    Rac1 is essential in cocaine-induced structural plasticity of nucleus accumbens neurons

    Repeated cocaine administration increases the dendritic arborization of nucleus accumbens neurons, but the underlying signaling events remain unknown. Here, we show that repeated cocaine negatively regulates the active form of Rac1, a small GTPase that controls actin remodeling in other systems. We show further, using viral-mediated gene transfer, that overexpression of a dominant negative mutant of Rac1, or local knockout of Rac1 from floxed Rac1 mice, is sufficient to increase the density of immature dendritic spines on nucleus accumbens neurons, whereas overexpression of a constitutively active Rac1 mutant, or light activation of a photoactivatible form of Rac1, blocks the ability of repeated cocaine to produce this effect. Downregulation of Rac1 activity in nucleus accumbens likewise promotes behavioral responses to cocaine, with Rac1 activation producing the opposite effect. These findings establish an important role for Rac1 signaling in mediating structural and behavioral plasticity to cocaine

After that, this paper also explores Huawei’s core technology, industrial layout, after-sales service and other aspects. This paper suggests that Huawei should adhere to independent innovation, improve brand awareness and influence, raise awareness of its intellectual property knowledge protection, and establish a strict internal risk control mechanism. Through the comparison of the experimental and simulation results, the noise reduction effect of the secondary sound source on the computer case is obtained.     The Effect of Flow Field on the Initial Formation of Thrombus in Ventricular Assist Devices

    Abstract Thrombosis is the main reason for the failure of ventricular assist devices (VADs). It has been acknowledged that the platelet activation induced by the nonphysiological blood flow leads to the increased thrombotic risk. However, due to the complicated influence of the VADs’ flow field and the difficulty in real‐time in situ observation, the mechanisms and process of thrombus formation in VADs remain unclear. In this work, the process of thrombus formation in VADs in vitro experiments is observed. The thrombus is found to form on the middle of the inlet guide vanes first and it is mainly caused by the immediate activation of platelets induced by the high shear rate of the flow field around the vanes and also affected by the rotation of the impeller. Then, subsequent thrombus is found in the tail of guide vanes and around the axle journal, where the blood flow is stagnated and the platelets are activated by the accumulated bioagonists. These findings clarify that the thrombus formed on the inlet guide vanes and the axle journal are dominated by two different mechanisms. This work provides unique insights into the initial formation of the thrombus on VADs and helps to reduce the thrombotic risk

    Glycolysis-Related SLC2A1 Is a Potential Pan-Cancer Biomarker for Prognosis and Immunotherapy

    SLC2A1 plays a pivotal role in cancer glycometabolism. SLC2A1 has been proposed as a putative driver gene in various cancers. However, a pan-cancer analysis of SLC2A1 has not yet been performed. In this study, we explored the expression and prognosis of SLC2A1 in pan-cancer across multiple databases. We conducted genetic alteration, epigenetic, and functional enrichment analyses of SLC2A. We calculated the correlation between SLC2A1 and tumor microenvironment using the TCGA pan-cancer dataset. We observed high expression levels of SLC2A1 with poor prognosis in most cancers. The overall genetic alteration frequency of SLC2A1 was 1.8% in pan-cancer, and the SLC2A1 promoter was hypomethylation in several cancers. Most m6A-methylation-related genes positively correlated with the expression of SLC2A1 in 33 TCGA cancers. Moreover, SLC2A1 was mainly related to the functions including epithelial–mesenchymal transition, glycolysis, hypoxia, cell-cycle regulation, and DNA repair. Finally, SLC2A1 positively associated with neutrophils and cancer-associated fibroblasts in the tumor microenvironment of most cancers and significantly correlated with TMB and MSI in various cancers. Notably, SLC2A1 was remarkably positively correlated with PD-L1 and CTLA4 in most cancers. SLC2A1 might serve as an attractive pan-cancer biomarker for providing new insights into cancer therapeutics

    Background: The synergistic effects of antiangiogenic inhibitor bevacizumab and epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKI) therapy were encouraging in patients with EGFR-mutant advanced NSCLC, though some controversy remains. The specific subgroup of patients who might benefit most from the EGFR-TKI and bevacizumab combination therapy is yet to be determined.Methods: Randomized clinical trials (RCTs) that had compared the clinical efficacy of EGFR-TKI and bevacizumab combination therapy with EGFR-TKI monotherapy in treating EGFR-mutant advanced NSCLC patients published before 23 December 2022 were searched in the Cochrane, PubMed and Embase. We performed a meta-analysis for the overall survival (OS), progression-free survival (PFS), objective response rate (ORR), and treatment-related adverse events with a grade equal or more than 3 (grade≄3 TRAEs). Subgroup analyses of PFS and OS stratified by clinical characteristics and treatment were conducted.Results: We included 10 RCTs involving 1520 patients. Compared with EGFR-TKI monotherapy, addition of bevacizumab to EGFR-TKI resulted in a significantly higher PFS (hazard ratio (HR) = 0.74, 95% confidence interval (95% CI): 0.62–0.87)) and ORR (risk ratio (RR) = 1.07, 95% CI: 1.01–1.13). However, no significant difference in OS (HR = 0.96, 95% CI: 0.83–1.12) was noticed. Patients with EGFR-mutant advanced NSCLC receiving combination therapy showed PFS improvement regardless of gender (male or female), Eastern Cooperative Oncology Group performance status (0 or 1), baseline central nervous system (CNS) metastasis (presence or absence) and EGFR mutation type (19del or 21L858R). Subgroup analyses showed that, with the treatment of bevacizumab and EGFR-TKI, patients who ever smoked achieved significantly better OS and PFS benefits (HR = 0.68, 95% CI: 0.48–0.95; HR = 0.59, 95% CI: 0.46–0.74, respectively), and those aged Conclusion: Addition of bevacizumab to EGFR-TKI therapy provided significantly better PFS and ORR for EGFR-mutant advanced NSCLC patients, though with higher risk of grade≄3 TRAEs.     Latticed-Confined Conversion Chemistry of Battery Electrode

    Electrochemical conversion reaction, usually featured by multiple redox processes and high specific capacity, holds great promise in developing high-energy rechargeable battery technologies. However, the complete structural change accompanies by spontaneous atomic migration and volume variation during the charge/discharge cycle leads to electrode disintegration and performance degradation, therefore severely restrict the application of conventional conversion-type electrodes. Herein, we propose latticed-confined conversion chemistry, where the “intercalation-like” redox behaviour is realized on the electrode with a “conversion-like” high capacity. By delicately formulating the high-entropy compounds, the pristine crystal structure can be preserved well by the inert lattice framework, thus enabling an ultra-high initial Coulombic efficiency of 92.5 and a long cycling lifespan over thousand cycles after quasistatic charge-discharge cycle. This lattice-confined conversion chemistry unfolds ubiquitous insights into the localized redox reaction and sheds new lights on developing high-performance electrodes toward next-generation high-energy rechargeable batteries
