    The Alcoholic Bark Extract of Terminalia Arjuna Exhibits Cytotoxic and Cytostatic Activity on Jurkat Leukemia Cells

    Abstract: Background: Natural products are characterized by complex chemical composition and are capable of concurrently modulate several signalling pathways. Considering the biological com- plexity of carcinogenesis, natural products represent key components of the therapeutic armamen- tarium for oncological diseases. The bark of Terminalia arjuna is used in traditional Ayurvedic medicine for its astringent, expectorant, cardiotonic, styptic, and antidysenteric properties. Along- side its traditional uses, Terminalia arjuna exhibits different biological activities including an- timutagenic and anticarcinogenic. Objective: This study was designed to evaluate the toxic effects of an alcoholic extract obtained from the bark of T. arjuna on a human T-lymphoblastic cell line (Jurkat). We explored the phyto- chemical composition and investigated the cytotoxic, cytostatic, genotoxic, and anti-genotoxic effects. Methods: The phytochemical composition was analyzed using spectrophotometric methods; all the biological endpoints were assessed through flow cytometry. Results: The phytochemical screening showed that polyphenols represent about 64% of the extract. Moreover, the extract was cytotoxic on Jurkat cells by inducing both apoptosis and necrosis and blocked the cell cycle in the G2/M phase. Additionally, it was found that the extract lacks any geno- toxic effect, but was not effective in protecting Jurkat cells from the DNA damage induced by H2O2 and etoposide. Conclusion: The results of our study show the toxic effects of Terminalia arjuna on Jurkat cells and confirm the pivotal role played by natural compounds in the oncological field. Further studies should be performed to better understand its clinical potential and deepen its toxicological profile

    Towards computing an optimal abstraction for structural causal models

    Working with causal model at different levels of abstraction is an important feature of science. Existing work has already considered the problem of expressing formally the relation of abstraction between causal models. In this paper, we focus on the problem of learning abstractions. We start by defining the learning problem formally in terms of the optimization of a standard measure of consistency. We then point out the limitation of this approach, and we suggest extending the objective function with a term accounting for information loss. We suggest a concrete measure of information loss, and we illustrate its contribution to learning new abstractions

    Agronomic strategies to enhance the early vigor and yield of maize. Part I: the role of seed applied biostimulant, hybrid and starter fertilization on rhizosphere bacteria profile and diversity

    The sustainable intensification of maize-based systems may reduce greenhouse-gas emissions and the excessive use of non-renewable inputs. Considering the key role that the microbiological fertility has on crop growth and resilience, it is worth of interest studying the role of cropping system on the rhizosphere bacterial communities, that affect soil health and biological soil fertility. In this work we monitored and characterized the diversity and composition of native rhizosphere bacterial communities during the early growth phases of two maize genotypes of different early vigor, using a nitrogen (N)-phosphorus (P) starter fertilization and a biostimulant seed treatment, in a growth chamber experiment, by polymerase chain reactiondenaturing gradient gel electrophoresis of partial 16S rRNA gene and amplicon sequencing. Cluster analyses showed that the biostimulant treatment affected the rhizosphere bacterial microbiota of the ordinary hybrid more than that of the early vigor, both at plant emergence and at the 5-leaf stage. Moreover, the diversity indices calculated from the community profiles, revealed significant effects of NP fertilization on richness and the estimated effective number of species (H2) in both maize genotypes, while the biostimulant had a positive effect on plant growth promoting community of the ordinary hybrid, both at the plant emergence and at the fifth leaf stage. Our data showed that maize genotype was the major factor shaping rhizosphere bacterial community composition suggesting that the root system of the two maize hybrids recruited a different microbiota. Moreover, for the first time, we identified at the species and genus level the predominant native bacteria associated with two maize hybrids differing for vigor. These results pave the way for further studies to be performed on the effects of cropping system and specific crop practices, considering also the application of biostimulants, on beneficial rhizosphere microorganisms

    Microdispersions of ellagic acid and pomegranate extracts as new potential nutraceutical ingredients

