Modelling of pulse-like velocity ground motion during the 2018 M_w 6.3 Hualien earthquake, Taiwan
The 2018 February 6 M_w 6.3 Hualien earthquake caused severe localized damage in Hualien City, located 20 km away from the epicentre. The damage was due to strong (>70 cm s⁻¹) and sharp (duration ∼2.5 s) velocity pulses. The observed peak ground-motion velocity in Hualien City symmetrically decays with distance from the nearby Milun fault. Waveforms observed on the opposite sides of the fault show reversed polarity on the vertical and N–S components while the E–W component is almost identical. None of the published finite-fault slip models can explain the spatially highly localized large velocity pulses. In this study, we show that an M_w 5.9 strike-slip subevent on the Milun fault at 2.5 km depth, rupturing from north to south at ∼0.9Vs speed, combined with site effects caused by surficial layers with low S-wave speed, can explain the velocity pulses observed at the dense strong-motion network stations. This subevent contributes only 25 per cent of the total moment of the 2018 Hualien earthquake, suggesting that a small local slip patch near a metropolis can dominate the local hazard. Our result strongly suggests that seismic hazard assessments should consider large ground-motion variabilities caused by directivity and site effects, as observed in the 2018 Hualien earthquake
Develop a High-Throughput Screening Method to Identify C-P4H1 (Collagen Prolyl 4-Hydroxylase 1) Inhibitors from FDA-Approved Chemicals
Collagen prolyl 4-hydroxylase 1 (C-P4H1) is an α-ketoglutarate (α-KG)-dependent dioxygenase that catalyzes 4-hydroxylation of proline on collagen. C-P4H1-induced prolyl hydroxylation is required for proper collagen deposition and cancer metastasis. Therefore, targeting C-P4H1 is considered a potential therapeutic strategy for collagen-related cancer progression and metastasis. However, no C-P4H1 inhibitors are available for clinical testing, and the high content assay is currently not available for C-P4H1 inhibitor screening. In the present study, we developed a high-throughput screening assay by quantifying succinate, a byproduct of C-P4H-catalyzed hydroxylation. C-P4H1 is the major isoform of collagen prolyl 4-hydroxylases (CP4Hs) that contributes the majority prolyl 4-hydroxylase activity. Using C-P4H1 tetramer purified from the eukaryotic expression system, we showed that the Succinate-GloTM Hydroxylase assay was more sensitive for measuring C-P4H1 activity compared with the hydroxyproline colorimetric assay. Next, we performed high-throughput screening with the FDA-approved drug library and identified several new C-P4H1 inhibitors, including Silodosin and Ticlopidine. Silodosin and Ticlopidine inhibited C-P4H1 activity in a dose-dependent manner and suppressed collagen secretion and tumor invasion in 3D tissue culture. These C-P4H1 inhibitors provide new agents to test clinical potential of targeting C-P4H1 in suppressing cancer progression and metastasis
Probing Primordial Gravitational Waves: Ali CMB Polarization Telescope
In this paper, we will give a general introduction to the project of Ali CMB
Polarization Telescope (AliCPT), which is a Sino-US joint project led by the
Institute of High Energy Physics (IHEP) and has involved many different
institutes in China. It is the first ground-based Cosmic Microwave Background
(CMB) polarization experiment in China and an integral part of China's
Gravitational Waves Program. The main scientific goal of AliCPT project is to
probe the primordial gravitational waves (PGWs) originated from the very early
Universe.
The AliCPT project includes two stages. The first stage referred to as
AliCPT-1, is to build a telescope in the Ali region of Tibet with an altitude
of 5,250 meters. Once completed, it will be the worldwide highest ground-based
CMB observatory and open a new window for probing PGWs in northern hemisphere.
AliCPT-1 telescope is designed to have about 7,000 TES detectors at 90GHz and
150GHz. The second stage is to have a more sensitive telescope (AliCPT-2) with
the number of detectors more than 20,000.
Our simulations show that AliCPT will improve the current constraint on the
tensor-to-scalar ratio by one order of magnitude with 3 years' observation.
