Fontes protéicas e atividade de enzimas digestivas em jundiás (Rhamdia quelen)
As enzimas digestivas influenciam a utilização dos alimentos em peixes, e seu conhecimento é importante para otimizar a formulação de dietas. Este trabalho descreve a atividade de enzimas digestivas em juvenis de jundiá alimentados com fontes protéicas. Os peixes foram alimentados com seis dietas (90 dias): MBY (farinha de carne e ossos + levedura de cana), SY (farelo de soja + levedura de cana), S (farelo de soja), MBS (farinha de carne e ossos + farelo de soja), FY (farinha de peixe + levedura de cana) e FS (farinha de peixe + farelo de soja). A cada 30 dias, foram analisadas as enzimas digestivas (tripsina, quimiotripsina e amilase) no intestino. No estômago, foi mensurada a protease ácida. Foram estimados os índices digestivo e hepato-somático, quociente intestinal, comprimento do trato digestório e ganho em peso. As atividades de tripsina e quimiotripsina foram maiores (p < 0,0001) nos peixes alimentados com as dietas com fontes de origem animal (MBY, MBS, FY e FS). As proteases alcalinas foram afetadas negativamente pelo farelo de soja nas dietas. A atividade de amilase apresentou grandes variações. Os peixes alimentados com as dietas MBY e MBS apresentaram maior atividade de protease ácida (p < 0,0001). Os ganhos em peso foram maiores nos peixes alimentados com as dietas MBS e FS (p < 0,05). Os maiores valores (p < 0,05) de comprimento do trato digestório foram verificados nos peixes alimentados com as dietas MBS e FY. A composição da dieta influencia na atividade de enzimas digestivas do jundiá.Digestive enzymes activity influence feed utilization by fish, and its understanding is important to optimize diet formulation. This study reports the digestive enzyme activities of jundiá juveniles fed diets with protein sources. Fish were fed six experimental diets for 90 days: MBY (meat and bone meal + sugar cane yeast), SY (soybean meal + sugar cane yeast), S (soybean meal), MBS (meat and bone meal + soybean meal), FY (fish meal + sugar cane yeast) and FS (fish meal + soybean meal), and then sampled every 30 days and assayed in two intestine sections for digestive enzymes - trypsin, chymotrypsin and amylase - activities; gastric protease was assayed in the stomach. Digestive and hepatosomatic index, intestinal quotient, digestive tract length and weight gain were also measured. Trypsin and chymotrypsin activities were higher (p < 0.0001) in fish fed diets containing animal protein sources (MBY, MBS, FY and FS diets). Alkaline proteases were negatively affected by dietary soybean meal in the SY and S diets. Amylase activity had greater variation between diets and intestine sections. Fish fed MBY and MBS diets showed higher gastric protease activity (p < 0.0001). Weight gains were higher in fish fed the MBS and FS diets (p < 0.05). Highest values (p < 0.05) of digestive tract length were observed in fish fed MBS and FY diets, but no correlation with enzymes activities was found. Acetylcholinesterase enzyme activity in carp brain and muscle after acute exposure to diafuran
Sublethal adverse effects may result from exposure of aquatic organisms to insecticides at environmentally relevant concentrations. Fingerlings of the common carp (Cyprinus carpio, Linnaeus, 1758), grass carp (Ctenopharyngodon idella, Valenciennes, 1844), and bighead carp (Aristichthys nobilis, Richardson, 1845) were exposed to diafuran, an insecticide widely used during rice cultivation in Southern Brazil. The aim of this study was to verify the relationship between the lethal concentration (LC50) of diafuran and the acetylcholinesterase (AChE) activity in brain and muscle tissues of these species as a possible early biomarker of exposure to this insecticide. LC50 was determined for fish exposed to diafuran concentrations during 96 h (short term): common carp: control, 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0 mg L-1; grass carp: control, 1.0, 2.0, 3.0 and 3.5 mg L-1 and, bighead carp: control, 0.5, 1.0, 1.5, 2.0, 3.0 and 4.0 mg L-1, as well as the determination of AChE at concentrations near LC50 for these species. LC50 values (nominal concentrations) were 1.81 mg L-1 for the common carp, 2.71 mg L-1 for the grass carp and, 2.37 mg L-1 for the bighead carp. All carps exposed to diafuran were lethargic (lower concentrations) or immobile. Diafuran inhibited