    LambdaOpt: Learn to Regularize Recommender Models in Finer Levels

    Recommendation models mainly deal with categorical variables, such as user/item ID and attributes. Besides the high-cardinality issue, the interactions among such categorical variables are usually long-tailed, with the head made up of highly frequent values and a long tail of rare ones. This phenomenon results in the data sparsity issue, making it essential to regularize the models to ensure generalization. The common practice is to employ grid search to manually tune regularization hyperparameters based on the validation data. However, it requires non-trivial efforts and large computation resources to search the whole candidate space; even so, it may not lead to the optimal choice, for which different parameters should have different regularization strengths. In this paper, we propose a hyperparameter optimization method, LambdaOpt, which automatically and adaptively enforces regularization during training. Specifically, it updates the regularization coefficients based on the performance of validation data. With LambdaOpt, the notorious tuning of regularization hyperparameters can be avoided; more importantly, it allows fine-grained regularization (i.e. each parameter can have an individualized regularization coefficient), leading to better generalized models. We show how to employ LambdaOpt on matrix factorization, a classical model that is representative of a large family of recommender models. Extensive experiments on two public benchmarks demonstrate the superiority of our method in boosting the performance of top-K recommendation.Comment: Accepted by KDD 201

    Nanomedicine‐boosting icaritin-based immunotherapy of advanced hepatocellular carcinoma

    Traditional treatments for advanced hepatocellular carcinoma (HCC), such as surgical resection, transplantation, radiofrequency ablation, and chemotherapy are unsatisfactory, and therefore the exploration of powerful therapeutic strategies is urgently needed. Immunotherapy has emerged as a promising strategy for advanced HCC treatment due to its minimal side effects and long-lasting therapeutic memory effects. Recent studies have demonstrated that icaritin could serve as an immunomodulator for effective immunotherapy of advanced HCC. Encouragingly, in 2022, icaritin soft capsules were approved by the National Medical Products Administration (NMPA) of China for the immunotherapy of advanced HCC. However, the therapeutic efficacy of icaritin in clinical practice is impaired by its poor bioavailability and unfavorable in vivo delivery efficiency. Recently, functionalized drug delivery systems including stimuli-responsive nanocarriers, cell membrane-coated nanocarriers, and living cell-nanocarrier systems have been designed to overcome the shortcomings of drugs, including the low bioavailability and limited delivery efficiency as well as side effects. Taken together, the development of icaritin-based nanomedicines is expected to further improve the immunotherapy of advanced HCC. Herein, we compared the different preparation methods for icaritin, interpreted the HCC immune microenvironment and the mechanisms underlying icaritin for treatment of advanced HCC, and discussed both the design of icaritin-based nanomedicines with high icaritin loading and the latest progress in icaritin-based nanomedicines for advanced HCC immunotherapy. Finally, the prospects to promote further clinical translation of icaritin-based nanomedicines for the immunotherapy of advanced HCC were proposed

    Pseudogap, Superconducting Energy Scale, and Fermi Arcs in Underdoped Cuprate Superconductors

    Through the measurements of magnetic field dependence of specific heat in La2xSrxCuO4La_{2-x}Sr_xCuO_4 in zero temperature limit, we determined the nodal slope vΔv_\Delta of the quasiparticle gap. It is found that vΔv_\Delta has a very similar doping dependence of the pseudogap temperature TT^* or value Δp\Delta_p. Meanwhile the virtual maximum gap at (π,0\pi,0) derived from vΔv_\Delta is found to follow the simple relation Δq=0.46kBT\Delta_q=0.46k_BT^* upon changing the doping concentration. This strongly suggests a close relationship between the pseudogap and superconductivity. It is further found that the superconducting transition temperature is determined by both the residual density of states of the pseudogap phase and the nodal gap slope in the zero temperature limit, namely, TcβvΔγn(0)T_c \approx \beta v_\Delta \gamma_n(0), where γn(0)\gamma_n(0) is the extracted zero temperature value of the normal state specific heat coefficient which is proportional to the size of the residual Fermi arc karck_{arc}. This manifests that the superconductivity may be formed by forming a new gap on the Fermi arcs near nodes below TcT_c. These observations mimic the key predictions of the SU(2) slave boson theory based on the general resonating-valence-bond (RVB) picture.Comment: 6 pages, 6 figures, to be published in Phys. Rev.