Besides the PGWs, the AliCPT will also enable a precise measurement on the CMB
rotation angle and provide a precise test on the CPT symmetry. We show 3 years'
observation will improve the current limit by two order of magnitude.Comment: 11 pages, 7 figures, 2 table

BACKGROUND:Our previous studies showed that Trichinella spiralis paramyosin (TsPmy) is an immunomodulatory protein that inhibits complement C1q and C8/C9 to evade host complement attack. Vaccination with recombinant TsPmy protein induced protective immunity against T. spiralis larval challenge. Due to the difficulty in producing TsPmy as a soluble recombinant protein, we prepared a DNA vaccine as an alternative approach in order to elicit a robust immunity against Trichinella infection. METHODS AND FINDINGS:The full-length TsPmy coding DNA was cloned into the eukaryotic expression plasmid pVAX1, and the recombinant pVAX1/TsPmy was transformed into attenuated Salmonella typhimurium strain SL7207. Oral vaccination of mice with this attenuated Salmonella-delivered TsPmy DNA vaccine elicited a significant mucosal sIgA response in the intestine and a systemic IgG antibody response with IgG2a as the predominant subclass. Cytokine analysis also showed a significant increase in the Th1 (IFN-γ, IL-2) and Th2 (IL-4, 5, 6, 10) responses in lymphocytes from the spleen and MLNs of immunized mice upon stimulation with TsPmy protein. The expression of the homing receptors CCR9/CCR10 on antibody secreting B cells may be related to the translocation of IgA-secreted B cells to local intestinal mucosa. The mice immunized with Salmonella-delivered TsPmy DNA vaccine produced a significant 44.8% reduction in adult worm and a 46.6% reduction in muscle larvae after challenge with T. spiralis larvae. CONCLUSION:Our results demonstrated that oral vaccination with TsPmy DNA delivered by live attenuated S. typhimurium elicited a significant local IgA response and a mixed Th1/Th2 immune response that elicited a significant protection against T. spiralis infection in mice
Forecasts of CMB lensing reconstruction of AliCPT-1 from the foreground cleaned polarization data
Cosmic microwave background radiation (CMB) observations are unavoidably
contaminated by emission from various extra-galactic foregrounds, which must be
removed to obtain reliable measurements of the cosmological signal. In this
paper, we demonstrate CMB lensing reconstruction in AliCPT-1 after foreground
removal, combine the two bands of AliCPT-1 (90 and 150~GHz) with Planck HFI
bands (100, 143, 217 and 353~GHz) and with the WMAP-K band (23~GHz). In order
to balance contamination by instrumental noise and foreground residual bias, we
adopt the Needlet Internal Linear Combination (NILC) method to clean the E-map
and the constrained Internal Linear Combination (cILC) method to clean the
B-map. The latter utilizes additional constraints on average frequency scaling
of the dust and synchrotron to remove foregrounds at the expense of somewhat
noisier maps. Assuming 4 modules observing 1 season from simulation data, the
resulting effective residual noise in E- and B-map are roughly and , respectively. As a
result, the CMB lensing reconstruction signal-to-noise ratio (SNR) from
polarization data is about SNR4.5. This lensing reconstruction
capability is comparable to that of other stage-III small aperture millimeter
CMB telescopes.Comment: 12 pages, 6 figure
Accuracy assessment of CAMS and MERRA-2 reanalysis PM2.5 and PM10 concentrations over China
Rapid industrialization and urbanization significantly contribute to air pollution in China. Essential constituents of air pollution are fine and coarse particulate matter which are the total mass of aerosol particles with aerodynamic diameters smaller than ≤2.5 μm (PM2.5) and ≤10 μm (PM10), respectively. These particles may cause severe health effects, and impact the atmospheric environment and climate. However, the limited number of ground-based measurements at sparsely distributed air quality monitoring stations hamper long-term air pollution impact studies over large areas. Although spatial data on PM2.5 and PM10 are available from reanalysis models, the accuracy of such data may be reduced in comparison with ground data and may vary regionally and seasonally. Therefore, a long-term evaluation of reanalysis-based PM2.5 and PM10 against ground-based measurements is needed for China. In this study, surface-level PM2.5 and PM10 concentrations from 2014 to 2020 obtained from the Copernicus Atmospheric Monitoring Service (CAMS), and from the second version of Modern-Era Retrospective analysis for Research and Applications (MERRA-2) were evaluated using ground-based measurements obtained from 1675 air quality monitoring stations distributed across China. High PM2.5 and PM10 (μg/m3) concentrations from ground-based measurements were observed in many parts of China (including the North China Plain: NCP, Yangtse River Delta:YRD, Pearl River Delta: PRD, Central China, Sichuan Basin: SB, and northwestern region: Tarim Basin). The patterns of the spatial distributions of PM2.5 and PM10 obtained from CAMS and MERRA-2 across China are similar to those of the ground-based monitoring data, but the concentrations from both models are substantially different. CAMS significantly overestimates PM2.5 and PM10 over most regions, in particular over urban and desert areas, whereas MERRA-2 seasonal and annual mean concentrations were more accurate over the highly polluted areas in central and eastern China. The lowest PM2.5 and PM10 concentrations were observed over the Tibetan Plateau and Qinghai, where CAMS and MERRA-2 datasets were substantially underestimated. Furthermore, both CAMS and MERRA-2 under-and over-estimate the PM concentrations in both low and high pollution conditions. Overall, this study contributes to understanding of the reliability of reanalysis data and provides a baseline document for improving the CAMS and MERRA-2 datasets for studying local and regional air quality in China
