    Modifications at the C-Terminus To Improve Pyrrole−Imidazole Polyamide Activity in Cell Culture

    Pyrrole−imidazole (Py-Im) hairpin polyamides are a class of small molecule DNA minor groove binding compounds that have been shown to modulate endogenous gene expression in cell culture. Gene regulation by polyamides requires efficient cellular uptake and nuclear localization properties for candidate compounds. To further optimize Py-Im polyamides for enhanced potency in cell culture, a focused library of polyamides possessing various modifications at the C-terminus was synthesized and tested. Comparison of polyamide biological activity in two cell lines revealed tolerance for structural modifications and agreement in activity trends between cell lines. The use of an oxime linkage between the polyamide and an aromatic functionality on the C-terminus resulted in a ~20-fold increase in the potency of polyamides targeted to the androgen response element (ARE) in LNCaP cells by measuring AR-activated PSA expression

    On the coupling between an ideal fluid and immersed particles

    In this paper we use Lagrange-Poincare reduction to understand the coupling between a fluid and a set of Lagrangian particles that are supposed to simulate it. In particular, we reinterpret the work of Cendra et al. by substituting velocity interpolation from particle velocities for their principal connection. The consequence of writing evolution equations in terms of interpolation is two-fold. First, it gives estimates on the error incurred when interpolation is used to derive the evolution of the system. Second, this form of the equations of motion can inspire a family of particle and hybrid particle-spectral methods where the error analysis is "built-in". We also discuss the influence of other parameters attached to the particles, such as shape, orientation, or higher-order deformations, and how they can help with conservation of momenta in the sense of Kelvin's circulation theorem.Comment: to appear in Physica D, comments and questions welcom

    Approximate theoretical performance evaluation for a diverging rocket

    A simplified combustion model, which is motivated by available performance studies on the diverging rocket reactor, has been used as basis for an engine performance evaluation. Comparison with conventional rocket configurations shows that an upper performance limit for the diverging reactor is comparable with performance estimates for engines using an adiabatic work cycle. Development of the diverging reactor for engine applications may, however, offer some advantages for very hot, high-energy, propellant systems

Material selection criteria are discussed; the material chosen is described, and results of tests performed after environmental exposure at five facilities are given. The facilities used for exposure to ultraviolet radiation/thermal-vacuum environments and the equipment used for testing physical properties before and after exposure are described.     We consider a particle dragged through a medium at constant temperature as described by a Langevin equation with a time-dependent potential. The time-dependence is specified by an external protocol. We give conditions on potential and protocol under which the dissipative work satisfies an exact symmetry in its fluctuations for all times. We also present counter examples to that exact fluctuation symmetry when our conditions are not satisfied. Finally, we consider the dissipated heat which differs from the work by a temporal boundary term. We explain when and why there can be a correction to the standard fluctuation theorem due to the unboundedness of that temporal boundary.