the acetylcholinesterase activity in brain (~38%) and muscle (~50%) of all species. Muscle of bighead carp under control treatment showed higher specific AChE activity than brain (14.44 against 5.94 µmol min-1 g protein-1, respectively). Concentrations of diafuran used for rice cropping may affect Cyprinus carpio, Ctenopharyngodon idella and Aristichthys nobilis behaviors and the AChE activities in brain and muscle of these species may be an early biomarker of toxicity of this insecticide.Exposição a inseticidas em concentrações elevadas no ambiente podem ocasionar efeitos adversos subletais em organismos aquáticos. Alevinos de carpa húngara (Cyprinus carpio, Linnaeus, 1758), carpa capim (Ctenopharyngodon idella, Valenciennes, 1844) e carpa cabeça grande (Aristichthys nobilis, Richardson, 1845) foram expostos ao diafuran, um inseticida utilizado na cultura do arroz no sul do Brasil. O objetivo deste estudo foi verificar a relação entre concentração letal mediana (CL50) do diafuran e a atividade da enzima acetilcolinesterase (AChE) em cérebro e músculo dessas espécies, como um possível biomarcador inicial da exposição a este inseticida. A CL50 foi determinada com peixes expostos a concentrações de diafuran em 96 h: carpa húngara: controle; 0,5; 1,0; 1,5; 2,0; 2,5 e 3,0 mg L-1; carpa capim: controle; 1,0; 2,0; 3,0 e 3,5 mg L-1 e carpa cabeça grande: controle; 0,5; 1,0; 1,5; 2,0; 3,0 e 4,0 mg L-1, bem como a determinação da AChE em concentrações próximas da CL50 para essas espécies. Valores de CL50 (concentrações nominais) foram de 1,81 mg L-1 para carpa húngara, 2,71 mg L-1 para carpa capim e 2,37 mg L-1 para carpa cabeça grande. Todas as carpas expostas ao diafuran estavam letárgicas (menores concentrações) ou imóveis. Diafuran inibiu significativamente a atividade da AChE em cérebro (~38 %) e músculo (~50 %) de todas as espécies estudadas. Atividade da AChE em músculo para carpa cabeça grande foi mais alta que cérebro (14,44 contra 5,94 µmol min-1 g proteína-1, respectivamente). Anesthesia of silver catfish with eugenol: time of induction, cortisol response and sensory analysis of fillet
The aim of this study was to identify the time of anesthetic induction and recovery of silver catfish (Rhamdia quelen) exposed to eugenol. It was also determined the efficacy of the anesthetic as a stress reducing agent and performed a sensory analysis of the fillets from fish exposed to this substance. The silver catfish were exposed to air for 1min to carry out biometry, and blood was collected at 0, 1 and 4 hours later. Eugenol can be used in the range of 20-50mg L-1 for anesthetic induction in silver catfish, and recovery time from anesthesia was not affected by eugenol concentration. The control group showed significantly higher cortisol levels 4 hours after biometry than at time zero. Fish anesthetized with eugenol (50mg L-1) presented significantly lower plasma cortisol levels than control fish at the same time. These data indicate that eugenol inhibits the rise of cortisol in the blood. The sensory analysis test demonstrated that eugenol modifies the flavor of the fillet and therefore is contra-indicated for anesthetization of silver catfish that are intended for human consumption
Oxidative stress parameters and antioxidant response to sublethal waterborne zinc in a euryhaline teleost Fundulus heteroclitus : protective effects of salinity
Zinc is an essential trace metal, but many aspects of its toxicity remain unclear. In this study, we investigated zinc effects on oxidative stress parameters and antioxidant profile in four tissues (gill, liver, kidney, and white muscle) of Fundulus heteroclitus. Possible interactive effects of salinity were also studied. Killifish were exposed to sublethal level (500 μgL(-1)) of waterborne zinc for 96 h in 0% (fresh water), 10% (3.5 ppt), 30% (10.5 ppt) and 100% sea water (35 ppt). Salinity per se had no effect on any parameter in the control groups. Zinc exposure clearly induced oxidative stress, and responses were qualitatively similar amongst different tissues. Salinity acted as a strong protective factor, with the highest levels of reactive oxygen species (ROS) and greatest damage (protein carbonyls, lipid peroxidation as indicated by thiobarbituric acid reactive substances (TBARS) in 0 ppt, the least in 100% sea water (35 ppt), and gradations in between in many of the observed responses. Increases in total oxidative scavenging capacity (TOSC) occurred at higher salinities, correlated with increases in the activities of superoxide dismutase (SOD) and glutathione-S-tranferase (GST), as well as in tissue glutathione (GSH) concentrations. However, TOSC was depleted in zinc-exposed fish at 0 ppt, accompanied by decreases in SOD, GST, GSH, and also catalase (CAT) activity. Our results confirm that sublethal waterborne zinc is an oxidative stressor in fish, and highlight the important protective role of higher salinities in ameliorating the oxidative stress associated with zinc toxicity in this model estuarine teleost
