    Antimicrobial activity of some ethnomedicinal plants used by Paliyar tribe from Tamil Nadu, India

    BACKGROUND: Antimicrobial activity of 18 ethnomedicinal plant extracts were evaluated against nine bacterial strains (Bacillus subtilis, Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus faecalis, Escherichia coli, Klebsiella pneumonia, Pseudomonas aeruginosa, Ervinia sp, Proteus vulgaris) and one fungal strain (Candida albicans). The collected ethnomedicinal plants were used in folk medicine in the treatment of skin diseases, venereal diseases, respiratory problems and nervous disorders. METHODS: Plants were collected from Palni hills of Southern Western Ghats and the ethnobotanical data were gathered from traditional healers who inhabit the study area. The hexane and methanol extracts were obtained by cold percolation method and the antimicrobial activity was found using paper disc diffusion method. All microorganisms were obtained from Christian Medical College, Vellore, Tamil Nadu, India. RESULTS: The results indicated that out of 18 plants, 10 plants exhibited antimicrobial activity against one or more of the tested microorganisms at three different concentrations of 1.25, 2.5 and 5 mg/disc. Among the plants tested, Acalypha fruticosa, Peltophorum pterocarpum, Toddalia asiatica,Cassia auriculata, Punica granatum and Syzygium lineare were most active. The highest antifungal activity was exhibited by methanol extract of Peltophorum pterocarpum and Punica granatum against Candida albicans. CONCLUSION: This study evaluated the antimicrobial activity of the some ethnomedicinal plants used in folkloric medicine. Compared to hexane extract, methanol extract showed significant activity against tested organisms. This study also showed that Toddalia asiatica, Syzygium lineare, Acalypha fruticosa and Peltophorum pterocarpum could be potential sources of new antimicrobial agents

    Medicinal plants used by traditional healers in Kancheepuram District of Tamil Nadu, India

    An ethnobotanical survey was undertaken to collect information from traditional healers on the use of medicinal plants in Kancheepuram district of Tamil Nadu during October 2003 to April 2004. The indigenous knowledge of local traditional healers and the native plants used for medicinal purposes were collected through questionnaire and personal interviews during field trips. The investigation revealed that, the traditional healers used 85 species of plants distributed in 76 genera belonging to 41 families to treat various diseases. The documented medicinal plants were mostly used to cure skin diseases, poison bites, stomachache and nervous disorders. In this study the most dominant family was Euphorbiaceae and leaves were most frequently used for the treatment of diseases. This study showed that many people in the studied parts of Kancheepuram district still continue to depend on medicinal plants at least for the treatment of primary healthcare. The traditional healers are dwindling in number and there is a grave danger of traditional knowledge disappearing soon since the younger generation is not interested to carry on this tradition

    Molecular Docking of Flindersine with some targets related to β-cells Protection

    Diabetes mellitus (DM) is the most widespread metabolic disorder affecting millions worldwide. Molecular docking studies are useful in identifying some useful ligands which could be used to target proteins related to β-cell protection.  Flindersine isolated from the plant Toddalia asiatica (L.) Lam. (Rutaceae) has been shown by us to possess antidiabetic property. With a view to identify in silico the possible mode of docking with different target proteins like PPARγ and GLUT4 which play important roles in protecting β-cells from damage. Chemical characteristics of Flindersine were retrieved from pubchem database http://pubchem.ncbi.nlm.nih.gov. The docking analysis in the active sites of 2PRG and Homology modeled protein structure of GLUT4 were performed by the Auto dock program. The docking results showed good binding interactions of the ligand with both the targets at very low energy level. In our in silico analysis, flindersine isolated from Toddalia asiatica clearly demonstrated that it could improve  diabetic condition by increasing insulin secretion from remnant or regenerated pancreatic beta cells and could promote insulin sensitization and glucose uptake activities. When compared with standard drug Rosiglitazone that is commercially available flindersine can further diminish the degree of shrinkage and necrosis of beta cells of pancreas. Thus flindersine can be considered for developing into a potent antidiabetic drug

    Larvicidal, ovicidal and repellent activities of marine sponge Cliona celata (Grant) extracts against Anopheles stephensi Liston (Diptera: Culicidae)

    AbstractObjectiveTo evaluate the larvicidal, ovicidal and repellent properties of solvent extracts of marine sponge Cliona celata (C. celata) (Grant) against the malarial vector Anopheles stephensi (An. stephensi) Liston.MethodsMarine sponge C. celata was thoroughly washed with distilled water and shade dried for 48 h. Then the sponges were homogenized and extracted sequentially with hexane, ethyl acetate and methanol. Larvicidal and ovicidal activities were tested at four different concentrations viz., 62.5, 125.0, 250.0 and 500.0 ppm. For repellent study extracts were taken in three different concentrations viz., 5.0, 2.5, 1.0 mg/cm at.ResultsAmong the three solvent extracts of C. celata, methanol extract showed the highest larvicidal activity at 500 ppm against the fourth instar larvae of An. stephensi. The LC50 and LC90 values of C. celata methanol extract were recorded as 80.61 and 220.81 ppm against An. stephensi larvae respectively. High ovicidal activity of 91.2% was recorded at 500 ppm concentration of methanol extract. The haxane extract was found to be the most effective protectant against the adult female mosquitoes of An. stephensi. The mean protection time recorded in hexane extract was up to 245 min at 5 mg/cm2 dosage against An. stephensi adults.ConclusionsThe screening results suggest that the hexane and methanol extracts of C. celata are promising in mosquito control. Considering these bioactivities, C. celata could be probed further to obtain some novel pesticidal molecules

