Expressão gênica em grãos de café submetidos a diferentes tratamentos pós-colheita-uma análise preliminar.
O conhecimento sobre como os tipos de processamento pós-colheita interferem na qualidade final de bebida é essencial para que cafés com qualidades especiais possam ser desenvolvidos. Dentre as características que podem ser diretamente influenciadas pelo processamento, a composição química do grão é a de maior interesse. Fatores como a regulação da expressão de genes e a atividade de enzimas específicas são determinantes para esta composição. Neste contexto, o objetivo deste estudo preliminar foi avaliar como os processamentos por via seca e úmida afetam o padrão de expressão dos genes isocitrato liase (ICL), ?-mananase (MAN) e ?-galactosidase (GAL). Frutos cereja foram submetidos a condições de secagem natural, despolpamento e desmucilamento. A expressão dos genes foi quantificada pela metodologia de RT-PCR em tempo real. A análise comparativa da expressão dos 3 genes indicou que no processamento via seca a expressão de ICL e MAN nas sementes foi maior do que em sementes de frutos processados por via úmida. Para o gene GAL, não foram observadas diferenças expressivas no perfil de expressão em sementes dos frutos submetidos aos diferentes tratamentos. Esses resultados diferiram dos obtidos em outros estudos já descritos na literatura, em que, apesar de ter sido avaliada a expressão de genes, foram utilizadas etapas de processamento distintas das utilizadas neste trabalho. Esses resultados revelaram, portanto, que há necessidade de se padronizarem os processamentos experimentais em pesquisas visando avaliar efeitos dos tratamentos pós-colheita sobre a qualidade do café
Machine-Readable Privacy Certificates for Services
Privacy-aware processing of personal data on the web of services requires
managing a number of issues arising both from the technical and the legal
domain. Several approaches have been proposed to matching privacy requirements
(on the clients side) and privacy guarantees (on the service provider side).
Still, the assurance of effective data protection (when possible) relies on
substantial human effort and exposes organizations to significant
(non-)compliance risks. In this paper we put forward the idea that a privacy
certification scheme producing and managing machine-readable artifacts in the
form of privacy certificates can play an important role towards the solution of
this problem. Digital privacy certificates represent the reasons why a privacy
property holds for a service and describe the privacy measures supporting it.
Also, privacy certificates can be used to automatically select services whose
certificates match the client policies (privacy requirements).
Our proposal relies on an evolution of the conceptual model developed in the
Assert4Soa project and on a certificate format specifically tailored to
represent privacy properties. To validate our approach, we present a worked-out
instance showing how privacy property Retention-based unlinkability can be

Composição química de café conilon (Coffea canephora).
O objetivo neste trabalho foi estudar a composição química de um grupo de clones de Coffea canephora pertencentes ao programa de pesquisa em melhoramento genético da espécie no Instituto Capixaba de Pesquisa e Extensão Rural - Incaper, no Estado do Espírito Santo. Foram avaliados, em 49 clones, os teores de cafeína, trigonelina, ácidos clorogênicos e sólidos solúveis totais. Verificou-se a existência de expressiva variação entre os diferentes clones analisados, com valores extremos variando entre de 1,51 a 2,64% para cafeína; de 0,64 a 1,30% para trigonelina; de 3,61 a 5,64% para ácidos clorogênicos e de 29,36 a 36,36% para sólidos solúveis. Esses resultados indicam a existência de variabilidade genética para as características estudadas, bem como a possibilidade de seleção de plantas possuidoras de características qualitativas de maior interesse comercial
Dietary Supplementation with Probiotics Improves Hematopoiesis in Malnourished Mice
