    Does community deprivation determine longevity after the age of 75? A cross-national analysis

    Objectives: Analyze the association between socioeconomic deprivation and old-age survival in Europe, and investigate whether it varies by country and gender. Methods: Our study incorporated five countries (Portugal, Spain, France, Italy, and England). A 10-year survival rate expressing the proportion of population aged 75–84 years who reached 85–94 years old was calculated at area-level for 2001–11. To estimate associations, we used Bayesian spatial models and a transnational measure of deprivation. Attributable/prevention fractions were calculated. Results: Overall, there was a significant association between deprivation and survival in both genders. In England that association was stronger, following a dose–response relation. Although lesser in magnitude, significant associations were observed in Spain and Italy, whereas in France and Portugal these were even weaker. The elimination of socioeconomic differences between areas would increase survival by 7.1%, and even a small reduction in socioeconomic differences would lead to a 1.6% increase. Conclusions: Socioeconomic deprivation was associated with survival among older adults at ecological-level, although with varying magnitude across countries. Reasons for such cross-country differences should be sought.     Distance to parks and non-residential destinations influences physical activity of older people, but crime doesn’t: a cross-sectional study in a southern European city

    Background: Physical activity (PA) has numerous health benefits, but older adults live mostly sedentary lifestyles. The physical and social neighborhood environment may encourage/dissuade PA. In particular, neighborhood crime may lead to feeling unsafe and affect older adults’ willingness to be physically active. Yet, research on this topic is still inconclusive. Older population, probably the age group most influenced by the neighborhood environment, has been understudied, especially in Southern Europe. In this study, we aimed to analyze the association between leisure-time physical activity (LTPA) in older adults and objective crime, alongside other neighborhood characteristics. Methods: We obtained data from a population-based cohort from Porto (2005–2008) to assess LTPA. Only adults aged 65 years or more were included (n = 532). A Geographic Information System was used to measure neighborhood characteristics. Neighborhood crime was expressed as crime rates by category (incivilities, criminal offenses with and without violence and traffic crime). Neighborhood characteristics such as socioeconomic deprivation, land gradient, street density, transportation network, distance to parks, non-residential destinations and sport spaces were also included. Generalized Additive Models were fitted to estimate the association between neighborhood characteristics and the participation (being active vs. inactive) and frequency (min/day) of LTPA. Results: Forty-six percent of the men and 61 % of the women did not engage in any kind of LTPA. Among the active participants, men spent on average 50.5 (35.2 Standard Deviation, SD) min/day in LTPA, whereas the average among women was 36.9 (35.1 SD) min/day (p < 0.001). Neighborhood crime was unrelated to the participation in, or frequency of, LTPA. On the other hand, two neighborhood characteristics – distance to the nearest park (β = −0.0262, p = 0.029) and to the nearest non-residential destination (β = −0.0735, p = 0.019) – were associated with time spent on LTPA, but only among active older women. No neighborhood characteristic was related to participation in LTPA. Conclusions: From a public health point of view, the provision of parks and non-residential destinations (shops, schools, cultural and worship places) might contribute to elevate PA levels of already active older women. On the other hand, in this setting, crime was not a big issue

    Cutaneous streptococcal abscess treated by photodynamic therapy

    Background: Photodynamic therapy has been investigated in different areas of health through experimental conditions. Its action can alter fundamental structures for the survival of microorganisms without any development of microbial resistance.Materials and Methods: Young sheep presenting with abscess in the left forelimb caused by Streptococcus spp. was previously treated with antibiotics. There was no clinical improvement with the treatments, and the bacteria presented sensitivity in vitro. Therefore, Photodynamic therapy associating methylene blue and red laser (660 nm) was used to treat the abscess.Results: After a day of treatment, complete healing was witnessed with no recurrence was observed during the 3-month follow-up period.Conclusion: The scientific results of the antimicrobial effect of PDT proved to be a therapeutic option with great potential for clinical application.Keywords: Photoinactivation, Laser, Sheep, Streptococcus spp

    Geometric compensation applied to image analysis of cell populations with morphological variability: A new role for a classical concept

    Immunofluorescence is the gold standard technique to determine the level and spatial distribution of fluorescent-tagged molecules. However, quantitative analysis of fluorescence microscopy images faces crucial challenges such as morphologic variability within cells. In this work, we developed an analytical strategy to deal with cell shape and size variability that is based on an elastic geometric alignment algorithm. Firstly, synthetic images mimicking cell populations with morphological variability were used to test and optimize the algorithm, under controlled conditions. We have computed expression profiles specifically assessing cell-cell interactions (IN profiles) and profiles focusing on the distribution of a marker throughout the intracellular space of single cells (RD profiles). To experimentally validate our analytical pipeline, we have used real images of cell cultures stained for E-cadherin, tubulin and a mitochondria dye, selected as prototypes of membrane, cytoplasmic and organelle-specific markers. The results demonstrated that our algorithm is able to generate a detailed quantitative report and a faithful representation of a large panel of molecules, distributed in distinct cellular compartments, independently of cell's morphological features.     Keratocystic odontogenic tumor overexpresses invadopodia-related proteins, suggesting invadopodia formation

