Response of Micro-Tom to Low Light
Yield is proportional to light absorption, but maximum light cannot always be supplied to crops in either space station production settings or in teaching situations. It is therefore important to determine how crops grow in light-limited conditions. ‘Micro-Tom’ tomato was grown in fluorescent light under shade cloth to provide 100, 200, and 300 µmol m-2 s-1 for 16-h photoperiods at 26/22 °C for 117 days.Yield and harvest index increased as light increased. There were significant amounts of unripe fruit after 117 days. During ripening, the fraction of dry matter decreases within the fruit.The higher yield was likely due to better fruit set in higher light, as evidenced by higher numbers of fruit with additional light. Higher light did not influence ripening, and the fraction of red fruit stayed constant with all light. ‘Micro-Tom’ was able to set and ripen fruit even at extremely low light levels
Educational leadership in schools of the 21st century: A paradigm shift in style and practice
The role of schools in the 21st century is to prepare students to be the leaders of tomorrow. Constant change is now a characteristic of our society and schools need to create an interdisciplinary curriculum that is relevant, challenging and caters for the individual needs of each student. In addition to the traditional learning areas, our students will need to be able to cope with constant change, to have a thorough understanding of, and proficient skills in information technology and to be creative problem solvers. Innovation, creativity and an improved capacity to adapt to changing circumstances will be the forefront of future learning. The schools of the 21st century will need committed; active participation in inquiry based learning and problem solving, rather than the passive transmission and reception of information. The Head’s role in an international school is that of the leading learner. The Head needs to be an exemplary educator and administrator ready to share his/her skills within a collaborative culture of learning and quality management. New skills will be required in addition to the traditional skills of human, physical, site and financial management. These skills must include a capacity to lead collaboratively; an ability to develop and articulate a shared educational vision, a commitment to the empowerment and staff and students and the ability to foster a school culture of continuous improvement. These skills together with the traditional responsibilities of school leadership will lead to the creation of a vibrant, exciting learning community

Crystal Structure and Substrate Specificity of Drosophila 3,4-Dihydroxyphenylalanine Decarboxylase
3,4-Dihydroxyphenylalanine decarboxylase (DDC), also known as aromatic L-amino acid decarboxylase, catalyzes the decarboxylation of a number of aromatic L-amino acids. Physiologically, DDC is responsible for the production of dopamine and serotonin through the decarboxylation of 3,4-dihydroxyphenylalanine and 5-hydroxytryptophan, respectively. In insects, both dopamine and serotonin serve as classical neurotransmitters, neuromodulators, or neurohormones, and dopamine is also involved in insect cuticle formation, eggshell hardening, and immune responses.In this study, we expressed a typical DDC enzyme from Drosophila melanogaster, critically analyzed its substrate specificity and biochemical properties, determined its crystal structure at 1.75 Angstrom resolution, and evaluated the roles residues T82 and H192 play in substrate binding and enzyme catalysis through site-directed mutagenesis of the enzyme. Our results establish that this DDC functions exclusively on the production of dopamine and serotonin, with no activity to tyrosine or tryptophan and catalyzes the formation of serotonin more efficiently than dopamine.The crystal structure of Drosophila DDC and the site-directed mutagenesis study of the enzyme demonstrate that T82 is involved in substrate binding and that H192 is used not only for substrate interaction, but for cofactor binding of drDDC as well. Through comparative analysis, the results also provide insight into the structure-function relationship of other insect DDC-like proteins
Optical Spectroscopy of the Surface Population of the rho Ophiuchi Molecular Cloud: The First Wave of Star Formation
We present the results of optical spectroscopy of 139 stars obtained with the
Hydra multi-object spectrograph. The objects extend over a 1.3 square degree
area surrounding the main cloud of the rho Oph complex. The objects were
selected from narrowband images to have H alpha in emission. Using the presence
of strong H alpha emission, lithium absorption, location in the
Hertzsprung-Russell diagram, or previously reported x-ray emission, we were
able to identify 88 objects as young stars associated with the cloud. Strong H
alpha emission was confirmed in 39 objects with line widths consistent with
their origin in magnetospheric accretion columns. Two of the strongest
emission-line objects are young, x-ray emitting brown dwarf candidates with M8
spectral types. Comparisons of the bolometric luminosities and effective