    Simultaneous determination of human plasma protein binding of bioactive flavonoids in Polygonum orientale by equilibrium dialysis combined with UPLC–MS/MS

    AbstractA simple and selective ultra performance liquid chromatography–electrospray ionization tandem mass spectrometry (UPLC–ESI-MS/MS) assay was developed for the determination of the human plasma protein binding of four bioactive flavonoids (such as orientin and vitexin) in Polygonum orientale. Protein precipitation was used for sample preparation. Equilibrium dialysis technique was applied to determine the plasma protein binding under physiological conditions. The separation was achieved through a Waters C18 column with a mobile phase composed of 0.1% formic acid in acetonitrile and 0.1% aqueous formic acid using step gradient elution at a flow rate of 0.35mL/min. A Waters ACQUITY™ TQD system was operated under the multiple reaction monitoring (MRM) mode of positive electrospray ionization. All of the recovery, precision, accuracy and stability of the method met the requirements. Good correlations (r>0.99) of the four compounds were found, which suggested that these compounds can be simultaneously determined with acceptable accuracy. Results showed that the plasma protein bindings of the four bioactive flavonoids were in the range of 74–89% over the six concentrations studied. The binding parameters containing protein binding affinity, protein binding dissociation constant, and protein binding site were studied. The maximum ability to bind with protein was also determined in the assay in order to understand the drug-protein binding of each compound better

    The prevalence of canine Leishmania infantum infection in western China detected by PCR and serological tests

    <p>Abstract</p> <p>Background</p> <p>Canine leishmaniasis (CanL) is endemic in western China, resulting in important public health problem. It is essential to evaluate the prevalence of canine <it>Leishmania infantum </it>infection for designing control policy. In the present study we report for the first time prevalence of <it>Leishmania </it>infection in dogs living in Jiuzhaigou County (Sichuan Provence, China), which is not only an important endemic area of CanL but also a tourism scenic spot, detected by PCR, ELISA and dipstick test. The results could provide key information for designing control programs against canine and human leishmaniasis. In addition, the complete sequence of the <it>Leishmania </it>isolate from Sichuan Province has not been reported to date and we present the sequences of 116 base-pair (bp) fragment of the conserved region in the minicircle kinetoplast DNA (kDNA) and the results of phylogenetic analyses based on the sequence of the amplified fragment.</p> <p>Results</p> <p>The proportion of dogs infected with <it>Leishmania </it>in Jiuzhaigou County was 36.79%, 9.43%, and 51.88% detected by ELISA, dipstick test, and PCR, respectively. The ELISA and PCR tests were more sensitive than dipstick test. The PCR method is the most sensitive way to detect dogs infected with <it>Leishmania </it>parasites. The total positive rate for infected dogs in the area was 59.43% by the three methods. The PCR products of 116-bp fragment amplified from the kDNA conserved region of dog blood samples and laboratory maintained <it>L. infantum </it>were DNA sequenced and the variation of the sequences was observed. The phylogenetic tree based on the sequences of 116-bp fragment reveals that <it>L. infantum </it>is more genetically related to visceralizing species <it>L. donovani </it>than to the <it>Leishmania </it>species associated with cutaneous disease.</p> <p>Conclusions</p> <p>More than half of dogs living in the endemic Jiuzhaigou County were infected by <it>L. infantum</it>. Control measures, such as treatment or eradication of infected dogs, or prohibition of maintaining dogs, must be taken against these infected dogs due to their role in the transmission of the infection to vectors. The phylogenetic tree based on the sequences of conserved region in kDNA of <it>Leishmania </it>can effectively distinguish species of <it>Leishmania</it>.</p

    Fatigue Detection for Ship OOWs Based on Input Data Features, from The Perspective of Comparison with Vehicle Drivers: A Review

    Ninety percent of the world’s cargo is transported by sea, and the fatigue of ship officers of the watch (OOWs) contributes significantly to maritime accidents. The fatigue detection of ship OOWs is more difficult than that of vehicles drivers owing to an increase in the automation degree. In this study, research progress pertaining to fatigue detection in OOWs is comprehensively analysed based on a comparison with that in vehicle drivers. Fatigue detection techniques for OOWs are organised based on input sources, which include the physiological/behavioural features of OOWs, vehicle/ship features, and their comprehensive features. Prerequisites for detecting fatigue in OOWs are summarised. Subsequently, various input features applicable and existing applications to the fatigue detection of OOWs are proposed, and their limitations are analysed. The results show that the reliability of the acquired feature data is insufficient for detecting fatigue in OOWs, as well as a non-negligible invasive effect on OOWs. Hence, low-invasive physiological information pertaining to the OOWs, behaviour videos, and multisource feature data of ship characteristics should be used as inputs in future studies to realise quantitative, accurate, and real-time fatigue detections in OOWs on actual ships