    Flavonoids are plant secondary metabolites. They are mainly classified into four major groups, such as flavanols, flavones, anthocyanidins, and isoflavonoids. Furthermore, they are divided into some subclasses. They are available in dietary foods and they cure various diseases. Certain plants and spices contain flavonoids, which have been commonly used for thousands of years in traditional medicine. Some of the flavonoids have been clinically used in many countries. Baicalein and its glycosides are one among them to have been experimented clinically. Flavonoids have the capability to regulate cell division and proliferation in an important pathway. They have medicinal activities including anticancer properties. The isoflavone analog rotenone is one of the flavonoid compounds, which has been revealed to be actual anticancer agent. Scutellaria species having flavones retain cytotoxic activities against many human cancer cell lines. At the same time, they do not harm the myeloid cells, normal peripheral and normal epithelial blood cells.     Hexane, chloroform and ethyl acetate extracts of the leaves of Atalantia monophylla (L.) Correa. were evaluated for their antifeedant and larvicidal activities along with their effect on pupation and adult emergence against bhendi fruit borer, Earias vittella Fab. at 5, 2.5 and 1.25% concentrations. Commercial botanical insecticide, Vijay Neem was used as positive control. Hexane extract at 5% concentration showed significant antifeedant and larvicidal activity of 70.89 and 85.33% respectively. A reduction of 16.66% in adult emergence was recorded in hexane extract followed by 25% reduction in ethyl acetate extract at 5% concentration. The results revealed that hexane extract showed significant antifeedant, and larvicidal activity and good reduction in adult emergence. This extract could be further investigated to isolate the active principles

    Antimicrobial, antimycobacterial and antibiofilm properties of Couroupita guianensis Aubl. fruit extract

    BACKGROUND: Couroupita guianensis Aubl. (Lecythidaceae) is commonly called Ayahuma and the Cannonball tree. It is distributed in the tropical regions of northern South America and Southern Caribbean. It has several medicinal properties. It is used to treat hypertension, tumours, pain, inflammatory processes, cold, stomach ache, skin diseases, malaria, wounds and toothache. METHODS: The fruits of Couroupita guianensis were extracted with chloroform. Antimicrobial, antimycobacterial and antibiofilm forming activities of the chloroform extract were investigated. Quantitative estimation of Indirubin, one of the major constituent, was identified by HPLC. RESULTS: Chloroform extract showed good antimicrobial and antibiofilm forming activities; however it showed low antimycobacterial activity. The zones of inhibition by chloroform extract ranged from 0 to 26 mm. Chloroform extract showed effective antibiofilm activity against Pseudomonas aeruginosa starting from 2 mg/mL BIC, with 52% inhibition of biofilm formation. When the chloroform extract was subjected to HPLC-DAD analysis, along with Indirubin standard, in the same chromatographic conditions, it was found that Indirubin was one of the major compounds in this plant (0.0918% dry weight basis). CONCLUSIONS: The chloroform extract showed good antimicrobial and antibiofilm properties. Chloroform extract can be evaluated further in drug development programmes

    Antimicrobial activity of some actinomycetes from Western Ghats of Tamil Nadu, India

    Introduction: Microbial diseases are increasing year by year and they are becoming a big threat to public health. There are more than 200 known diseases transmitted by bacteria, fungi, viruses, prions, rickettsia and other microbes to humans. The emergence of drug resistance to chemical drugs is the biggest threat in controlling human pathogens. Hence novel antimicrobial agents from actinomycetes are timely needed for the control of several human pathogens.Aim: The aim was to find some actinomycetes with antimicrobial metabolites.Methods: Soil samples were collected from Nilgiris district in Western Ghats of Tamil Nadu, India. Actinomycetes were isolated using serial dilution and plating techniques on actinomycetes isolation agar. Streptomycin and ketoconazole (25 lg/disc) were used as reference controls. The active strains were identified by 16S rRNA and phylogenetic tree was constructed; the sequences were submitted in the GenBank.Results: Totally 106 actinomycete strains were isolated and cross streaked against various human microbial pathogens. Only 44 (41.50%) exhibited good antimicrobial activity against different pathogenic microbes. Five isolates (FMS-20, TGH-30, TGH-31, TGH-31-1 and IS-4) were chosen for secondary screening using filtrate. Among them FMS-20 filtrate showed good inhibition on the 16th day against all tested microbial pathogens. Further the intracellular methanol extract of FMS- 20 showed maximum zone of inhibition against A. brasiliensis (22 mm) at 5 mg/disc. Similarly the extracellular ethyl acetate extract of FMS-20 showed maximum zone of inhibition against B. subtilis (25 mm).Conclusions: The present work revealed that, among 106 actinomycetes screened, Streptomyces rimosus (FMS-20) (Accession No-KT827106) showed promising antimicrobial activity against all the tested human microbial pathogens.Keywords: Streptomyces rimosus; Antimicrobial activity; Disc diffusion; Zone of inhibitio