    OBJECTIVE: Keratocystic odontogenic tumor (KOT) is an odontogenic neoplasm that shows aggressive clinical behavior and local invasiveness. Invadopodia are actin-rich cellular protrusions exhibiting proteolytic pericellular activity, thereby inducing focal invasion in neoplastic cells and increasing neoplasms aggressiveness. Thus, this study aimed to evaluate immunoexpression of invadopodia-related proteins, cortactin, MT1-MMP, Tks4, and Tks5, in KOT. STUDY DESIGN: Immunohistochemistry of 16 cases of KOT, eight cases of calcifying cystic odontogenic tumor (CCOT), and eight samples of the oral mucosa (OM) was carried out to assess the expression of the above described invadopodia-related proteins in the basal and suprabasal layer. RESULTS: KOT samples showed higher and significant immunoexpression of cortactin, MT1-MMP, TKs4, and TKs5 compared with the CCOT and OM samples. Significant expression of all these proteins was observed in the basal layer compared with the suprabasal layer in KOT. CONCLUSIONS: Overexpression of cortactin, MT1-MMP, TKs4, and TKs5 was observed in KOT compared with samples of CCOT and OM. These proteins were also overexpressed in the basal over the suprabasal layer of KOT samples. Taken together, these results suggest the participation of invadopodia-related proteins on the pathogenesis of this lesion

    Patients' perspective and lung function correlation:the importance of questionnaires in home mechanical ventilation

    Classical physiological variables used to monitor respiratory function in patients under non-invasive ventilation (NIV), such as FEV1 or FVC, correlate poorly with reported impairment of physical function or overall health status and hence provide an incomplete picture of impaired health. Aim: to investigate the association between the Portuguese S3-non-invasive ventilation (S3-NIV) questionnaire score and objective measures of lung function. Consecutive adult patients with chronic respiratory failure, established on home NIV for at least 30 days were recruited in one outpatient clinic. Correlations between physiological variables and S3-NIV score were computed with Spearman rank coefficient. We studied 230 stable patients (126 male, 54.8%) with a mean age of 69.2 (± 11,0) years. Demographic and spirometry characteristics, and S3-NIV respiratory and total scores are presented on the table, according to disease groups. Spearman coefficient values are also presented. Neither the S3-NIV total score nor the respiratory symptoms subscore correlate with lung function impairment in any of the disease groups. This reinforces the notion that symptoms questionnaires and patient reported outcome measures must always be obtained directly from the patient and should be included in regular treatment monitoring

    Effect of partially purified fumonisins on cellular immune response in experimental murine paracoccidioidomycosis

    Fumonisins are mycotoxins produced mainly by Fusarium verticillioides, which can modulate the immune response. Paracoccidioidomycosis (PCM), caused by the fungus Paracoccodioides brasiliensis (Pb), is one of the most important systemic mycoses in Latin America. The aim of this study was to evaluate the effect of the partially purified fumonisins on cellular immune response in mice infected with Pb. Four groups of male BALB/c mice were used. Groups PB and PB/FB were inoculated i.v. with 1 × 105 Pb yeast cells and, after 28 days, groups FB and PB/FB were inoculated (s.c.) with partially purified fumonisin B1 from F. verticillioides (5 × 2.25 mg FB1/kg body weight). After 7 days, cellular immune response was evaluated by delayed-type hypersensitivity (DTH) and lymphoproliferative assays (LA) using spleen cells. Nitric oxide (NO) production by spleen cells was also evaluated. The specific LA response to Pb antigen was higher in group PB than in FB and CTR groups (p&lt; 0.05) but not significant with PB/FB. The DTH response was higher in infected than non infected groups (p&lt;0.05) but also no significantly with PB and PB/FB groups. The lyphoproliferative response to ConA was decreased in FB or PB/FB in relation to CTR (p&lt;0.05) but not with PB/FB and also a reduction of NO levels was observed in fumonisin treated in relation to control group FB1/kg (p&lt;0.05). In conclusion, fumonisin B1 or other components of F. verticillioides extracts significantly suppress the unspecific cellular immune response and the NO production by splenocytes from P. brasiliensis infected or not infected BALB/c mice.Keywords: Fumonisin, Paracoccodioides brasiliensis, lymphoproliferative assay, nitric oxideAfrican Journal of Biotechnology Vol. 12(42), pp. 6126-613

    The influence of socioeconomic, biogeophysical and built environment on old-age survival in a Southern European city

    Old-age survival is a good indicator of population health and regional development. We evaluated the spatial distribution of old-age survival across Porto neighbourhoods and its relation with physical (biogeophysical and built) and socioeconomic factors (deprivation). Smoothed survival rates and odds ratio (OR) were estimated using Bayesian spatial models. There were important geographical differentials in the chances of survival after 75 years of age. Socioeconomic deprivation strongly impacted old-age survival (Men: least deprived areas OR=1.31(1.05-1.63); Women OR=1.53(1.24-1.89)), explaining over 40% of the spatial variance. Walkability and biogeophysical environment were unrelated to old-age survival and also unrelated to socioeconomic deprivation, being fairly evenly distributed through the city
