Nucleic Acid Lateral Flow Immunoassay for the Detection of Pathogenic Bacteria from Food

    Get PDF
Nucleic acid lateral flow immunoassay (NALFIA) is a method combining molecular biological principle of detection with immunochemical principle of visualisation. Following isolation of DNA from the sample, a duplex PCR with two primer sets, of which one was labelled with biotin and the other with digoxigenin or fluorescein, respectively, was performed. The PCR solution and carbon particles conjugated with avidin are directly added to the nitrocellulose membrane with two test lines of immobilised antibodies specific for digoxigenin and fluorescein. The appearance of a black line indicates the presence of specific amplicon. We would like to present the NALFIA for the simultaneous detection of L. monocytogenes in particular and the genus Listeria in general, in food. Bacteria from the genus Listeria frequently contaminate a large variety of foods. Occurrence of Listeria strains in food may indicate errors in good hygienic and manufacturing practice, only L. monocytogenes is a significant human and animal pathogen responsible for the serious illness listeriosis. Conventional microbiological methods for L. monocytogenes detection are laborious and take several days to achieve a confirmed identification

Expanding the spectrum of impulse control disorders in Parkinson's disease: the phenomenology of sweet craving

Thesis (M.A.)--Boston UniversityBackground: The recognized spectrum of impulse control disorders in Parkinson’s disease (PD) includes pathologic gambling, hypersexuality, compulsive buying, and binge eating, and is commonly related to exposure to dopamine agonist medications. Sweet craving in the general population is a phenomenon that is closely linked to several factors, including poor impulse control. Craving for sweets, though recognized to occur in PD, has not previously been studied. Methods: First, patients with idiopathic PD and normal controls who reported craving sweets completed craving questionnaires (CQ), taste threshold testing, assessment of mood symptoms and olfactory testing. CQ scores were correlated with these results and other demographic information. A pathologic craving score was identified as the 75th percentile of the mean CQ score for all PD patients. Second, patients with PD and controls completed a series of questionnaires addressing the presence of a variety of impulse control disorders and sweet craving to determine the prevalence of sweet craving in PD and disease and medication-related factors that are associated with each. Results: Craving for sweets is present in about 5.5% of patients with idiopathic PD. Similar to the determinants of other ICDs, determinants of sweet craving include female gender (p=0.0001), decreased olfactory function (t-test: p=0.0001; fisher’s exact method: comparing QSIT scores of 0 and 2 (p=0.0179), 0 and 3 (p=0.0182)), self-reported current or past history of depression (p=0.048) and obsessive-compulsive traits (p=0.044), higher Hoehn & Yahr staging (p=0.0001; fisher’s exact method: no significance), younger onset of disease (p=0.015), and longer duration of disease (p=0.003). Conclusions: Craving for sweets in PD is similar in its phenomenology to other recognized ICDs in this population of patients. Though the potential implications of this behavior may be less hazardous than those of other pathologic behaviors, it broadens the spectrum of ICDs that should be recognized in PD and discussed with patients

The Wister Mud Pot Lineament: Southeastward Extension or Abandoned Strand of the San Andreas Fault?

We present the results of a survey of mud pots in the Wister Unit of the Imperial Wildlife Area. Thirty-three mud pots, pot clusters, or related geothermal vents (hundreds of pots in all) were identified, and most were found to cluster along a northwest-trending line that is more or less coincident with the postulated Sand Hills fault. An extrapolation of the trace of the San Andreas fault southeastward from its accepted terminus north of Bombay Beach very nearly coincides with the mud pot lineament and may represent a surface manifestation of the San Andreas fault southeast of the Salton Sea. Additionally, a recent survey of vents near Mullet Island in the Salton Sea revealed eight areas along a northwest-striking line where gas was bubbling up through the water and in two cases hot mud and water were being violently ejected

19091.The increase in loading ability and life expectancy of gears is a problem which decision considerably is defined now by the questions linked not only with requirements to use new techniques and materials and to raise their efficiency, but also with a load on environment. In a paper possibilities of application of thin firm coverages in the gears working in interaction with organic lubricants are considered. The paper basically is directed on possibility to use a coverage of the materials approaching for manufacture of tooth gears, and for discussion of scratch-tests results of slicks, plotted on the selected materials 12061, 14220, 15340 and 19091

A one-way dissection of high-order compact scheme for the solution of 2D Poisson equation

We present a one-way dissection formulation of high-order compact scheme for the solution of 2D Poisson equation. One-way dissection is a type of matrix reordering, divide and conquers procedure. Efficient and concise compact schemes of 4th and 6th orders are derived using the truncation errors of the Taylors’ series expansion of the governing equation. The system is split into sub-domains and each sub-domain is treated separately. Two test problems are solved to show the fourth order performance of the scheme. The direct method is used to achieve a quick solution to the problems

The State Long-Term Care Ombudsman program (SLTCOP)operates as a unit within the Iowa Department on Aging. Duties of all long-term care ombudsmen are mandated by the Older Americans Act. This office serves people living in nursing, skilled nursing, residential care, and nursing facilities in hospitals as well as elder group homes and assisted living facilities. In order to carry out all of the mandates of the Older Americans Act this office recommends to establish a fully functioning volunteer ombudsman program that meets the criteria set forth in the Federal Older Americans Act as well as to strengthen family & resident councils, and to increase collaboration between the SLTCOP and Iowa’s Aging Network