temperatures with theoretical models suggest a medianage for this population of
2.1 Myr which is signifcantly older than the ages derived for objects in the
cloud core. It appears that these stars formed contemporaneously with low mass
stars in the Upper Scorpius subgroup, likely triggered by massive stars in the

Pleural Fluid Mesothelin as an Adjunct to the Diagnosis of Pleural Malignant Mesothelioma
Rationale:. The diagnosis of pleural malignant mesothelioma (MM) by effusion cytology may be difficult and is currently controversial. Effusion mesothelin levels are increased in patients with MM but the clinical role of this test is uncertain. Objectives:. To determine the clinical value of measuring mesothelin levels in pleural effusion supernatant to aid diagnosis of MM. Methods and Measurements. Pleural effusion samples were collected prospectively from 1331 consecutive patients. Mesothelin levels were determined by commercial ELISA in effusions and their relationship to concurrent pathology reporting and final clinical diagnosis was determined. Results:. 2156 pleural effusion samples from 1331 individuals were analysed. The final clinical diagnosis was 183 MM, 436 non-MM malignancy, and 712 nonmalignant effusions. Effusion mesothelin had a sensitivity of 67% for MM at 95% specificity. Mesothelin was elevated in over 47% of MM cases in effusions obtained before definitive diagnosis of MM was established. In the setting of inconclusive effusion cytology, effusion mesothelin had a positive predictive value of 79% for MM and 94% for malignancy. Conclusions:. A mesothelin-positive pleural effusion, irrespective of the identification of malignant cells, indicates the likely presence of malignancy and adds weight to the clinical rationale for further investigation to establish a malignant diagnosis
Auto-Antibodies to β-F1-ATPase and Vimentin in Malignant Mesothelioma
Patients with Malignant Mesothelioma (MM) develop unidentified auto-antibodies to MM tumour antigens. This study was conducted to identify the targets of MM patient auto-antibodies in order to try to understand more of the anti-tumour response and to determine if these antibodies might be helpful for diagnosis or prognostication. Using MM patient sera in a Western immunoblott screening strategy, no common immunoreactive proteins were identified. The sera from one long-term survivor recognised a protein band of 50–60 kDa present in cell lysates from four of five MM cell lines tested. The immunoreactive proteins in this band were identified by 2D electrophoretic separation of a MM cell line protein lysate, followed by analysis of excised immunoreactive proteins on a MALDI TOF mass spectrometer and peptide mass fingerprinting. The immunoreactive proteins identified were vimentin (accession gi55977767) and the ATP synthase (F1-ATPase) beta chain (accession gi114549 and gi47606749). ELISA assays were developed for antibodies to these proteins. Neither vimentin (median and 95% CI 0.346; 0.32–0.468 for MM patients, 0.327; 0.308–0.428 for controls) nor ß-F1-ATPase (0.257; 0.221–0.453 for MM patients, 0.263; 0.22–0.35 for controls) showed significant differences in autoantibody levels between a group of MM patients and controls. Using a dichotomized antibody level (high, low) for these targets we demonstrated that vimentin antibody levels were not associated with survival. In contrast, high ß-F1-ATPase antibody levels were significantly associated with increased median survival (18 months) compared to low ß F1 ATPase antibody levels (9 months; p = 0.049). Immunohistochemical analysis on a MM tissue microarray showed cytoplasmic staining in 28 of 33 samples for vimentin and strong cytoplasmic staining in14 and weak in 16 samples for ß-F1-ATPase. Therefore antibodies to neither vimentin nor ß-F1-ATPase are useful for differential diagnosis of MM, however high antibody levels to ß-F1-ATPase may be associated with increased survival and this warrants further investigation
Changes in anemia management and hemoglobin levels following revision of a bundling policy to incorporate recombinant human erythropoietin
In April 2006, Japan's health insurance system instituted a bundling policy that included recombinant human erythropoietin (rHuEPO) in outpatient hemodialysis therapy. To evaluate outcomes of this, we analyzed a prospective cohort of hemodialysis patients in the Japan Dialysis Outcomes and Practice Patterns Study, in 53 facilities using prevalent cross-sections of 1584 patients before and 1622 patients after the rHuEPO reimbursement change. Patient data included hemoglobin levels, iron management profiles, and anemia treatment with rHuEPO and intravenous iron. No significant differences were found in pre- or post-policy cross-sections for hemoglobin distributions or the percentage of patients prescribed rHuEPO. Among patients receiving rHuEPO, the mean dose significantly decreased by 11.8 percent. The percentage of patients prescribed intravenous iron over 4months significantly increased; however, the mean dose of iron did not significantly change. Thus, this bundling policy was associated with reduced rHuEPO doses, increased intravenous iron use, and stable hemoglobin levels in Japanese patients receiving hemodialysis
