Application of a stir-tank bioreactor for perfusion culture and continuous harvest of Glycyrrhiza inflata suspension cells

    Get PDF
We developed a perfusion stir-tank bioreactor that allowed continuous cultivation and harvest of plant cells and challenged for Glycyrrhiza inflata cell suspensions. Continuous separation of cell cultures from the medium occurred using a gravitational sedimentation column. We then conducted cell retention studies of this bioreactor by detecting the cell loss in the discarded medium. We determined that complete cell retention had been achieved when the perfusion rate (perfusion rate is the ratio ofmedium feeding volume per day to bioreactor working volume) was lower than 100% d-1 and the cell suspension recirculation speed was more than 0.5 ml s-1. Growth kinetic measurements showed that the maximum cell concentration reached 25 ± 0.5 g l-1 DW (dry weight). The maximum growth rate occurred on Day 18. Under the given perfusion medium feed rate, the bioreactor operated smoothly, maintaining a relatively stable concentration of 20 g l-1 through continuous cell harvest. The bioreactorwe used could be an efficient cell culture system and demonstrates industrial potential

Interaction and Cooperative Nucleation of InAsSbP Quantum Dots and Pits on InAs(100) Substrate

    Get PDF
An example of InAsSbP quaternary quantum dots (QDs), pits and dots–pits cooperative structures’ growth on InAs(100) substrates by liquid phase epitaxy (LPE) is reported. The interaction and surface morphology of the dots–pits combinations are investigated by the high-resolution scanning electron microscope. Bimodal growth mechanism for the both QDs and pits nucleation is observed. Cooperative structures consist of the QDs banded by pits, as well as the “large” pits banded by the quantum wires are detected. The composition of the islands and the pits edges is found to be quaternary, enriched by antimony and phosphorus, respectively. This repartition is caused by dissociation of the wetting layer, followed by migration (surface diffusion) of the Sb and P atoms in opposite directions. The “small” QDs average density ranges from 0.8 to 2 × 109 cm−2, with heights and widths dimensions from 2 to 20 nm and 5 to 45 nm, respectively. The average density of the “small” pits is equal to (6–10) × 109 cm−2 with dimensions of 5–40 nm in width and depth. Lifshits–Slezov-like distribution for the amount and surface density of both “small” QDs and pits versus their average diameter is experimentally detected. A displacement of the absorption edge toward the long wavelength region and enlargement toward the short wavelength region is detected by the Fourier transform infrared spectrometry

New Terms for the Compact Form of Electroweak Chiral Lagrangian

The compact form of the electroweak chiral Lagrangian is a reformulation of its original form and is expressed in terms of chiral rotated electroweak gauge fields, which is crucial for relating the information of underlying theories to the coefficients of the low-energy effective Lagrangian. However the compact form obtained in previous works is not complete. In this letter we add several new chiral invariant terms to it and discuss the contributions of these terms to the original electroweak chiral Lagrangian.Comment: 3 pages, references adde

    High Performance Perovskite Solar Cells

    Get PDF
Perovskite solar cells fabricated from organometal halide light harvesters have captured significant attention due to their tremendously low device costs as well as unprecedented rapid progress on power conversion efficiency (PCE). A certified PCE of 20.1% was achieved in late 2014 following the first study of long-term stable all-solid-state perovskite solar cell with a PCE of 9.7% in 2012, showing their promising potential towards future cost-effective and high performance solar cells. Here, notable achievements of primary device configuration involving perovskite layer, hole-transporting materials (HTMs) and electron-transporting materials (ETMs) are reviewed. Numerous strategies for enhancing photovoltaic parameters of perovskite solar cells, including morphology and crystallization control of perovskite layer, HTMs design and ETMs modifications are discussed in detail. In addition, perovskite solar cells outside of HTMs and ETMs are mentioned as well, providing guidelines for further simplification of device processing and hence cost reduction

The tuned absorptance in multilayer graphene-dielectric structures by intraband transition

© 2017 Author(s). In this work, using the transfer-matrix method, the optical transport process is investigated, with graphene inserted into multilayer dielectric structures, theoretically and numerically in the THz regime. When the incident frequency is lower than the graphene Fermi energy, the optical intra-band transitions provide the main contribution to the graphene surface current. The absorptance can be enhanced to about 50% with only one graphene/dielectric layer in air. When increasing the number of dielectric layers coated with graphene, the absorption increases. Periodic absorption peaks are observed in multilayer structures. The positions of the absorption peaks as a function of the frequency and the incident angle are in accordance with the positions of the abrupt change in the reflection coefficient phase and of the imaginary solution of the Bloch wavevector in expanding periodic structures using Bloch theorem

Coherent quantum transport in ferromagnet/superconductor/ferromagnet structures

The Blonder-Tinkham-Klapwijk (BTK) approach is extended to study coherent quantum transport in ferromagnet/superconductor/ferromagnet (FM/SC/SM) double tunnel junctions. In order to guarantee current conservation it is necessary to simultaneously consider spin-polarized electron currents along one direction and spin-polarized hole currents along the opposite direction, and to determine self-consistently the chemical potential in SC. It is found that all the reflection and transmission coefficients'in BTK theory as well as conductance spectra oscillate with energy, exhibiting different behavior in the metallic and tunnel limits.published_or_final_versio

Inhibitory mechanism of 3-hydroxypropionaldehyde accumulation in 1,3-propanediol synthesis with Klebsiella pneumoniae

    Get PDF
3-Hydroxypropionaldehyde accumulation may cause the cessation of 1,3-propanediol sustained production with glycerol by Klebsiella pneumoniae. The impeller tip speed shift from higher to lower speed at glycerol excess or the pulsed glycerol feeding could lead to an abrupt increase of the 3- hydroxypropionaldehyde concentration (up to 10 mmol/l) in 10 min. The intracellular consequence of the 3-hydroxypropionaldehyde accumulation has not yet been elucidated. The rapid accumulation of 3- hydroxypropionaldehyde relying on the impeller tip speed shift was employed to investigate the influences of 3-hydroxypropionaldehyde to the activities of nine key enzymes related to glycerol metabolism, CO2 and O2 levels in off-gas, cell growth and 1,3-propanediol synthesis. Compared with that at 1.19 mmol/l 3-hydroxypropionaldehyde in broth, the residual enzymatic activities of the nine key enzymes ranged from 9.44 to 74.68% in the cultures at 7.5 mmol/l 3-hydroxypropionaldehyde in broth. The inhibitions of cell growth and the 1,3-propanediol synthesis were unnoticeable at the low level of 3- hydroxypropionaldehyde. By contrast, the CO2 and O2 levels changes in off-gas response to the 3- hydroxypropionaldehyde accumulation were less than 15 min. These results suggest that 3- hydroxypropionaldehyde inhibited the growth and metabolism of K. pneumoniae in a more complicated manner. Keywords: Fermentation, glycerol, 3-hydroxypropionaldehyde, Klebsiella pneumoniae, 1,3-propanediol
