A Public-Private Partnership vélt és valós hangsúlyai Magyarországon = Assumed and actual emphases of Public-Private Partnership in Hungary
ĂŤrásunk a Public-Private Partnership (PPP) magyarországi megvalĂłsulásának hangsĂşlyairĂłl szĂłl. Azt vizsgáljuk, hogy a szakmai közvĂ©lemĂ©ny milyen kĂ©pet alakĂthatott ki e projektekrĹ‘l a szaksajtĂł olvasásán keresztĂĽl. Három tágabb elvi szempont sajtĂłbeli megjelenĂ©sĂ©nek sĂşlyát elemezzĂĽk: a PPP cĂ©ljai, a feladatok hatĂ©konyabb elvĂ©gzĂ©sĂ©nek mĂłdja, illetve a társadalmi kontroll megvalĂłsulása áll Ă©rdeklĹ‘dĂ©sĂĽnk közĂ©ppontjában. A tartalomelemzĂ©s matematikai-statisztikai mĂłdszereit alkalmazzuk. KövetkeztetĂ©sĂĽnk, hogy a PPP-projektek általában mĂ©g nem elvi lĂ©nyegĂĽk szerint valĂłsultak meg Magyarországon, bár az Ă©vek során Ă©rzĂ©kelhetĹ‘ pozitĂv irányĂş elmozdulás: tanulási folyamat törtĂ©nik. This working paper focuses on the perceptions of the Hungarian implementation of Public-Private Partnership (PPP) projects. It was examined how PPP was perceived in the wider public through the communication of the most read daily and weekly business journals. We analyzed the proportion of three broad aspects in the articles: the main goals of PPP, efficiency improvement as a substantial opportunity for value creation, and the role of democratic control. We applied the mathematical-statistical methods of content analysis. Our conclusion is that PPP-projects in Hungary are far from the normative model of implementation as discerned from literature, but from year to year a positive shift, a learning process can be observed. Therefore, instead of discarding PPP as a progressive government solution, Hungary should focus on improving the design and implementation of PPP projects

Developmentally regulated expression of the BRI1 brassinosteroid receptor in Arabidopsis thaliana
Brassinosteroids (BRs) are important regulators of morphogenic events during plant development. The lack of active transport and well-characterized biosynthesis offer ideal conditions for studying the local and temporal effects of this hormone group. While recent studies have found clear coincidence between the sites of BR accumulation and organ differentiation, they have also provided evidence for developmental changes in hormone susceptibility. In order to investigate the role of the BR receptor BRI1 in the modulation of hormone sensitivity, we studied the time course and localization of BRI1 gene activity in Arabidopsis seedlings. To this end, we generated transgenic lines carrying BRI1 promoter-driven luciferase or GUS reporter genes and characterized the expression patterns of these chimeric genes. Our results showed increased BRI1 expression in dark grown seedlings, particularly in the elongation zone of the hypocotyl, and also at the sites of organ development in green seedlings. These data indicate that, in addition to local increases in the hormone level, the abundance of the receptor can also be instrumental in eliciting the BR response
A szkizofrĂ©nia diagnosztizálására alkalmas periferális genetikai marker azonosĂtása = Identification of a peripheral genetic marker suitable for diagnosis of schizophrenia
A szkizofrĂ©nia az egyik leggyakoribb neuropszichiátriai elváltozás, amely a human populáciĂł mintegy 1 %-át Ă©rinti. A szkizofrĂ©nia jelenlegi diagnĂłzisa komplex klinikai tĂĽnetek összetett Ă©rtĂ©kelĂ©sĂ©n alapul. A könnyen kimutathatĂł periferális molekuláris markerek használata alapvetĹ‘en segĂthetnĂ© a pszichiátriai betegsĂ©gek diagnĂłzisát. NemrĂ©g megjelent tanulmányok azt mutatják, hogy a dopamin receptorok D1 Ă©s D2 szubtĂpusai a periferális limfocitákban is kifejezĹ‘dnek. A dopamin receptor D3 (DRD3) gĂ©njĂ©nek tĂşlműködĂ©sĂ©t találták szkizofrĂ©n betegek limfocitáiban Ă©s javasolták a betegsĂ©g diagnosztikai markerĂ©nek. Jelen tanulmányunkban 13 drug naiv ill. drug free szkizofrĂ©n beteg limfocitáját vizsgáltuk meg, abbĂłl a cĂ©lbĂłl hogy a szkizofrĂ©nia további periferális markereit találjuk meg. Vizsgálataink nagy elĹ‘nye volt, hogy