    Sequences Analysis of a Gene Encoding Extracellular Xylanase in Streptomyces costaricanus 45I-3

    Streptomyces costaricanus 45I-3 is a bacterial strain belongs to actinomycetes group isolated from peat soil. The bacterium is known to produce extracellular xylanase. The aims of this study were to analyze DNA sequence and sub-clone gene involved in the synthesis of extracellular xylanase. Complete DNA sequence predicted to encode xylanase genes was isolated from bacterial genome using Inverse Polymerase Chain Reaction (I-PCR). Total DNA sequence of 1664 bp in size obtained from I-PCR consisted of two open reading frames (ORF) in opposite direction. ORF1 was 1029 bp and ORF2 (partial sequence) was 309 bp. Analysis sequence using BlastX indicated that ORF1 was homologous with xylanase bacterium enrichment culture clone Xyl8B8 (GenBank accession No. AFH35005.1), i.e. 95% in identity and 99% in similarity. In addition, ORF2 was homologous with glyoxalase bacterium enrichment culture clone Xyl8B8 (GenBank accession No. AFH35007.1), i.e. 95% in identity and 98% in similarity. Analysis of amino acid sequence revealed that ORF1 consisted of 2 domains, i.e. glyco-hydrolase 11 (GH11) and Carbohydrate Binding Type 2 (CBM2). Active site was found at 130th amino acid on GH11 domain. Visualization of 3-dimension structure showed that 1029 bp fragment is of 19 areas

    Studi Keanekaragaman Mikroalga di Sungai dan Danau Bengkulu sebagai Bioindikator Perairan

    This exploratory study aims to analyze the water quality of Selagan River and Tes Lake based on diversity of microalgae. Sampling was done by systematic purposive sampling. Results of study found 38 species (8 classes) of microalgae in Selagan River and 48 species (9 classes) in Tes Lake. The bundance of microalgae at the upstream, middle, and downstream stations of Selagan River were 3714 ind/ml, 105 ind/ml, and 315 ind/ml, with diversity index 0.25; 1.93; and 1.72. The microalgae dominance index in river is 0.95; 0.58; and 0.75 low to high criteria. The evenness index value for each station is 0.09; 0.78; and 0.63 low to high categories. Meanwhile, abundance of microalgae in Tes Lake at the inlet, middle, and outlet stations were 192 ind/ml, 204 ind/ml, and 200 ind/ml, with  diversity index of 2.70; 2.55; and 2.05. The microalgae dominance index was 0.10; 0.28; and 0.2 low category. The evenness index value for each station is 0.78; 0.75; 0.64 high category. Conclusion, condition of Selagan River at the upstream station is classified as heavily polluted, the middle and downstream stations are classified as clean. Meanwhile, condition of Tes Lake is clean and shows normal tolerance range for microalgae life

    The aim of this study is to explore the impact of interactive-conceptual-instruction with visualization on logic thinking, conceptual understanding and spiritual aspect embedding. The sampling technique used in this research was convenience sampling. This research used mixed methods with embedded experiment design. The data of logic thinking and conceptual understanding was collected through pre-test and post-test. The questionnaires were used to collect data on spiritual attitudes and were analyzed qualitatively. The results showed that logic thinking-ability improves from no logic thinking to inductive-deductive logic thinking. The improvement of logic thinking abilities was in the medium category. Understanding the concept shifted from incompetent to conceptual understanding with improvement, in the medium category. The questionnaires analysis showed that the students got spiritual values related to the awareness of God believing behind the natural phenomena that impact on their spiritual attitudes. This study also tried to apply the results of pure science research to support learning activities. Based on the result of the research, it can be concluded that the learning process using interactive conceptual with visualization can improve logic thinking, conceptual comprehension, and spiritual attitude embeding

