    Effect of Intravenously Applied Furosemide on Endocochlear DC Potential and Cochlear Microphonics Correlated with Structural Changes in Guinea Pigs

    In the scala media, the DC potential of about +80 mV exists between endolymph and neighboring tissue of the earth potential. Furosemide, which is a strong diuretics, is known to inhibit Na-Cl cotransport across the luminar cell of the thick ascending limb of the Henle loop and it produces the changes in cochlear functions. In this study, furosemide was applied systematically at the dosages of 80 mg/kg, 40 mg/kg and 20 mg/kg. Depending upon the doses, furosemide reduced the DC potential to 0 mV or even negative potential and decreased the amplitude of the cochlear microphonics. These changes started within a few min after application of the drug, were maintained another a few min and then recovered completely to the initial level within an hour or so. It is noteworthy to describe that microphonics recovers faster than DC potential. Electronmicroscopic investigation indicated that the shrinkage and projection took place in the cells of stria vascularis. Action of furosemide is explained from the blockage Na-Cl co-transport across the endocochlear side of the marginal cell membrane and, consequently, blockage Na-K pump at the opposite side of the cell membrane

    Theoretical study on novel electronic properties in nanographite materials

    Antiferromagnetism in stacked nanographite is investigated with using the Hubbard-type model. We find that the open shell electronic structure can be an origin of the decreasing magnetic moment with the decrease of the inter-graphene distance, as experiments on adsorption of molecules suggest. Next, possible charge-separated states are considered using the extended Hubbard model with nearest-neighbor interactions. The charge-polarized state could appear, when a static electric field is present in the graphene plane for example. Finally, superperiodic patterns with a long distance in a nanographene sheet observed by STM are discussed in terms of the interference of electronic wave functions with a static linear potential theoretically. In the analysis by the k-p model, the oscillation period decreases spatially in agreement with experiments.Comment: 8 pages; 6 figures; accepted for publication in J. Phys. Chem.     Effect of enhanced expression of connexin 43 on sunitinib-induced cytotoxicity in mesothelioma cells

    AbstractConnexin (Cx) makes up a type of intercellular channel called gap junction (GJ). GJ plays a regulatory role in cellular physiology. The Cx expression level is often decreased in cancer cells compared to that in healthy ones, and the restoration of its expression has been shown to exert antiproliferative effects. This work aims to evaluate the effect of the restoration of connexin 43 (Cx43) (the most ubiquitous Cx subtype) expression on sunitinib (SU)-induced cytotoxicity in malignant mesothelioma (MM) cells. Increased Cx43 expression in an MM cell line (H28) improved the ability of SU to inhibit receptor tyrosine kinase (RTK) signaling. Moreover, higher Cx43 expression promoted SU-induced apoptosis. The cell viability test revealed that Cx43 enhanced the cytotoxic effect of SU in a GJ-independent manner. The effect of Cx43 on a proapoptotic factor, Bax, was then investigated. The interaction between Cx43 and Bax was confirmed by immunoprecipitation. Furthermore, higher Cx43 expression increased the production of a cleaved (active) form of Bax during SU-induced apoptosis with no alteration in total Bax expression. These findings indicate that Cx43 most likely increases sensitivity to SU in H28 through direct interaction with Bax. In conclusion, we found that Cx43 overcame the chemoresistance of MM cells