olyan betegek vĂ©rĂ©t vizsgáltuk, akik nem álltak gyĂłgyszeres kezelĂ©s alatt. Ez kizárta annak a lehetĹ‘sĂ©gĂ©t, hogy gyĂłgyszerek hatását vizsgáljuk, a betegsĂ©g által okozott gĂ©nexpressziĂłs változások helyett. DNS chip vizsgálattal a dopamin receptor 2 (DRD2) Ă©s egy kálium csatorna gĂ©n (az inwardly rectifiying potassium channel , Kir2.3) fokozott expressziĂłját állapĂtottuk meg. A megnövekedett mRNS szintet kvantitativ real time PCR- val (QRT-PCR) erĹ‘sĂtettĂĽk meg, SybrGreen festĂ©ket Ă©s kettĹ‘sen jelölt Taqman prĂłbákat használva. MindkĂ©t molekuláris marker használata a szkizofrĂ©nia gyorsabb Ă©s pontosabb diagnĂłzisát teszi lehetĹ‘vĂ©, Ă©s segĂthet az optimális gyĂłgykezelĂ©s kidolgozásában. | Schizophrenia is one of the most common neuropsychiatric disorders affecting nearly 1% of human population. Current diagnosis of schizophrenia is based on complex clinical symptoms. Peripheral molecular markers that can be easily analyzed could substantially help in the diagnosis of psychiatric disorders. Recent studies showed that peripheral blood lymphocytes (PBL) express subtypes of D1 and D2 subclass of dopamine receptors. Dopamine D3 (DRD3) receptor was found to be overexpressed in schizophrenic PBL by quantitative RT-PCR and proposed to be a diagnostic and follow-up marker for schizophrenia. In this study we screened drug-naive and drug-free schizophrenic PBL to identify additional markers that are differentially expressed compared to healthy individuals using microarray and quantitative real-time PCR (QRT-PCR) techniques. We analyzed lymphocytes of 7 female and 6 male drug-free or drug-naive schizophrenic patients and 5-5 sex-matched control individuals. Among others, genes for dopamine D2 receptor (DRD2) and inwardly rectifying potassium channel (Kir2.3) were found to be overexpressed in microarray analysis. Increased mRNA levels were confirmed by QRT-PCR using SybrGreen method and dual labeled TaqMan probes. Use of both molecular markers enable us for more rapid and precise prediction of schizophrenia and might help follow the efficiency of drugs in therapy in order to provide a more tailored medication for schizophrenic patients

A támogatási idĹ‘szak során a brasszinoszteroidok (BR-ok, növĂ©nyi szteroid hormonok) bioszintĂ©zisĂ©ben rĂ©sztvevĹ‘ gĂ©nek funkciĂłit Ă©s szabályozását vizsgáltuk Arabidopsis modellnövĂ©nyben. Molekuláris genetikai mĂłdszerek segĂtsĂ©gĂ©vel meghatároztuk a CYP90C1 Ă©s CYP90D1 gĂ©nek által kĂłdolt ezimek szerepĂ©t a hormon szintĂ©zisĂ©ben, Ă©s eredmĂ©nyeink alapján Ăşj bioszintĂ©zis modellt dolgoztunk ki. ElvĂ©geztĂĽk a BR szintĂ©zis kulcsenzimeit kĂłdolĂł CPD/CYP90A1 Ă©s CYP85A2 gĂ©nek kifejezĹ‘dĂ©sĂ©nek rĂ©szletes vizsgálatát, Ă©s kimutattuk, hogy expressziĂłs szintjĂĽk emelkedĂ©se az aktĂv hormon felhalmozĂłdását eredmĂ©nyezi. E kĂ©t gĂ©n napszakos szabályozásának tanulmányozása során bizonyĂtottuk a fĂ©ny induktĂv szerepĂ©t, Ă©s tisztáztuk hogy a fĂ©nyhatás közvetĂtĂ©sĂ©ben a fitokrĂłm B fotoreceptorrĂłl kiindulĂł szignálátviteli Ăştnak van meghatározĂł jelentĹ‘sĂ©ge. | In the frame of the current project we studied the functions and regulation of the genes that are involved in the biosynthesis of brassinosteroids (BRs, plant steroid hormones) in Arabidopsis. Using molecular genetic approaches, we identified the reactions catalyzed by the CYP90C1 and CYP90D1 genes, and based on these results we revised the existing model of the BR biosynthetic pathway. We carried out a detailed expressional analysis of the CPD/CYP90A1 and CYP85A2 genes that encode rate limiting enzymes of BR synthesis, and demonstrated that their increased activity results in the accumulation of the bioactive hormone. These results strongly suggest a key role for the transcriptional regulation of BR-biosynthetic genes in the setting of local BR levels. Studies of the diurnal regulation of these genes revealed the inductive role of light, and we determined that this light control depends primarily on signals originating from the phytochrome B photoreceptor.