    AQUAPONIC APPLICATION OF ORGANIC VEGETABLES-CATFISH IN BUCKETS (ASOILELEDAMBER) IN BENGKULU CITY. Bengkulu City is the capital of Bengkulu Province with the highest population in Bengkulu Province so it requires the availability of high food consumption as well. Required foodstuffs such as vegetables, meat, and fish, not all are available from Bengkulu City, so it is not uncommon the price on the market becomes more expensive. To fulfill the needs of foodstuffs, some people of Bengkulu City make effort to cultivate vegetables and maintain fish in ponds, but this method is also difficult to do because of the limited land area. Based on the description, it is necessary to apply the Appropriate Technology (TTG) for the people of Bengkulu City to fulfill family food security, one of which is the application of aquaponic technology: Asoileledamber (Organic Vegetable-Catfish Aquaponics in Buckets). This community service activity aims to provide information and training to the group of service partners in Bengkulu City about the benefits of Asoileledamber. Community service activities include Asoileledamber counseling activities, the provision of guidebooks, provision of tools and materials for cultivation such as fish seedlings, vegetable seeds and pellet feed. The results perceived by the service partners were harvesting Catfish from the second month of maintenance to the fourth month of maintenance. Organic vegetables are harvested starting from 2 weeks after planting time (SMT) until the 4th month of maintenance with an average harvest of 1-2 bunches per bucket

    SOCIALIZATION OF COVID-19 AND PRACTICE OF MAKING DISINFECTANT LIQUIDS FOR FLOOR MOP MATERIALS IN ISLAMIC MODERN BOARDING SCHOOL OF DARUSALAM, KEPAHIANG REGENCY, BENGKULU PROVINCE. Floor cleaner liquid is a disinfectant used to prevent the transmission of environmental pathogenic microorganisms, especially on the floors. The high price of floor cleaner in the market and difficult to find in the condition of the current Covid-19 outbreak, make this community service activity need to be done and it aims to overcome this problem by holding out socialization about Covid-19 and the practice of making disenfectant fluids as floor cleaning materials at the Islamic Modern Boarding School of Darussalam, Kepahiang Regency. It is expected, by this activity can create a small business to manufacture floor mop from disinfectant with good management in this school. Community Service Activities (PPM) were well received by the students and the teaching staff of the Islamic Modern Boarding School of Darussalam. Before giving service materials to community service participants, the participants or students did not really know what was the cause and prevention of Covid-19 in the surrounding environment and how to make disinfectants. The giving of the pre-test stage questionnaire after the end of the socialization was considered as feedback for participants resulting in an increase in understanding of Covid-19, increasing awareness of the importance of maintaining clean floors and understanding of how to make disinfectant fluids as floor mops made from sodium hypochlorite

    BAKTERI PENGHASIL AMILASE YANG DIISOLASI DARI EKOENZIM LIMBAH BUAH-BUAHAN

    Many of potential enzymes can be found in the ecoenzyme, one of them is amylase. Amylase is an enzyme that is able to hydrolize the glycoside bonds of starch or starch into dextrin, glucose and maltose which is widely used in various industries such as beverages. This study aims to isolate the ecoenzyme bacteria and test their ability to produce amylase. Isolation of ecoenzyme bacteria was carried out using serial dilution methods 10-1, 10-3, 10-5. A total of 0.1 ml of each dilution series was pipetted and spread on the Nutrient agar (NA) medium. The bacterial isolates that grew were then purified and identified by morphological observation, Gram staining and their enzymatic activity was tested using Starch Agar (SA) media qualitatively. The results of this study showed there were 39 bacterial isolates from ecoenzyme with different morphological characteristics. The amylase activity test were found that 34 isolates had a positive activity to hydrolize the starch in the SA media which was indicated by the formation of a clear zone around the bacterial colonies. Each bacterial isolate had a different hydrolysis index value, which ranged from 9.45 to 23.65. The highest clear zone diameter index value from starch hydrolysis was EJM 15 isolate. &nbsp
