease and 4) identify factors that impact on the presence of severe disease. Severe disease was defined as 5% or more female vines in a block showing the systemic symptoms of green shoot wilt and cane dieback. To determine Psa effects on productivity historical data from 2599 ‘Hayward’ orchards were analysed. No reduction in productivity was found until 1 year after initial detection of Psa, after controlling for other orchard inputs that affect productivity. A crosssectional survey was sent to all Psa confirmed ‘Hayward’ orchards and 430 growers provided information about one of their ‘Hayward’ orchard blocks. The survey found 84% of orchard blocks were affected by disease and 57% had green shoot-wilt and/or cane dieback reported. Blocks typically had a low within block prevalence of systemic symptoms (Median = 5% of vines). In 194 orchards that were asymptomatic at the start of the study period the probability of disease developing in a block increased in association with use of Psa protectant sprays immediately post-pruning and using artificial pollination. A lower probability of disease developing was associated with undertaking summer girdling and with the presence of older male vines. The probability of developing severe disease was investigated in 331 orchard blocks that were symptomatic. The probability increased with time after Psa was first detected in a block and was highest when frost damage occurred, when poplar, cypress or pine shelter belts were present and when artificial pollination was used. The probability of severe bacterial canker was lower when spring girdling of female vines was undertaken. The results of this study can be used to prioritise future research. The thesis has also demonstrated the utility of observational studies for plant disease research

    Importance of Leptosphaeria biglobosa as a cause of phoma stem canker on winter oilseed rape in the UK

    Phoma stem canker is a major disease of oilseed rape in the UK, leading to annual yield losses worth more than £100M. The disease is caused by two closely related species, Leptosphaeria maculans and L. biglobosa. L. maculans is generally considered more damaging, causing stem base canker; L. biglobosa is generally less damaging, causing upper stem lesions. Therefore, previous work has mainly focused on L. maculans and there has been little work on L. biglobosa. This work investigated the contribution of L. biglobosa to stem canker epidemics by assessing the amounts of DNA of L. maculans and L. biglobosa in upper stem lesions or stem base cankers on winter oilseed rape cultivars with different types of resistance against L. mac ulans. Diseased upper stem and stem base samples were collected from nine oilseed rape cultivars in a 2011/2012 field experiment at Rothamsted. The presence of L. maculans or L. biglobosa in each stem sample was detected by speciesspecific PCR. The abundance of L. maculans or L. biglobosa in each stem sample was measured by quantification of L. maculans DNA and L. biglobosa DNA using quantitative PCR (qPCR). The amounts of L. biglobosa DNA were greater than those of L. maculans DNA in both upper stem and stem base samples. These results suggest that the severe upper stem lesions and stem base cankers in the 2011/2012 cropping season were mainly caused by L. biglobosa, suggesting that L. biglobosa can sometimes cause considerable yield loss in the UK. There were differences between cultivars in the amounts of L. maculans DNA and L. biglobosa DNA, with the susceptible cultivar Drakkar having more L. maculans DNA than L. biglobosa DNA while resistant cultivars had less L. maculans DNA than L. biglobosa DNA. These results suggest that L. biglobosa can be an important cause of phoma stem canker on oilseed rape in the UK.Peer reviewedFinal Published versio

    Effect of mycoviruses on growth, spore germination and pathogenicity of the fungus Fusarium circinatum

    Producción CientíficaAim of the study: To assess the impact on two mycoviruses recently described in F. circinatum mitovirus 1, and 2-2 (FcMV1 and FcMV2-2) on i) mycelial growth, ii) spore germination and iii) relative necrosis. Material and methods: Fourteen monosporic strains of F. circinatum (one of each pair infected with mycoviruses and the other without them) of the pathogen with and without viruses were selected for the assay. The statistical analysis, were a linear mixed model of analysis of variance considering one between-subjects factor (isolate) and one within-subjects factor with four levels (1=without viruses, 2=only virus FcMV1, 3=only virus FcMV2-2 and 4=both viruses). Main results: Colony growth rates of F. circinatum isolates were significantly reduced in presence of mycoviruses (p=0.002). The spore germination was also reduced in the F. circinatum isolates containing mycovirus as compared to mycovirus-free isolates (p<0.001). No significant differences in lesion lengths caused by F. circinatum were found in relation to the presence/absence of mycovirus (p<0.61). Research highlights: Reduction of the percentage of spore germination in the isolates of F. circinatum with mycovirus infections, as compared to free isolates, provides indications of reduction of metabolic activity and plant physiology are discussed.     Susceptibility of Several Northeastern Conifers to Fusarium circinatum and Strategies for Biocontrol

    Producción CientíficaFusarium circinatum, the causal of pine pitch canker disease (PPC), is now considered among the most important pathogens of Pinaceae in the world. Although in Europe PPC is only established in the Iberian Peninsula, the potential endangered areas cover over 10 million hectares under the current host distribution and climatic conditions. It is therefore a priority to test the susceptibility of those species and their provenances, within Central and Northern Europe and find biological control agents (BCAs) against the disease. In this study, the susceptibility of Pinus sylvestris, P. mugo and Picea abies Romanian provenances to F. circinatum was tested using three inoculum doses. In parallel, the potential use of Trichoderma atroviride and Trichoderma viride as BCAs against F. circinatum was also tested. This study has demonstrated, for the first time, the susceptibility of P. mugo to F. circinatum. Likewise, the susceptibility of P. abies was also confirmed. The fact that the Romanian provenance of P. sylvestris has not been susceptible to F. circinatum suggests genetic resistance as a potential tool to manage the disease.     Insects Associated With Butternut and Butternut Canker in Minnesota and Wisconsin

    Butternut, Juglans cinerea, is being killed throughout its native range in North America by the fungus Sirococcus clavigignenti-juglandacearum. In addition to rain splashed spores, it is thought that the fungus may have spread over long distances to infect widely scattered butternut by insect vectors During surveys in 1995 and 1996 we found several insect species in close association with diseased butternut trees, and spores of S. clavigignenti-juglandacearum were isolated in pure culture from the bodies of some of these insects. Potential insect vectors were species in the coleopteran genera Eubulus (Curculionidae), Laemophlaeus (Laemophloeidae), and Glischrochilus (Nitidulidae). Because several insect species become contaminated with fungal spores, further study is needed to determine if any of these insects might transmit the fungus to healthy trees and thereby infect them

    Susceptibility to Apple Scab, Nectria Cancer and Powdery Mildew of Different Unsprayed Apple Varieties

    Twentyfive apple varieties were evaluated for Apple scab (Venturia inaqualis), Nectria canker (Nectria galligena) and Powdery mildew (Podosphaera leucotricha) for 4 to 7 years. The trees were grown unsprayed. The weather conditions at the experimental orchard normally results in high infections of apple scab and low infections of powdery mildew. Infections of Apple scab is often seen also in scab resistant varieties. Williams Pride, Katinka and Katrina (scabresistant varieties) were the only varieties not showing scab in the experimental period. The two scabresistant varieties Angold and Produkta (varieties having a poor fruit quality) was unsusceptible to Nectria canker. The reference variety Elstar/Elshof was in all plantings among the most resistant varieties. Powdery mildew is not a problem at the experimental orchard. Most varieties had low susceptibility to Powdery mildew. Varieties with low susceptibility to diseases in general were Angold, Produkta, Rajka, Rubinola, Ahrista, Gerlinde, Gul Richard, Sukkertop, Katinka, Katrina, Marian and Primicia