BACKGROUND: Lactobacillus rhamnosus CRL1505 (Lr) administered during the repletion of immunocompromised-malnourished mice improves the resistance against intestinal and respiratory infections. This effect is associated with an increase in the number and functionality of immune cells, indicating that Lr could have some influence on myeloid and lymphoid cell production and maturation. OBJECTIVE: This study analyzed the extent of the damage caused by malnutrition on myeloid and lymphoid cell development in the spleen and bone marrow (BM). We also evaluated the impact of immunobiotics on the recovery of hematopoiesis affected in malnourished mice. METHODS: Protein malnourished mice were fed on a balanced conventional diet for 7 or 14 consecutive d with or without supplemental Lr or fermented goat's milk (FGM). Malnourished mice and well-nourished mice were used as controls. Histological and flow cytometry studies were carried out in BM and spleen to study myeloid and lymphoid cells. RESULTS: Malnutrition induced quantitative alterations in spleen B and T cells; however, no alteration was observed in the ability of splenic B cells to produce immunoglobulins after challenge with LPS or CpG. The analysis of BM B cell subsets based on B220, CD24, IgM and IgD expression showed that malnutrition affected B cell development. In addition, BM myeloid cells decreased in malnourished mice. On the contrary, protein deprivation increased BM T cell number. These alterations were reverted with Lr or FGM repletion treatments since normal numbers of BM myeloid, T and B cells were observed in these groups. CONCLUSIONS: Protein malnutrition significantly alters B cell development in BM. The treatment of malnourished mice with L. rhamnosus CRL1505 was able to induce a recovery of B cells that would explain its ability to increase immunity against infections. This work highlights the possibility of using immunobiotics to accelerate the recovery of lymphopoyesis in immunocompromised-malnourished hosts
The erythropoietin receptor expressed in skeletal muscle is essential for mitochondrial biogenesis and physiological exercise
Erythropoietin (EPO) is a haematopoietic hormone that regulates erythropoiesis, but the EPO-receptor (EpoR) is also expressed in non-haematopoietic tissues. Stimulation of the EpoR in cardiac and skeletal muscle provides protection from various forms of pathological stress, but its relevance for normal muscle physiology remains unclear. We aimed to determine the contribution of the tissue-specific EpoR to exercise-induced remodelling of cardiac and skeletal muscle. Baseline phenotyping was performed on left ventricle and m. gastrocnemius of mice that only express the EpoR in haematopoietic tissues (EpoR-tKO). Subsequently, mice were caged in the presence or absence of a running wheel for 4 weeks and exercise performance, cardiac function and histological and molecular markers for physiological adaptation were assessed. While gross morphology of both muscles was normal in EpoR-tKO mice, mitochondrial content in skeletal muscle was decreased by 50%, associated with similar reductions in mitochondrial biogenesis, while mitophagy was unaltered. When subjected to exercise, EpoR-tKO mice ran slower and covered less distance than wild-type (WT) mice (5.5 ± 0.6 vs. 8.0 ± 0.4 km/day, p < 0.01). The impaired exercise performance was paralleled by reductions in myocyte growth and angiogenesis in both muscle types. Our findings indicate that the endogenous EPO-EpoR system controls mitochondrial biogenesis in skeletal muscle. The reductions in mitochondrial content were associated with reduced exercise capacity in response to voluntary exercise, supporting a critical role for the extra-haematopoietic EpoR in exercise performance. Converging evidence from different species indicates that some newborn vertebrates, including humans, have visual predispositions to attend to the head region of animate creatures. It has been claimed that newborn preferences for faces are domain-relevant and similar in different species. One of the most common criticisms of the work supporting domain-relevant face biases in human newborns is that in most studies they already have several hours of visual experience when tested. Search for new phenomena with the MT2 variable in the all-hadronic final state produced in proton-proton collisions at s=13 TeV.
A search for new phenomena is performed using events with jets and significant transverse momentum imbalance, as inferred through the MT2 variable. The results are based on a sample of proton-proton collisions collected in 2016 at a center-of-mass energy of 13 TeV with the CMS detector and corresponding to an integrated luminosity of 35.9 fb-1 . No excess event yield is observed above the predicted standard model background, and the results are interpreted as exclusion limits at 95% confidence level on the masses of predicted particles in a variety of simplified models of R-parity conserving supersymmetry. Depending on the details of the model, 95% confidence level lower limits on the gluino (light-flavor squark) masses are placed up to 2025 (1550) GeV . Mass limits as high as 1070 (1175) GeV are set on the masses of top (bottom) squarks. Information is provided to enable re-interpretation of these results, including model-independent limits on the number of non-standard model events for a set of simplified, inclusive search regions