    The health properties attributed to several fruits (i.e. pomegranates, raspberries, strawberries, blackberry, chestnuts, walnuts, pecan), herbs (tea) and seeds (berries seeds) are attributed to an important group of natural polyphenols classified as hydrolysable tannins (HT) named Ellagitannins (ETs), that have shown in vitro multi-target biological properties relevant to the treatment of several human diseases. In vivo, ETs are rather not absorbed, and they are hydrolysed providing mainly Ellagic acid (EA). EA is endowed with the same biological properties of ETs and it could be considered as the responsible of their health benefits. Unfortunately, EA cannot be exploited for in vivo applications because of its poor water solubility (9.7 \u3bcg/mL) and accordingly low bioavailability. At first, aiming to increase EA solubility, an EA solid microdispersion (EA-md) was realized by employing only water and low methoxylated pectin, as a food compatible excipient, by applying spray drying technology. EA-md showed a 22% (w/w) Drug Loading (DL), a 30 times improved water solubility maintaining a remarkable radical scavenging activity [1]. It has been analytically characterised and used for in vivo pharmacological treatments in order to evaluate it as potential nutraceutical ingredient. Adult (3-6 months old) and old (20-22 old months) male mice were chronically administered EA-md dissolved in the drinking water (about 150 mg / Kg) for 14 days. During this period, animals were monitored for the spontaneous motor activity and for curiosity before, during and at the end of the EA-md treatment. Adult and old mice were then sacrificed for \u201cex vivo, in vitro\u201d analysis to test the efficiency of noradrenaline release from cortical nerve endings. It is known that noradrenaline exocytosis from cortical nerve endings is significantly impaired during ageing. We found that the chronic administration of EA-md did not alter the noradrenaline exocytosis from cortical nerve endings of adult mice, but significantly recovered the reduced noradrenaline overflow in aged mice. Further investigations are needed to explore the cellular cascade of events accounting for the beneficial effect. In a second step, pomegranate, as a natural source of EA, has been considered to similarly prepare and investigate an analogous formulation. Since pomegranate fruit is recognized as one of the most important sources of ETs, mainly localized in the by-products obtained after industrial juice squeezing, a method to convert the squeezing marcs into a potential nutraceutical ingredient has been explored. In particular, Pulsed Ultrasound-Assisted Extraction (PUAE), using just water as solvent, resulted to be suitable for extracting the water-soluble bioactive molecules (PEx), whose content in hydrolysable tannins, standardized in EA, has been determined. Furthermore, the already mentioned spray drying microdispersion has been employed to formulate and to stabilize it over time.     Native mycorrhizal communities in maize roots as affected by plant genotype, starter fertilization and a seed-applied biostimulant

    Background and aims: One of the most promising strategies for sustainable intensification of crop production involves the utilization of beneficial root-associated microorganisms, such as plant growth-promoting bacteria and arbuscular mycorrhizal fungi (AMF). The aim of this study was to investigate whether a seed-applied biostimulant, based on the bacterial strain Bacillus amyloliquefaciens IT-45 and a plant polysaccharide extract, and crop enhancement tools, such as hybrids with contrasting early vigor and nitrogen (N) plus phosphorus (P) starter fertilization, and their interactions, shape the communities of native root-colonizing AMF symbionts in maize. Methods: A factorial growth chamber experiment was set up with two maize genotypes in natural soil. Mycorrhizal colonization was evaluated after root staining. The diversity and composition of AMF communities were assessed by PCR-DGGE of the 18S rRNA gene and amplicon sequencing. Results: N and P fertilization determined a consistent reduction of AMF root colonization and, in combination of biostimulant, a reduction of AMF richness. The biostimulant alone generally did not affect AMF colonization or the community biodiversity. In addition the effect of the two factors were modulated by maize genotype. In all treatments, predominant AMF were represented by Glomus sp. and Funneliformis mosseae, while populations of the genus Rhizoglomus were rarely detected in biostimulant and NP fertilization treatments. Conclusion: The results of this study increase our understanding of how the biostimulant seed treatment may affect native AMF communities, depending on NP fertilization and maize genotype and may improve the implementation of innovative tools in sustainable and resilient agroecosystems

    Neuroinflammation in Aged Brain: Impact of the Oral Administration of Ellagic Acid Microdispersion

    The immune system and the central nervous system message each other to preserving central homeostasis. Both systems undergo changes during aging that determine central age-related defects. Ellagic acid (EA) is a natural product which is beneficial in both peripheral and central diseases, including aging. We analyzed the impact of the oral administration of a new oral ellagic acid micro-dispersion (EAm), that largely increased the EA solubility, in young and old mice. Oral EAm did not modify animal weight and behavioral skills in young and old mice, but significantly recovered changes in "ex-vivo, in vitro" parameters in old animals. Cortical noradrenaline exocytosis decreased in aged mice. EAm administration did not modify noradrenaline overflow in young animals, but recovered it in old mice. Furthermore, GFAP staining was increased in the cortex of aged mice, while IBA-1 and CD45 immunopositivities were unchanged when compared to young ones. EAm treatment significantly reduced CD45 signal in both young and old cortical lysates; it diminished GFAP immunopositivity in young mice, but failed to affect IBA-1 expression in both young and old animals. Finally, EAm treatment significantly reduced IL1beta expression in old mice. These results suggest that EAm is beneficial to aging and represents a nutraceutical ingredient for elders

    Efficacy of fingolimod and interferon beta-1b on cognitive, MRI, and clinical outcomes in relapsing-remitting multiple sclerosis: an 18-month, open-label, rater-blinded, randomised, multicentre study (the GOLDEN study)