Ammonia excretion at different life stages of silver catfish
This study examined ammonia excretion at different life stages (eggs, larvae and juveniles) in silver catfish (Rhamdia quelen) and determined the influence of fasting time on ammonia excretion. Eggs and larvae were collected from incubators at different times after fecundation and placed in chambers. Juveniles were separated into two weight classes (2–50 g and 150-320 g) and placed in individual chambers after feeding. Water was collected from each chamber to determine ammonia excretion. Ammonia excretion by the eggs was low, but when hatching began approximately 28h after fecundation, excretion increased until 48h after fecundation. In fasting silver catfish juveniles, there was a significant negative correlation between ammonia excretion and weight. Moreover, ammonia excretion decreased significantly after 12 and 48h of fasting (compared to 6h fasting) in the smallest and largest specimens, respectively. Consequently, during incubation of silver catfish eggs, water renovation must be increased at hatching time to avoid a build-up in the concentration of ammonia. In addition, as ammonia excretion in this species increases after feeding, feed must be discontinued when ammonia levels in the tanks are high to avoid a further increase of this metabolite and consequent mortality of silver catfish.Verificou-se a excreção de amônia em diferentes estágios de vida (ovos, larvas e juvenis) e determinou-se a influência do tempo de jejum na excreção de amônia no jundiá, Rhamdia quelen. As águas foram coletadas de cada recipiente para determinação da excreção de amônia. A excreção de amônia pelos ovos foi baixa, mas iniciada a eclosão – 28h após a fecundação, ela aumentou até as 48h. Nos juvenis em jejum, houve significativa correlação negativa entre a excreção de amônia e o peso. Além disso, a excreção de amônia diminuiu significativamente após 12 e 48h de jejum nos
espécimes menores e maiores, respectivamente. Portanto, a renovação da água deve ser aumentada no momento da eclosão dos ovos de jundiá para evitar aumento da concentração de amônia. Como nessa espécie a excreção de amônia aumenta após a alimentação, quando os níveis nos tanques forem elevados a alimentação deve ser suspensa para evitar aumento adicional deste metabólito e mortalidade

Association of Lipids with Oxidative Stress Biomarkers in Subclinical Hypothyroidism
Objective. The aim of the present study was to evaluate the oxidative stress biomarkers in patients with subclinical hypothyroidism (n=20) and health controls (n=20). Subjects and Methods. Total cholesterol (TC), triglycerides (TGs), low-density lipoprotein-cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), thiobarbituric acid reactive substances (TBARSs), catalase (CAT), superoxide dismutase (SOD), and arylesterase (ARE) were analyzed. Results. TC, LDL-C, TBARS, and CAT were higher in subclinical hypothyroidism patients, whereas SOD did not change. Arylesterase activity was significantly lower in the SH group, compared with the control group. Correlation analyses revealed the association of lipids (TC and LDL-C) with both oxidative stress biomarkers and thyrotropin (TSH). Thyroid hormones were correlated only with triglyceride levels. In addition, TSH was significantly correlated with TBARS, CAT, and SOD. However, no significant correlations were observed after controlling TC levels. Conclusions. We found that SH patients are under increased oxidative stress manifested by reduced ARE activity and elevated lipoperoxidation and CAT activity. Secondary hypercholesterolemia to thyroid dysfunction and not hypothyroidism per se appears to be associated with oxidative stress in subclinical hypothyroidism
