Investigation of ABCA1 C69T polymorphism in patients with type 2 diabetes mellitus
Introduction: Non insulin dependent diabetes mellitus is the most common type of diabetes. Genetic factors, lipid profiles, hypertension are potential risk factors for diabetes mellitus. Adenosine binding cassette transporter proteins 1 (ABCA1) plays a role in cholesterol metabolism, especially high density lipoprotein (HDL-cholesterol). There are multiple mechanisms by which HDL-cholesterol can be atheroprotective, it is clear that the relative activity of ABCA1 plays a major role. We aimed to in-vestigate association of ABCA1 C69T gene polymorphism with lipid levels in Turkish type 2 diabetic patients.
Materials and methods: After isolation of DNA by ethanol precipitation we determined ABCA1 gene polymorphism by using polimerase chain reaction - restriction fragment lenght polymorphism (PCR-RFLP) method in 107 type 2 diabetic patients and 50 healthy controls.
Results: We have observed that the frequency of TT genotype is significantly higher in healthy con-trols compared to patients (14% vs. 3%; P = 0.008). Also frequency of T allele was higher in controls than in patients (34% vs. 21%; P = 0.020; OR (95% CI) = 0.52 (0.30-0.88)). There was no association of lipid levels and ABCA1 C69T polymorphism subgroups.
Conclusion: We have found significantly higher frequency of both T allele and genotype in control group when compared to patients that made us think that T allele may be a protective factor against diabetes mellitus. But, we could not find a relationship between genotypes and lipid concentrations in our two groups. Larger studies will help us to understand the relationship between ABCA1 C69T genotype and lipid parameters in diabetes mellitus
Plasma Kisspeptin-54 levels in gastric cancer patients
AbstractBackgroundKisspeptin (Kisspeptin-54; KP-54) is a 54-amino acid peptide was originally known as metastin that was implicated in suppression of tumor metastasis and circulating kisspeptin has been proposed as a tumor marker for numerous cancers in humans. However, the plasma level of KP-54 in gastric cancer (GC) remains undetermined.AimWe aimed to investigate the plasma levels of KP-54 in patients with GC.MethodsPlasma KP-54 levels were quantified with enzyme-immunoassay from blood samples of 40 patients with GC at their initial staging and 59 age-matched controls.ResultsPlasma KP-54 levels were significantly higher in GC patients (63.3 ± 17.9) than in controls (49.0 ± 12.7) (p = 0.000). Cut-off value for KP-54 was determined as 44 ng/ml and sensitivity, specificity, positive predictive value and negative predictive value, were 60%, 78%, 63%, and 74% respectively. Plasma KP-54 levels were not correlated with any clinicopathological features of GC patients (p > 0.05).ConclusionsResult of our preliminary study suggest that plasma KP-54 levels might be a useful parameter in diagnosis of GC

Current status of Great Bustard Otis tarda in Turkey: population size, distribution, movements, and threats
The Great Bustard Otis tarda is a globally threatened species with populations inhabiting the steppe zones of Turkey. In recent decades, its populations in Turkey have suffered severe declines in range and size. Although the remaining populations are in urgent need of protection, there has been no national-scale study on the species since 2009, and huge information gaps remain concerning its range, abundance, and movements in the country. Here, we combined data from five years of fieldwork together with all available sight and literature records to present up-To-date estimates of distribution and population size in Turkey, to better understand its movement patterns, to reveal its recent and historical population changes, and to assess its national extinction risk and threats. We find that the species' breeding population has shrunk by 20-29% over the last five years, and there are only 559-780 breeding Great Bustards in Turkey distributed in two discrete subpopulations. Comparison with historical records shows that the species' range has shrunk by at least 60% since the beginning of the 20th century. We suggest possible migratory routes within and through Turkey and that Turkey might have a higher regional importance for the species than previously thought. Illegal hunting, agricultural intensification, shift to irrigated crops, overgrazing, collision with powerlines, and disturbance are the most severe threats to the species in Turkey. Our national Red List assessment yields an Endangered categorisation. Further studies are needed to understand the metapopulation structure and movements of the species and to conserve its remaining populations in Turkey
