    Research on the Dimerization Sequence of Exo70 and Its Role in Breast Cancer Cell Metastasis

    Exo70是胞外分泌复合体Exocyst的关键亚基,可通过参与Exocyst复合体或自身聚合促进细胞迁移。我们在研究中发现,Exo70自身形成二聚体的能力显著强于与Exocyst其它7个亚基的结合,提示Exo70二聚化的重要作用。进一步构建Exo70缺失突变体,并通过一系列免疫共沉淀实验确定了Exo70形成二聚体的关键位置是位于氨基酸31~35与505~509的两段同样的序列SLEKS。进而利用这两个序列的缺失突变体,通过transwell迁移实验,证实这两段SLEKS序列在Exo70二聚化及乳腺癌细胞迁移过程中的关键作用。以上结果为Exo70独立于Exocyst复合体功能之外的新功能研究提供了依据,也为乳腺癌及其转移的治疗提供潜在的靶点。Exo70,the key subunit of Exocyst protein complex,promotes cell metastasisby self-dimerization or by involving in the Exocyst complex formation.In this research,we found that Exo70 was dramatically more capable of forming self-dimerization than binding to the other subunits of exocyst complex,and aindicative of the important roles of its self-dimerization.Then we constructed a set of truncation mutants of Exo70 in order to determine the core sequence for its dimerization.With co-immun oprecipitation analysis,we identified two identical protein sequences"SLEKS"located at amino acid 31~35 and 505~509 as critical sequencesfor the dimerization of Exo70.By "cell transwell"essay,we further found that the full length Exo70 significantly promotes breast cancer metastasis,while the two truncation mutants of Exo70(Δ31~35 and Δ505~509) don't process such ability,indicating that these two "SLEKS"sequences play pivotal role in breast cancer cell metastasis and invasion.Together,these results provide fundamental basis for researches on novel functions of Exo70 besides its well-known role in Exocyst complex formation and provide potential drug target for treating breast cancer and its metastasis.国家自然科学基金青年项目(31300970);; 福州大学科研启动项目(510402)共同资

    芪参益气滴丸对ApoE基因敲除小鼠动脉粥样硬化中调节性T细胞的影响

    目的:研究芪参益气滴丸对动脉粥样硬化中调节性T细胞的影响。方法:采用6周龄雄性C57BL/6J小鼠和Apo E基因敲除小鼠,分为正常组、模型组、芪参低剂量组、芪参高剂量组,每组6只。除正常组外,其他3组小鼠均给予高脂饲料喂养,药物组还给予高、低剂量的芪参益气滴丸,8周后评价内脏指数、动脉粥样硬化病变、调节性T细胞情况。结果:与模型组比较,芪参高剂量组可显著降低肝指数(P<0.05),减少动脉粥样硬化斑块面积(P<0.05,P<0.01),并增加脾脏调节性T细胞数量(P<0.01)。结论:芪参益气滴丸可能通过增加脾脏调节性T细胞数量抑制动脉粥样硬化形成。福建省自然科学基金项目(No.2016J01413);;广东省自然科学基金项目(No.2016A030313860);;教育部留学回国人员科研启动基金项目(No.2015-311);;教育部“新世纪优秀人才支持计划”项目(No.NCET-13-0505);;国家重点研发计划“重大慢性非传染性疾病防控研究”重点专项(No.2016YFC1305903)~

    首先介绍目前无线通信中常用的3种调度算法。为了提高频谱利用率,通过仿真比较了3种常用调度算法对系统性能的影响,提出基于QoS(服务质量)业务的比例公平调度算法。该调度算法兼容了其他调度算法的优点。实验中对用户配置不同的QoS参数,从吞吐量的角度来验证不同配置的用户有不同的吞吐量,而且系统会优先保障最优用户的需求。该调度算法提高了系统的最大吞吐量,满足了用户调度的公平性,给用户提供了不同的业务服务,保障了用户的不同需求

    利用分带值的方法调查了海南东寨港和清澜港的红树林群落结构组成状况,比较了4种非胎生红树植物(瓶花木、海漆、榄李和老鼠簕)的繁殖体散布特征(大小、质量和漂浮性)和萌发的初始时间和萌发率的情况.4种非胎生红树植物在潮间带的位置从海到陆依次是:瓶花木、海漆、榄李和老鼠簕.繁殖体的大小按长度依次是榄李>老鼠簕>瓶花木>海漆,按质量依次是老鼠簕>榄李>瓶花木>海漆.瓶花木和榄李有很好的漂浮性,其次是海漆,老鼠簕的漂浮性最差.榄李的种子具有休眠的特点,因此萌发实验用50 d湿处理对榄李进行解除休眠.盐度高于30时榄李不能萌发,而老鼠簕对盐度具有很好的耐受性,在30盐度以下,萌发率都很高,甚至达到了100%.4种非胎生红树植物繁殖体的散布特征和萌发情况,对于了解红树植物在潮间带的分带现象有重要意义.Sequencing scores of four non-viviparous mangrove species(Scyphiphora hydrophyllacea Gaertn.,Excoecaria agallocha L.,Lumnitzera racemosa(Jack.) Voigt.,and Acanthus ilicifolius L.) were obtained from mangrove communities in Dongzhai harbor and Qinglan harbor in Hainan.The differences in dispersal properties such as length,mass,buoyancy and germination of these four non-viviparous mangrove species were investigated.Distribution order of these species from low to high inter-tidal zones were S.hydrophyllacea,E.agallocha,L.racemosa,A.ilicifolius.The order of diaspore longevity was L.racemosa>A.ilicifolius>S.hydrophyllacea>E.agallocha,while diaspore masses were in the order of A.ilicifolius>L.racemosa>S.hydrophyllacea>E.agallocha.Among these four non-viviparous species,diaspore buoyancy was in the descending order of S.hydrophyllacea>L.racemosa>E.agallocha>A.ilicifolius.Due to seed dormancy of L.racemosa,the propagules for germination were stored in wet condition for 50 d.Propagules of L.racemosa failed to germinate at salinities over 30.Propagules of A.ilicifolius were more tolerant to high salinity than those of L.racemosa.It was even with 100% germination at salinities less than 30.Diaspore dispersal characters of these four non-viviparous species are important in understanding the phenomenon of mangrove zonation in inter-tidal zone.国家海洋局海洋公益性行业科研专项(200805072);国家海洋局908项目(908-02-04-04);教育部新世纪优秀人才计划资