    : Cognitive impairment (CI) affects 40-65% of multiple sclerosis (MS) patients. This study attempted evaluating the effects of fingolimod and interferon beta-1b (IFN ÎČ-1b) on CI progression, magnetic resonance imaging (MRI) and clinical outcomes in relapsing-remitting MS (RRMS) patients over 18 months. The GOLDEN study was a pilot study including RRMS patients with CI randomised (2:1) to fingolimod (0.5 mg daily)/IFN ÎČ-1b (250 Â”g every other day). CI was assessed via Rao's Brief Repeatable Battery and Delis-Kaplan Executive Function System test. MRI parameters, Expanded Disability Status Scale scores and relapses were measured. Overall, 157 patients were randomised, of whom 30 discontinued the study (fingolimod, 8.49%; IFN ÎČ-1b, 41.18%; p â‰€ 0.0001). Patients randomised to fingolimod had more severe clinical and MRI disease characteristics at baseline compared with IFN ÎČ-1b. At Month (M) 18, both treatment groups showed improvements in all cognitive parameters. At M18, relapse rate, total number and volume of T2/T1 gadolinium-enhancing lesions were higher with IFN ÎČ-1b, as well as the percentage brain volume change during the study. Safety and tolerability of both treatments were similar to previous studies. Both treatments showed improvements in cognitive parameters. Fingolimod demonstrated significantly better effects on MRI parameters and relapse rate. Imbalance in baseline characteristics and the drop-out pattern may have favoured IFN ÎČ-1b. A longer duration trial may be needed to observe the complete expression of differential effects on CI scales reflecting the between-groups differences on MRI. Although limited in size, the GOLDEN study confirms the favourable benefit-risk profile of fingolimod reported in previous studies

    We propose a multivariate statistical approach based on Principal Component Analysis (PCA) as an useful instrument to improve the Rules of Refinement and Reduction in in vivo animal experimentation. We analysed with PCA the preliminary data from a study on the effects of the oral administration of Tilia tomentosa bud extracts (TTBEs) on the behavioural skills of adult and aged male and female mice. PCA allows to rationalize the data set information and to dissect the results, showing connections among variables under study (behavioural parameters) and different trends in the experimental groups (control and TTBEs-administered animals). Our results show that PCA can give some important information that can be useful for the refinement of the experimental protocol, in order to reduce the number of the animals used in the experiments and/or the behavioural tests to get reliable information

    Analytical Challenges and Metrological Approaches to Ensuring Dietary Supplement Quality: International Perspectives

    The increased utilization of metrology resources and expanded application of its’ approaches in the development of internationally agreed upon measurements can lay the basis for regulatory harmonization, support reproducible research, and advance scientific understanding, especially of dietary supplements and herbal medicines. Yet, metrology is often underappreciated and underutilized in dealing with the many challenges presented by these chemically complex preparations. This article discusses the utility of applying rigorous analytical techniques and adopting metrological principles more widely in studying dietary supplement products and ingredients, particularly medicinal plants and other botanicals. An assessment of current and emerging dietary supplement characterization methods is provided, including targeted and non-targeted techniques, as well as data analysis and evaluation approaches, with a focus on chemometrics, toxicity, dosage form performance, and data management. Quality assessment, statistical methods, and optimized methods for data management are also discussed. Case studies provide examples of applying metrological principles in thorough analytical characterization of supplement composition to clarify their health effects. A new frontier for metrology in dietary supplement science is described, including opportunities to improve methods for analysis and data management, development of relevant standards and good practices, and communication of these developments to researchers and analysts, as well as to regulatory and policy decision makers in the public and private sectors. The promotion of closer interactions between analytical, clinical, and pharmaceutical scientists who are involved in research and product development with metrologists who develop standards and methodological guidelines is critical to advance research on dietary supplement characterization and health effects

    Sexual Dysfunction in People with Multiple Sclerosis: The Role of Disease Severity, Illness Perception, and Depression

    Despite being a common issue in people with multiple sclerosis (pwMS), sexual dysfunction is still underinvestigated. This work aims to assess the potential determinants of sexual dysfunction in pwMS by considering its relationship with disease severity (in terms of global disability), illness perception, and depressive symptoms. In this multicenter study, 1010 pwMS responded to an online survey. A serial mediation model considering negative illness perception and depressive symptoms as mediators of the relationship between disease severity and sexual dysfunction was conducted using the SPSS PROCESS Macro with bias-corrected bootstrapping (5000 samples). Disease severity exerts an indirect effect on sexual dysfunction via illness perception, both independently and through depressive symptoms. However, the results indicated that illness perception plays a more crucial role in sexual dysfunction in pwMS with mild disability than in pwMS with moderate-severe disability. This study suggests that higher disability increases its magnitude by enhancing negative illness perception, that, in turn, affects sexual dysfunction both directly and through depressive symptoms, especially in pwMS with mild disability. Modulating the effect of illness perception by favoring adaptive coping strategies might represent a valid approach to mitigate sexual dysfunction symptoms in MS
